Amyloid-b,tau pathology,and biomarkers of neurodegeneration make up the core diagnostic biomarkers of Alzheimer disease(AD).However,these proteins represent only a fraction of the complex biological processes underlying AD,and individuals with other brain diseases in which AD pathology is a comorbidity also test positive for these diagnostic biomarkers.More ADspecific early diagnostic and disease staging biomarkers are needed.In this study,we performed tandem mass tag proteomic analysis of paired cerebrospinal Fluid(CSF)and serum samples in a discovery cohort comprising 98 participants.Candidate biomarkers were validated by parallel reaction monitoring–based targeted proteomic assays in an independent multicenter cohort comprising 288 participants.We quantified 3,238 CSF and 1,702 serum proteins in the discovery cohort,identifying 171 and 860 CSF proteins and 37 and 323 serum proteins as potential early diagnostic and staging biomarkers,respectively.In the validation cohort,58 and 21 CSF proteins,as well as 12 and 18 serum proteins,were verified as early diagnostic and staging biomarkers,respectively.Separate 19-protein CSF and an 8-protein serum biomarker panels were built by machine learning to accurately classify mild cognitive impairment(MCI)due to AD from normal cognition with areas under the curve of 0.984 and 0.881,respectively.The 19-protein CSF biomarker panel also effectively discriminated patients with MCI due to AD from patients with other neurodegenerative diseases.Moreover,we identified 21 CSF and 18 serum stage-associated proteins re-flecting AD stages.Our findings provide a foundation for developing bloodbased tests for AD screening and staging in clinical practice.

Cancer of unknown primary(CUP)is a rare disease characterized by metastases in which the primary tumor is of unknown origin.The cerebrospinal Fluid(CSF)tumor microenvironment of CUP is still unknown.A Chinese male was diagnosed with leptomeningeal metastases from CUP(CUP-LM)based on the following medical examination results:partial leptomeningeal enhancement by brain magnetic resonance imaging,few malignant cells of diverse morphology in CSF,and no abnormalities or lymphadenopathy by systemic examination.

A modular microFluidic platform to study how Fluid shear stress alters estrogen receptor phenotype in ER+breast cancer cells

Metastatic breast cancer leads to poor prognoses and worse outcomes in patients due to its invasive behavior and poor response to therapy.It is still unclear what biophysical and biochemical factors drive this more aggressive phenotype in metastatic cancer;however recent studies have suggested that exposure to Fluid shear stress in the vasculature could cause this.In this study a modular microFluidic platform capable of mimicking the magnitude of Fluid shear stress(FSS)found in human vasculature was designed and fabricated.This device provides a platform to evaluate the effects of FSS on MCF-7 cell line,an estrogen receptor positive(ER+)breast cancer cell line,during circulation in the vessels.Elucidation of the effects of FSS on MCF-7 cells was carried out utilizing two approaches:single cell analysis and bulk analysis.For single cell analysis,cells were trapped in a microarray after exiting the serpentine channel and followed by immunostaining on the device(on-chip).Bulk analysis was performed after cells were collected in a microtube at the outlet of the microFluidic serpentine channel for western blotting(off-chip).It was found that cells exposed to an FSS magnitude of 10 dyn/cm^(2) with a residence time of 60 s enhanced expression of the proliferation marker Ki67 in the MCF-7 cell line at a single cell level.To understand possible mechanisms for enhanced Ki67 expression,on-chip and off-chip analyses were performed for pro-growth and survival pathways ERK,AKT,and JAK/STAT.Results demonstrated that after shearing the cells phosphorylation of p-AKT,p-mTOR,and p-STAT3 were observed.However,there was no change in p-ERK1/2.AKT is a mediator of ER rapid signaling,analysis of phosphorylated ERαwas carried out and no significant differences between sheared and non-sheared populations were observed.Taken together these results demonstrate that FSS can increase phosphorylation of proteins associated with a more aggressive phenotype in circulating cancer cells.These findings provide additional information that may help

