The study by Cao et al aimed to identify early second-trimester biomarkers that could predict gestational diabetes mellitus(GDM)development using advanced proteomic techniques,such as Isobaric tags for relative and absolute quantitation isobaric tags for relative and absolute quantitation and liquid chromatography-mass spectrometry liquid chromatography-mass *** analysis revealed 47 differentially expressed proteins in the GDM group,with retinol-binding protein 4 and angiopoietin-like 8 showing significantly elevated serum levels compared to *** these findings are promising,the study is limited by its small sample size(n=4 per group)and lacks essential details on the reproducibility and reliability of the protein quantification methods ***,the absence of experimental validation weakens the interpretation of the protein-protein interaction network identified through bioinformatics *** study's focus on second-trimester biomarkers raises concerns about whether this is a sufficiently early period to implement preventive interventions for *** GDM risk during the first trimester or pre-conceptional period may offer more clinical *** its limitations,the study presents valuable insights into potential GDM biomarkers,but larger,well-validated studies are needed to establish their predictive utility and generalizability.

gestational diabetes mellitus(GDM)is a high-prevalence disease and diagnosed in middle *** are a series of fatty acid esters of carnitine and play important roles in fatty acid and carbohydrate ***,the role of acylcarnitine on the development of GDM remains *** case-control study involving 214 study participants(107 GDM cases and 107 matched controls)was conducted in a cohort,in China,from 2013 to *** levels of carnitine and 36 acylcarnitines in serum samples collected at the early stage of pregnancy were determined by using ultra-high performance liquid chromatography coupled with tandem mass *** associations of the levels of the 37 targeted compounds with GDM risk were investigated by using binary conditional logistic regression *** in acylcarnitine levels were observed 9–17 weeks before GDM *** increases in levels of propionyl-carnitine,malonyl-carnitine,isovaleryl-carnitine,palmitoyl-carnitine and linoleoyl-carnitine were associated with GDM risk with odds ratios(ORs)per standard deviation(SD)increment greater than 1(p<0.05),after adjustment for potential confounding factors(pre-pregnancy body mass index and parity).On the contrary,the increases of decanoyl-carnitine,decenoyl-carnitine,tetradecenoyl-carnitine,tetradecandienoylcarnitine levels were associated with the reduced risk for GDM(ORs per SD<1,p<0.05).To our knowledge,the present study is the largest case-control study to investigate the association between early-pregnancy acylcarnitine levels in serum and GDM *** findings add to the evidence for the association between acylcarnitine levels and GDM risk.

This study was conducted to examine the association of perfluoroalkyl substance（PFAS）exposure with gestational diabetes mellitus（GDM） risk and postpartum fasting blood *** used a 1：2 matched case–control study with 84 GDM subjects and 168 healthy pregnant women from Beijing, China. The maternal blood was collected at 1–2 days before delivery, and eight linear isomers and fourteen branched isomers were determined in maternal *** regression analyses were performed to evaluate the associations after adjusting for potential confounders. The median of the sum of levels of total PFASs was 4.24 ng/m L with a interquartile range（IQR） of 2.82–6.54 ng/m L. Although maternal PFAS exposure was not associated with risk of GDM, significant positive associations were observed between evaluated exposure to specific PFAS congeners and increasing blood glucose. The odds ratio（ORs） of the highest category of postpartum fasting blood glucose for perfluoro-1-metylheptylsulfonat（1 m-PFOS）, perfluoro-3/4-metylheptylsulfonat（3 m＋4 m-PFOS）, perfluoro-5-metylheptylsulfonat（5 m-PFOS）, and perfluorohexane sulfonate（PFHx S） were 2.03（95% CI： 1.09–3.77）, 1.93（95% CI：1.04–3.58）, 2.48（95% CI： 1.33–4.65）, and 2.26（95% CI： 1.21–4.21）, respectively, suggesting negative effects of maternal exposure to specific PFAS compounds on glucose metabolism.

