Case of autoimmune hepatitis with markedly enlarged hepatoduodenal ligament lymph nodes

Autoimmune hepatitis(AIH) is a necroinflammatory liver disease of unknown *** disease is characterized histologically by interface hepatitis,biochemically by increased aspartate aminotransferase and alanine aminotransferase levels,and serologically by increased autoantibodies and immunoglobulin G *** we discuss AIH in a previously healthy 37-year-old male with highly elevated serum levels of soluble interleukin-2 receptor and markedly enlarged hepatoduodenal ligament lymph nodes(HLLNs,diameter,50 mm).Based on these observations,the differential diagnoses were AIH,lymphoma,or Castleman's *** biopsy revealed the features of interface hepatitis without bridging fibrosis along with plasma cell infiltration which is the typical characteristics of acute *** node biopsy revealed lymphoid follicles with inflammatory lymphocytic infiltration;immunohistochemical examination excluded the presence of lymphoma ***,he was administered corticosteroid therapy:after 2 mo,the enlarged liver reached an almost normal size and the enlarged HLLNs reduced in *** could not find AIH cases with such enlarged lymph nodes(diameter,50 mm) in our literature ***,we speculate that markedly enlarged lymph nodes observed in our patient may be caused by a highly activated,humoral immune response in AIH.

Probiotic Mixture VSL#3 Alleviates Dextran Sulfate Sodium-induced Colitis in Mice by Downregulating T Follicular Helper Cells

Clinical trials have shown beneficial effects of probiotics on inflammatory bowel diseases (IBD), although the exact mechanism remains unknown. VSL#3, a mixture of 8 probiotic bacteria, has been confirmed to have adjunctive therapeutic effects on colitis. T follicular helper (Tfh) cells, a new separate subset of CD4+ T helper cells, have been proved to play a vital role in autoimmunity. The present study aimed to identify the beneficial effect of the probiotic mixture VSL#3 on the mouse model of colitis by regulating Tfh cells. Dextran sulfate sodium (DSS) was used to induce chronic colitis in C57BL/6 mice. VSL#3 (3x109 live bacteria) was given to C57BL/6 mice every other day for 60 days by gavage. The disease activity index (DAI), histological activity index (HAI), colon length and myeloperoxidase (MPO) activity were detected. Immunofluorescence was used to visualize the location of Tfh cells. Immunoglobulins, Tfh cells and plasma cells were quantified by enzyme-linked immunosorbent assay (ELISA), flow cytometry, real-time PCR or Western blotting. The results showed that after DSS treatment, the humoral immunity was disordered in C57BL/6 mice, with increased IgM, IgG and IgA levels in colonic mucus and increased Tfh cells in mesenteric lymph nodes (MLN). VSL#3 treatment showed anti-inflammatory effects as evidenced by reduced DAI score, HAI score and MPO activity. IgM, IgG and IgA levels were significantly reduced in colon mucus, and the number of Tfh cells was markedly decreased in MLN after VSL#3 treatment. It was concluded that VSL#3 alleviates DSS-induced colitis by downregulating Tfh cells, and Tfh cells may become a potential therapeutic target for IBD.

Construction of multivalent DNA vaccines for Mycobacte-rium tuberculosis and its immunogenicity

The coding regions of Ag85B MPT-64, and ESAT-6 secreted proteins were cloned initially into the eu-karyotic expression vector pJW4303, then transformed to E. coli Top 10 strain for plasmid DNA extraction and further analysis. Plasmids containing the right insertion were se-quenced to confirm their identity. COS7 cells were trans-fected with a mixture containing serially diluted plasmid DNA encoding three secreted proteins and Lipofectin (Gibco). The supernatants and pellets prepared from various cell lines were run on SDS-PAGE gel and the expression of these proteins in COS7 cells were demonstrated by immunoblot using polyclonal or monoclonal antiserum of ***37Rv. 21 days after first vaccination of C57BL-6 mice by all three recombinant eukaryotic expressing vectors, antibody titer for Ag85B reached 1:3200. 21 days after second vaccination, the antibody titer reached 1:102400. Immunomodulatory activity of butanol fraction of Gentiana olivieri Griseb. on Balb/C mice

Objective:To explore the immunomodulatory properties of 80% ethanol extract and butanol fraction of Gentiana olivieri(G. olivieri) Griseb on Balb/C ***:The study was performed with basic models of immunomodulation such as the humoral antibody response(hemoglutination antibody titres), cell mediated immune response(delayed type hypersensitivity and in vivocarbon clearance or phagocytosis). Ethanol(80%) extract of flowering aerial parts of *** its butanol fraction were administered p.o.(orally) to the mice. Levamisole, 2.5 mg/kg was used as standard ***:There was a potentiation of immune response to sheep red blood cells by cellular and humoral mediated mechanisms comparable to levamisole(2.5 mg/kg) by both 80% ethanol extract and the butanol fraction at doses of 50-200 mg/kg in male Balb/C mice. Both significantly(Phumoral immune response in cyclophosphamide(250 mg/kg)immunosupressed mice at 100 and 200 mg/kg of each extract and fraction as compared to *** potentiation of delayed type hypersensitivity response was statistically significant(Phumoral immunity in experimental models of immunity in mice. Butanol fraction is the most effective at a dose level of 200 mg/kg.

