Down-Regulation of CD44 Contributes to the Differentiation of hl-60 cells Induced by ATRA or HMBA

CD44 is highly expressed in human acute myeloid leukemia （AML） cells. Some experiments had shown that it was possible to reverse differentiation blockage in AML cells by CD44 ligation with specific antibodies, indicating that CD44 was closely related to the differentiation of leukemia cells. The differentiation of acute promyelocytic leukemia cell line hl-60 cells could be induced by all trans-retinoic acid （ATRA） and hexamethylene bisacetamide （HMBA）, but so far the mechanism was not demonstrated clearly. In the present study, we investigated whether ATRA or HMBA induced the growth arrest of hl-60 cells by down-regulating the expression of CD44. The results indicated that the proliferation of hl-60 cells was obviously inhibited and the differentiation was induced by both ATRA and HMBA. The decreased expression of CD44 and cyclin E mRNA, and the increased expression of p27 and p21 at mRNA levels were observed. Furthermore, there was a negative correlation between the expression of CD44 and p27. It was concluded that ATRA and HMBA played a role in the differentiation induction of hl-60 cells, which was mediated by the down-regulation of CD44, accompanied by down-regulation of cyclin E, and up-regulation of p27 and p21 mRNA. cellular ＆ Molecular Immunology. 2007;4（1）：59-63.

In present study we studied the effect of all-trans retinoic acid(ATRA) and dimethylsulfoxide (DMSO) on the induction of apoptosis in hl-60 cell line. Based on morphological changes by Hochest 33342 staining and identification of internucleosomal DNA cleavage by gel electrophoresis, we observed aberrant nuclear chromatin eondensation and ladder-like pattern of DNA degradation. Using Flow Cytometric method, we found sub-G1 peak in RA-treated hl-60 cells starting 5 to 6 d after the initiation of the treatment. However, such an obvious apoptotic peak was not identified in DMSO-differentiated cells. Combining the research accomplished before, our study approves further that apoptosis could be a common mode of death of terminally differentiated hl-60 cells.

Objective:In order to study if Trichostatin A facilitate Arsenic Trioxide to induce differentiation of hl-60 cells and its underlying ***:the alteration in the cell proliferation was determined by MTT *** reduction assay and CD11b analysis by FACS were used to detect the differentiation of hl60 *** cycle arrest was measured by *** expressions of m RNA of P21,P27 and Cyclin D3,Cyclin E in hl60 cells after exposure to TSA and(or)Arsenic Trioxide were evaluated by semi-quantitative ***:It was found that the combinationof TSAand Arsenic Trioxide could inhibit proliferation and induce differentiation of hl60 cells obviously than Arsenic Trioxide *** up-regulationof the m RNA expression of P21,P27 but down-regulation of the expression of Cyclin D3,Cyclin E were related to the differentiation of ***:TSA has synergistic effect with Arsenic Trioxide in inducing differentiation of hl60 and it’s associated with the change of P21,P27 and Cyclin D3,Cyclin E expression.

报导了一系列多亚甲基双［酰胺］类化合物的合成，由－摩尔多亚甲基二甲酰氯分别和二摩尔２－氨基噻唑啉及５－氨基－１－甲基吡啶酮反应制得。测定了其体外对ＨＬ－６０人早幼粒白血病细胞的分化诱导活性，初步结果表明：Ｎ，Ｎ｀－双吡啶酮基六二甲酰胺和Ｎ，Ｎ｀－双噻唑啉基八亚甲基二甲酰胺分别在０．１ｍｍｏｌ／Ｌ和０．５ｍｍｏｌ／Ｌ浓度时，诱导分化百分率可达６０％。此浓度下细胞存活率分别为２６％及２２％，其有效诱导浓度比ＨＭＢＡ低十倍。
