＜正＞Objective The dopamine（DA）system in the retina is critical to normal visual development as lack of retinal DA signalling may contribute to myopic *** involvement of DA in myopic development is complex and may be different between form deprivation and hyperopic *** study aims to explore the possible different roles of DA signaling between the FDM and defocus-induced myopia using guinea *** This study evaluated effects of a non-selective DA receptor agonist,apomorphine（APO）on refractive development in guinea pigs treated with form deprivation or hyperopic defocus（from age of 21 to 32 days）.Changes in retinal concentration of DA and its metabolites（DOPAC）were also measured in the 2 animal models in order to assess the level of DA involvement in each of the models（the less the change,the lower the involvement）.Results（1）Similar myopic degree was induced in both the deprived and defocused eyes（-4.06 D vs.-3.64 D）at 11 days of the experiment.（2）DA and DOPAC levels were reduced in the deprived eyes but did not change significantly in the defocused eyes compared to the fellow and normal control eyes.（3）A subconjunctival injection of APO（from day 1 to 11）at concentrations from 2.5 ng to 250 ng/eye reduced axial myopia treated by form deprivation in a concentration-dependent manner. （4）By contrast,the effectiveness of APO treatment（at concentration from 2.5 ng up to 25 mg/eye）was significantly lower in inhibition of the defocus-induced myopia and the associated axial *** DA signaling may play a more critical role in form deprivation myopia than in defocus-induced myopia,raising a question whether the mechanisms of DA signaling are different under these two types of experimental myopia.

Objective deprivation of form vision causes excessive axial eye growth and a myopic refractive error in juveniles of several ***,the brain may also be involved in the control of eye *** this work,we investigated whether the retina-brain link is necessary for form-deprivation myopia in mice with optic nerve crush（ONC）.Methods Eye refraction was measured by an automated eccentric infrared photorefractor and ocular components were measured by optical coherence tomography（OCT）.C57BL/6 mice（21-days-old） were monocularly deprived of form vision with white translucent occluders for 4 *** assess the role of brain in form-deprivation myopia,the right optic nerve was crushed in mice（18-days-old） 3 days before the right eyes were occluded for 4 weeks *** The occluded eyes with intact optic nerve showed a significant myopic shift（-5.55±1.83 D）,relative to the contralateral fellow eyes,after 4-week visual *** mice with ONC（but without occluders）,the injured eyes became either myopic or hyperopic,compared to the fellow *** the group with myopic-shift（-5.73±4.81D）,the injured eyes exhibited a significant decrease in both lens and retinal thickness（P ＜0.05）.In the group with hyperopic-shift（4.59±3.03 D）,the injured eyes showed a decrease in corneal and retinal dnckness（P ＜0.05）.In mice with ONC,the occluded eyes all became myopic,compared to the fellow eyes,with a myopic shift（-9.69±4.17 D） even larger than that（-5.55±1.83 D）observed in occluded eyes in mice with intact optic ***,these occluded eyes exhibited a significant increase in axial length and vitreous chamber depth（P ＜0.05）,and a decrease in retinal thickness（P ＜0.001）,lens thickness and corneal radius of curvature（P ＜0.05）.Conclusion Intact eye-brain link is indispensable for normal ocular growth and refractive *** centers are involved in the development of myopia due to form deprivation.

Background:The use of ocular hypotensive drugs has been reported to attenuate myopia *** study explores whether brimonidine can slow myopia progression in the guinea pig form-deprivation(FD)***:Three-week-old pigmented male guinea pigs(Cavia porcellus)underwent monocular FD and were treated with 3 different methods of brimonidine administration(eye drops,subconjunctival or intravitreal injections).Four different concentrations of brimonidine were tested for intravitreal injection(2μg/μL,4μg/μL,20μg/μL,40μg/μL).All treatments continued for a period of 21 ***,retinoscopy,and A-scan ultrasonography were used to monitor intraocular pressure(IOP),refractive error and axial length(AL),*** day 21,guinea pigs were sacrificed for RNA sequencing(RNA-seq)to screen for associated transcriptomic ***:The myopia model was successfully established in FD animals(control eye *** eye,respectively:refraction at day 20,0.97±0.18 D vs.−0.13±0.38 D,F=6.921,P=0.02;AL difference between day 0 and day 21,0.29±0.04mm vs.0.45±0.03 mm,F=11.655,P=0.004).Among the 3 different brimonidine administration methods,intravitreal injection was the most effective in slowing myopia progression,and 4μg/μL was the most effective among the four different concentrations of brimonidine intravitreal injection *** AL and the refraction of the brimonidine intravitreal injection group was significantly shorter or more hyperopic than those of other 2 ***μg/μL produced the smallest difference in AL and spherical equivalent difference *** treatment significantly increased the *** was significantly lower at 1 day after intravitreal injections which was the lowest in FD eye of intravitreal injection of *** day 21,gene expression analyses using RNA-seq showed upregulation of Col1a1 and Mmp2 expression levels by intravitreal ***:Among the 3 different administration methods,intravitreal injection of brimonid
