Immune reactions to foreign or self-antigens lead to protective immunity and, sometimes, immune disorders such as allergies and autoimmune diseases. Antigen presenting cells （APC） including epidermal langerhans cells （LCs） play an important role in the course and outcome of the immune reactions. Epidermal powder immunization （EPI） is a technology that offers a tool to manipulate the LCs and the potential to harness the immune reactions towards prevention and treatment of infectious diseases and immune disorders.

Background:Skin contact has been proposed for peanut sensitization;however,we found previously that tolerance to peanut was achieved by applying peanut protein onto the intact skin of mice,which indicates that skin application of food allergen might be used as a prophylactic and/or therapeutic intervention for food *** mechanism of the skin-induced tolerance is not clear *** cells(LCs)are the only dendritic cells present in the epidermis at the steady state,and have been shown to have negatively regulatory role in many allergic and inflammatory skin ***,we explore the role of LCs in the mechanism of the skin-induced tolerance to ***:Langerin-DTA mice that constitutively defect in langerhans cells were ***-DTA mice and wide-type control were given 5 weekly cutaneous application of 1000 ug complete peanut extract(CPE)on the shaved back skin,followed by 2 weekly intra-peritoneal injection of CPE to induce allergic reactivity to *** various timepoints,the immunologic responses and allergic clinical manifestations to allergens were *** number and phenotype of LCs,dermal dendritic cells and regulatory T cells in skin and regional draining lymph node were analyzed after application of CPE onto intact skin in wide-type ***:Skin application of CPE to structurally intact skin did not lead to allergic sensitization to peanut in LangerinDTA or wide-type *** wide-type mice,cutaneous allergen application blocked the subsequent induction of sensitiza-tion including inhibiting ***-induced CPE-specific IgE,IgGl and IgG2a production,suppressing of peanut anaphylaxis,and modulating the sensitization-promoted cytokine ***,there were almost no inhibitions on the sensitization in Langerin-DTA *** number of migratory LCs and regulatory T cells increased in the regional draining lymph node of wide-type *** LCs also showed an immature phenotype with low level of CD8

A Study on Changes of langerhans cells in Human Epidermal Cell Culture

Human epidermal cells (ECs) were grown in vitro from normalhuman foreskin and the changes of langerhans cells (LCs) within theoriginal explants and the epidermal outgrowth were observed in culturewith ATPase staining, HLA - Dr monoclonal antibody indirectimmuno-fluorescence and enzyme labelling avidin-biotin complex (ABC)staining. The capacity of ECs to stimulate the proliferation of allogeniclymhocytes was tested in the mixed skin-lymphocyte culture response(MSLR). The ATPase HLA-Dr positive LCs were gradually decreasedand significantly deformed within the original explants in culture. 70% ofLCs was decreased 3 days after culture. 90% of LCs was lost and the LCsremained in the original explants were only seen their outline 7 days afterculture. LCs approximatively disappeared within the original explants 14to 21 days after culture. The ATPase, HLA - Dr positive LCs have neverbeen found within the epidermal outgrowth. The ECs which were freshlyisolated from skin explants potently stimulated the proliferation ofallogenic lymphocytes in the MSLR. The ECs which were separated fromthe original explants after 3 days of culture were significantly declinedtheir ability to stimulate the proliferation of allogenic lymphocytes in theMSLR. The ECs separated from the original explants after 7 days ofculture were lost their ability to stimulate the proliferation of allogeniclymphocytes in the MSLR. The ECs separated from the epidermaloutgrowth failed to stimulate the proliferation of allogenic lymphocytes inthe MSLR. These results suggested that the ECs grown in our culturesystem were declined their immunogenety of skin allografts.

A rare case of langerhans cell histiocytosis of the gastrointestinal tract

langerhans cell histiocytosis （LCH） is a group of idiopathic disorders characterized by the proliferation of specialized, bone marrow-derived langerhans cells and mature eosinophils. The clinical spectrum ranges from an acute, fulminant, disseminated disease called Letterer Siwe disease to solitary or few, indolent and chronic lesions of the bone or other organs called eosinophilic granuloma. Involvement of the gastrointestinal tract is very rare in LCH. We present the case of a 53-year-old woman referred by her primary care physician for a screening colonoscopy. A single sessile polyp, measuring 4 mm in size, was found in the rectum. Histopathological examination revealed that the lesion was relatively well circumscribed and comprised mainly a mixture of polygonal cells with moderate-to-abundant pink slightly granular cytoplasm. The nuclei within these cells had frequent grooves and were occasionally folded. Immunohistochemical staining was positive for CD la which confirmed the diagnosis of LCH. On further workup, there was no evidence of involvement of any other organ. On follow up colonoscopy one year later, there was no evidence of disease recurrence. Review of the published literature revealed that LCH presenting as solitary colonic polyp is rare. However, with the increasing rates of screening colonoscopy, more colonic polyps may be identified as LCH on histopathology. This underscores the importance of recognizing this rare condition and ensuring proper follow-up to rule out systemic disease.

Temporal effect of local hyperthermia on murine contact hypersensitivity

Background The sensitization and elicitation phases are involved in the immunopathogenesis of contact hypersensitivity （CHS）. langerhans cells （LCs） are believed to play pivotal roles in the sensitization stage of CHS. Local hyperthermia on skin induces the migration as well as maturation of epidermal LCs. Although fever-range whole body hyperthermia and local hyperthermia at 43℃ prior to sensitization were reported to suppress CHS, the effects of different temperatures and the timing sequence of local hyperthermia on CHS have not been tackled. Methods Local hyperthermia was applied to murine dorsal skin 3 days prior to, concurrent with, or 2 days post sensitization with fluorescein isothiocyanate （FITC） in BALB/c mice. Local hyperthermia temperatures at 37℃, 39℃, 41℃ and 43℃ were applied to mouse dorsal skin and the severity of CHS was calculated by measuring the swelling response of the challenged ears. Results Local hyperthermia at 39℃, 41℃ and 43℃ prior to sensitization reduced the severity of CHS, as compared with that at 37℃. The suppression of CHS was temperature dependant in that higher temperature had a stronger effect. On the contrary, the hyperthermia treatments, either concurrent with or post-sensitization, resulted in an enhanced temperature-dependant ear swelling response. Conclusions The severity of murine CHS could be influenced by local hyperthermia at the sensitization stage in a temperature dependant manner. The temporal effect of local hyperthermia suggested a novel factor in interpreting the severity of allergic contact dermatitis.