This article presents an in-depth exploration of the acoustoFluidic capabilities of guided flexural waves(GFWs)generated by a membrane acoustic waveguide actuator(MAWA).By harnessing the potential of GFWs,cavity-agnostic advanced particle manipulation functions are achieved,unlocking new avenues for microFluidic systems and lab-on-a-chip development.The localized acoustoFluidic effects of GFWs arising from the evanescent nature of the acoustic fields they induce inside a liquid medium are numerically investigated to highlight their unique and promising characteristics.Unlike traditional acoustoFluidic technologies,the GFWs propagating on the MAWA’s membrane waveguide allow for cavity-agnostic particle manipulation,irrespective of the resonant properties of the Fluidic chamber.Moreover,the acoustoFluidic functions enabled by the device depend on the flexural mode populating the active region of the membrane waveguide.Experimental demonstrations using two types of particles include in-sessile-droplet particle transport,mixing,and spatial separation based on particle diameter,along with streaming-induced counter-flow virtual channel generation in microFluidic PDMS channels.These experiments emphasize the versatility and potential applications of the MAWA as a microFluidic platform targeted at lab-on-a-chip development and showcase the MAWA’s compatibility with existing microFluidic systems.

The addition of surface acoustic wave(SAW)technologies to microFluidics has greatly advanced lab-on-a-chip applications due to their unique and powerful attributes,including high-precision manipulation,versatility,integrability,biocompatibility,contactless nature,and rapid actuation.However,the development of SAW microFluidic devices is limited by complex and time-consuming micro/nanofabrication techniques and access to cleanroom facilities for multistep photolithography and vacuum-based processing.To simplify the fabrication of SAW microFluidic devices with customizable dimensions and functions,we utilized the additive manufacturing technique of aerosol jet printing.We successfully fabricated customized SAW microFluidic devices of varying materials,including silver nanowires,graphene,and poly(3,4-ethylenedioxythiophene)polystyrene sulfonate(PEDOT:PSS).To characterize and compare the acoustic actuation performance of these aerosol jet printed SAW microFluidic devices with their cleanroom-fabricated counterparts,the wave displacements and resonant frequencies of the different fabricated devices were directly measured through scanning laser Doppler vibrometry.Finally,to exhibit the capability of the aerosol jet printed devices for lab-on-a-chip applications,we successfully conducted acoustic streaming and particle concentration experiments.Overall,we demonstrated a novel solution-based,direct-write,single-step,cleanroom-free additive manufacturing technique to rapidly develop SAW microFluidic devices that shows viability for applications in the fields of biology,chemistry,engineering,and medicine.

Techniques for high-definition micromanipulations,such as optical tweezers,hold substantial interest across a wide range of disciplines.However,their applicability remains constrained by material properties and laser exposure.And while microFluidic manipulations have been suggested as an alternative,their inherent capabilities are limited and further hindered by practical challenges of implementation and control.Here we show that the iterative application of laser-induced,localized flow fields can be used for the relative positioning of multiple micro-particles,irrespectively of their material properties.Compared to the standing theoretical proposal,our method keeps particles mobile,and we show that their precision manipulation is non-linearly accelerated via the multiplexing of temperature stimuli below the heat diffusion limit.The resulting flow fields are topologically rich and mathematically predictable.They represent unprecedented microFluidic control capabilities that are illustrated by the actuation of humanoid micro-robots with up to 30 degrees of freedom,whose motions are sufficiently well-defined to reliably communicate personal characteristics such as gender,happiness and nervousness.Our results constitute high-definition micro-Fluidic manipulations with transformative potential for assembly,micro-manufacturing,the life sciences,robotics and optohydraulically actuated micro-factories.