Background Epidemiological studies examining the direct and indirect effects of gestational diabetes mellitus(GDM)on offspring early childhood developmental vulnerability are ***,the aims of this study were to estimate the direct and indirect effects of GDM(through preterm birth)on early childhood developmental *** We conducted a retrospective population-based cohort study on the association between gestational diabetes mellitus and early childhood developmental vulnerability in children born in Western Australia(WA)using maternal,infant and birth records from the Midwives Notification,Hospitalizations,Developmental Anomalies,and the Australian Early Development Census(AEDC)*** used two aggregated outcome measures:developmentally vulnerable on at least one AEDC domain(DV1)and developmentally vulnerable on at least two AEDC domains(DV2).Causal mediation analysis was applied to estimate the natural direct(NDE),indirect(NIE),and total(TE)effects as relative risks(RR).Results In the whole cohort(n=64,356),approximately 22%were classified as DV1 and 11%as DV2 on AEDC *** of the natural direct effect suggested that children exposed to GDM were more likely to be classified as DV1(RR=1.20,95%CI:1.10-1.31)and DV2(RR=1.34,95%CI:1.19-1.50)after adjusting for potential *** 6%and 4%of the effect of GDM on early childhood developmental vulnerability was mediated by preterm birth for DV1 and DV2,*** Children exposed to gestational diabetes mellitus were more likely to be developmentally vulnerable in one or more AEDC *** biological mechanism for these associations is not well explained by mediation through preterm birth.

Objective:To assess the effects of Thunbergia laurifolia ***(TLE)on gestational diabetes mellitus(GDM)in a rat ***:Thunbergia laurifolin *** were subjected to ethanolic *** vivo study,50 pregnant rats were randomly divided into 5 groups(10 for each):non-GDM group,GDM induced by streptozotocin(STZ,60 mg/kg i.p.),metformin(MET)100 mg/kg,TLE 50,and 500 mg/kg *** was performed on gestation day 7 until term(day 21).The effects of TLE on blood glucose,insulin levels,lipid profiles,liver enzymes,and maternal performances were *** in vitro study,the effect of TLE was examined using the organ bath for uterine force ***:In in vivo study,TLE significantly reduced blood glucose as compared to GDM(P<0.05)with gradually increased insulin *** effect was consistent with islets of Langerhans ***,the uterine muscular layer displayed a marked increase in fiber area in response to both doses as compared to GDM(P<0.05).Additionally,TLE significantly reduced total cholesterol,triglyceride,and alanine transaminase levels(P<0.05).Intriguingly,TLE also led to a notable augmentation in gravid uterus size,live fetuses count,and implantation numbers,while significantly reducing the post-implantation loss rate associated with fetal classification(P<0.05).Thus,GDM improvements were close to those produced by *** in vitro study,TLE exerted a concentration-dependent inhibition of spontaneous uterine contractility(half-maximal inhibition concentration=1.2 mg/L).This inhibitory effect extended to potassium chloride depolarization and oxytocin-mediated *** combined with its major constituent,rosmarinic acid,TLE produced an enhanced inhibitory effect(P<0.05).Conclusions:TLE ameliorated blood glucose levels,enhanced uterine muscular structure,and improved maternal and fetal performance in *** also displayed tocolytic *** findings underscore the need for further explor

The placenta plays an important role in nutrient transport to maintain the growth and development of the *** diabetes mellitus(GDM),the most common complication during pregnancy,highly affects placental function in late *** glycation end-products(AGEs),a complex and heterogeneous group of compounds engaged by the receptor for AGEs(RAGE),are closely associated with diabetes-related *** this study,AGEs induced a decrease in the expression of tight junction(TJ)proteins in BeWo cells and increased the paracellular permeability of trophoblast cells by regulating RAGE/NF-κ***-Dawley(SD)rats injected with 100 mg/kg AGEs-rat serum albumin(RSA)via the tail vein from embryo day 2 were set as the placental barrier dysfunction model group(n=10).The effect of AGEs on placental permeability was determined using the Evans-Blue dye extravasation *** ultrastructure of the placenta samples was observed by transmission electron *** effects of AGEs on the placenta were confirmed by treating rats with RAGE antagonist FPS-ZM1 and soluble forms of RAGE(sRAGE).AGEs treatment increased placental permeability and disrupted the tight junctions in pregnant rat placenta,but has no effect on blood *** expression of TJ-related proteins,including ZO-1,Occludin,and Claudin 5,were downregulated after AGEs ***,AGEs treatment increased the expression of RAGE and nuclear factor-κB in the placenta of rats and upregulated the levels of vascular endothelial growth *** effects of AGEs on the placenta were blocked by RAGE antagonist FPS-ZM1 and *** study demonstrates the mechanism underlying AGEs-induced disturbance in placental function in pregnant rats and highlights the potential of AGEs in the treatment of GDM.
