Three new lignan glucosides from the roots of Scutellaria baicalensis

Three new lignan glucosides, baicalensinosides A–C(1–3), were isolated from the roots of Scutellaria baicalensis. The structural elucidation was achieved by in-depth spectroscopic examinations and qualitative chemical test. Structurally, these compounds belong to the 3,4-dibenzyltetrahydrofurantype lignan glycoside with a mono-hydroxyl substitution at the 70-position of benzylidene group on the numbering system of lignans being one of their shared critical features. The anti-osteoporotic activity of the isolated compounds was assessed in an in vitro osteoprotegerin(OPG) transcriptional activity assay using dual luciferase reporter detection. At 10 μmol/L, compounds 1–3 increased the relative activating ratio of OPG transcription to 1.83, 0.84 and 0.98 times that of the control group, respectively.

Further investigation on the aerial parts of Isodon enanderianus afforded a novel asymmetric ent kauranoid dimer, enanderinanin J (1). The structure of the dimer was elucidated by means of spectroscopic methods (including 2D NMR techniques). Enanderinanin J was a dimer of xerophilusin A and probably formed by cycloaddition.

Their structures were elucidated on the basis of their spectral properties, as well as X ray crystallographic analysis. The cytotoxicities of these two new compounds against human tumor cells K562 and T24 were also tested, and sculponeatin J showed significant inhibitory effects with IC 50 values less than 1.0 μg/mL.

Four new oleanane triterpene saponins were isolated and purified from the whole plant of Clinopodium urticifolium. They were 3B 16B, 23, 28-tetrahydroxyoleana-9 (11), 12(13)-diene-3-yl-[B-D-glucopyranosyl-(1-2)]-[B-D-glucopyranosyl-(1-3)]- B-D-fucopyranoside 1; 3B, 16B, 21B, 23, 28-pentahydroxyoleana-9(11), 12(13)-diene-3-yl-[B-D-glucopyranosyl-(1-2)]-[B-D-glucopyranosyl-(1-3)]- B-D-fucopyranoside 2; 3B, 16B, 23, 28-tetrahydroxyoleana-9(11), 12(13)-diene-3-yl-[B-D-glucopyranosyl-(1-6)-B-D-glucopyranosyl-(1-3)]-[B-D-glucopyranosyl-(1-2)]-B-D-fucopyranoside 3; 3B, 16B, 23, 28-tetrahydroxyoleana-9(11), 12(13)-diene-3-yl-[B-D-gluco-pyranosyl-(1-4)-B-D-glucopyranosyl-(1-6)-B-D-glucopyranosyl-(1-3)]-[B-D-glucopyranosyl-(1-2)]- B-D-fucopyranoside 4. Their structures were elucidated on the basis of interpretation of NMR and MS data and from chemical evidence.

A new phenylethanoid glycoside,seulponisidc（1） was isolated from the aerial parts of Isodon sculponeatus（Vaniot） Kudo, along with six known compounds martynoside（2）,verbascoside（3）,（＋）-hydroxypinoresinol-8-O- β-D-glucoside（4）,cedrusin（5）, 7-megastigmene-3S,5R,6R,7E,9S-tetrol（6） and 4-oxo- β-ionol- β-D-glucopyranoside（7）.Their chemical structures were elucidated from physieochemical data and by acidic hydrolysis.

Two novel ent-kauranoids with new sub-skeleton types, gesneroidins G and H were isolated as white powder from the ether extract of the leaves of Isodon gesneroides through normal phase column chromatography. Their structures were elucidated as 3β,6α,7β-triacetoxyl-15-hydroxyl-14-oxo-ent-15,16- ,seco-kaur-11,17-olide and 1α-hydroxyl-3β,6α,7β,11β- tetraacetoxyl-ent-nor-15,17-kaur-8,16-olide on the basis of the spectral evidences including 1D and 2D NMR spectra.

Two new ent-kaurane diterpenoids, xerophilusin E (1) and xerophilusin F (2), were isolated from the leaves of Isodon xerophilus. Their structures were determined as 3, 20: 7, 20-diepoxy-ent-kaur-16-en-15-one and 7 beta, 14 beta, 20 (R)-trihydroxy-11 beta-acetoxy-7,20-cyclo-ent-kaur-16-en-6, 15-dione, respectively, by spectral methods and X-ray crystallographic analysis.