Effects of Methionine on the Immune Function in Animals

Nutrition and immunity are the hot topics in animal’s production, and the effects of methionine on the immunity are already confirmed as the deep research on the nutrition of amino acid and immune function. However, the relationship of the methionine and immunity has not been elucidated clearly, this review aims to clarify the effects of methionine on immune function in the aspects of growth and development immune organs, the histological structure of the immune organs, non-specific immunity, humoral immunity, cellular immunity and cytokines, and to provide foundations for further studies on the relationship between methionine and immune function.

Analysis of the levels of in vivo immunological indicators in patients with different types of malignanat lymphoma

Objective:To analyze the levels of in vivo immunological indicators in patients with different types of malignanat ***: A total of 69 patients with different types of malignanat lymphoma who were admitted in our hospital were included in the study, among which 31 had primary extranodular malignant lymphoma (extranodular group), and 38 had primary nodular malignant lymphoma (nodular group). Moreover, 34 patients with pure lymphnoditis were served as the control group. FCM and full-automatic speed scattering turbidimetry were used to detect T lymphocyte subsets proportion, IgG, IgA, IgM, C3, and C4 in the three ***: The comparison of CD3+, CD4+, CD8+, NK cell proportion, and CD4+/CD8+ among the three groups was significantly different. When compared with the control group, CD3+, CD4+, CD8+, NK cell proportion, and CD4+/CD8+ in the extranodular group and nodular group were significantly reduced;moreover, CD3+, CD4+, CD8+, and CD4+/CD8+ in the extranodular group were significantly lower than those in the nodular group, while NK cell in the extranodular group was significantly higher than that in the nodular group. The comparison of IgG, IgA, and C3 levels among the three groups was statistically significant. When compared with the control group, IgG and IgA levels in the extranodular group and nodular group were significantly elevated, while C3 level was significantly reduced, and the comparison between the extranodular group and nodular group between the two groups was statistically significant. IgM level in the extranodular group and nodular group was elevated, C4 level was reduced, but the comparison with the control group was not statistically significant. THE EFFECT OF SYMPATHETIC NERVE AND ADRENAL ON THE VISCERA-AURICULO-ACUPOINT RESPONSE

The article summarizes the achievements of research on the mechanism of auriculo-acu-points by us in recent years.(1) By stimulating and recording the electric activities of the sympathetic nerve, it has been observed that the abnormal sympathetic excitation is the cause for the reduction of the electric resistance itt ear acupoints, the occurrence of the low-resistance spots in the auricle, and the auricular resistance has a phasic process of rising ahead and falling behind, we have observed that the quantity change of the low-resistance spots in the auricle is parallel to the quantity of the syrnpathetlc fibres, with local injection of the specific sympathetic-destroying agent, 6-OHDA.(2) It was confirmed with animal cross-circulation method that humour factors participate in the viscera-auriculo-acupoint response. Furthermore, the adrenal effect was proved with the adrenal extirpation. A several models of Yang-deficiency syndrome were rnade to analyze the action of some substaflces secreted from adrenal cortex and adrenal medulla.(3) On the basis of the above-mentioned outcome, a hypothesis that specific and nonspecif1c viscera-auriculo-acupoint responses are mixed has been rendered, The specific response here means the special relationship between certain parts of the auricle and certain viscera which forms the foundation of auriculo-acupoints, while the nonspecific one refers to the general or diffused response in the whole auricle resulting from the actions of all the viscera and other parts of the body. The specific response may be fulfilled by the sympathetic nerve, wh1le the nonspecific response is accomplished by both of sympathetic nerve and adrenal gland. Tumors and cancers of the gastrointestinal tract and pancreas are commonly derived from precursor lesions so that understanding the physiological, cellular, and molecular mechanisms underlying the pathogenesis of precursor lesions is critical for the prevention and treatment of those neoplasms. Pancreatic neuroendocrine tumors(PNETs) can also be derived from precursor lesions. Pancreatic α cell hyperplasia(ACH), a specific and overwhelming increase in the number of α cells, is a precursor lesion leading to PNET pathogenesis. One of the 3 subtypes of ACH, reactive ACH is caused by glucagon signaling disruption and invariably evolves into PNETs. In this article, the existing work on the mechanisms underlying reactive ACH pathogenesis is reviewed. It is clear that the liver secretes a humoral factor regulating α cell numbers but the identity of the liver factor remains elusive. Potential approaches to identify the liver factor are discussed.

Kanggan Granule Ameliorates Dexamethasone-Induced Immunosuppression in Mice

Objective: This study was to investigate the effect of the Chinese herbal compound Kanggan granule (KG) on immune function in a mouse model of immunosuppression and its possible mechanism of action. Method: ICR mice were randomly divided into a normal control group (untreated non-immunosuppressed, Control), untreated immunosuppressed group (Model), positive control group (immunosuppressed and treated with 1.6 g/kg astragalus granule [AG]), high-dose KG group (immunosuppressed and treated with 24 g/kg, KG-24), and low-dose KG group (immunosuppressed and treated with 6 g/kg, KG-6). Each group received intragastric administration once daily for 7 days. Immunosuppression was induced by an intraperitoneal injection of dexamethasone (25 mg/kg) once daily beginning on day 1 for 3 days. To illuminate the mechanism of immunomodulatory, we studied the effects of KG on nonspecific immunity, humoral immunity and cellular immunity in mice respectively. Results: KG improved organ weights and improved the phagocytic ability of mononuclear macrophages in immunosuppressed mice (p p humoral immunity, and cellular immunity were all enhanced.