Association of Fluid-attenuated inversion recovery vascular hyperintensity with ischaemic events in internal carotid artery or middle cerebral artery occlusion

Background and purpose Individuals with intracranial artery occlusion have high rates of ischaemic events and recurrence.It has been challenging to identify patients who had high-risk stroke using a simple,valid and non-invasive screening approach.This study aimed to investigate whether Fluid-attenuated inversion recovery(FLAIR)vascular hyperintensity(FVH),a specific imaging sign on the FLAIR sequence,could be a predictor of ischaemic events in a population with internal carotid artery(ICA)or middle cerebral artery(MCA)occlusion.Methods We retrospectively analysed 147 patients(mean 60.43±12.83 years)with 149 lesions,including 37 asymptomatic and 112 symptomatic cases of ICA or MCA occlusion.Symptomatic occlusion was considered if ischaemic events were present in the relevant territory within 90 days.FVH Alberta Stroke Program Early Computed Tomography Score(FVH-ASPECTS:0-7,with 0 indicating absence of FVH and 7 suggesting prominent FVH)and collateral circulation grade were assessed for each participant.Multivariable logistic regression analysis was performed to detect independent markers associated with symptomatic status.Results A lower FVH-ASPECTS was associated with a more favourable collateral circulation grade(rho=−0.464,p<0.0001).The FVH-ASPECTS was significantly lower in the asymptomatic occlusion group than in the symptomatic occlusion group(p<0.0001).FVH-ASPECTS(Odd ratio,2.973;95%confidence interval,1.849 to 4.781;p<0.0001)was independently associated with symptomatic status after adjustment for age,sex,lesion location and collateral circulation grade in the multivariate logistic regression.The area under the curve was 0.861 for the use of FVH-ASPECTS to identify symptomatic occlusion.Conclusions The ability to discriminate symptomatic from asymptomatic occlusion suggests that FVH may be a predictor of stroke.As a simple imaging sign,FVH may serve as a surrogate for haemodynamic impairments and can be used to identify high-risk stroke cases early in ICA or MCA occlusion.

Deformation behavior and Fluid action of quartz veins in the Xuelongshan metamorphic complex,Western Yunnan

The Xuelongshan metamorphic complex underwent Cenozoic sinistral strike-slip shearing and exhumation from deep to shallow in the southeast margin of the Tibet Plateau.We studied the deformation behavior and Fluid action of quartz veins in the metamorphic complex in depth based on macroscopic observation,electron backscatter diffraction fabric,and Fluid inclusion analysis.The mylonitic samples are characterized by strongly elongated and orientated porphyroclasts and a finegrained matrix with a shear strain of 2.4.The ultramylonitic samples are characterized by a strongly fine-grained matrix with a shear strain of 5.8.The{m}glide controls the quartz deformation in the mylonites and ultramylonites.As the dominant recovery mechanism,subgrain rotation dynamic recrystallization reduces the grain size and promotes rheological weakening.The porphyroclasts restrict the crystal orientation of the recrystallized quartz matrix.The mylonitic quartz veins have abundant single-phase and two-phase Fluid inclusions.However,the ultramylonitic quartz veins have a few Fluid inclusions,most of which are small and irregular toward the grain boundaries.The temperature and pressure of Fluid activity in quartz veins are 550–500℃and 391–218 MPa for ultramylonitic quartz veins,with a depth of 14.2–7.9 km and 450–400℃and 236–91 MPa for mylonitic quartz veins,with a depth of 8.6–3.3 km.The deformation process changes the shape of Fluid inclusions from mylonitization to ultramylonitization and causes Fluid leakage,an increase in local Fluid pressure at the boundary,and hydrolysis weakening,promoting lattice dislocation glide and recrystallization,which is directly related to regional continuous shear deformation and exhumation.

Introduction The ileocecal region is the end point of colonoscopy and comprises the junction of the ileum,cecum,and appendix.The ileocecal region has been reported as a common lesion site for various intestinal diseases(e.g.Crohn’s disease[1],intestinal tuberculosis[2],and primary colorectal lymphoma[3]).However,to date,there have been few studies on the intestinal microbiota of the ileocecal region,because the sampling is relatively difficult and with only a local biopsy it is difficult to represent the whole region without deviation.Compared with ileocecal mucosa sampling,the collection of ileocecal lavage Fluid is easier to be accepted by patients and can avoid the interference of human genomic DNA in the process of bacterial genome sequencing.

To the Editor:In neurosurgery,intraoperative Fluid administration is essential in maintaining appropriate systemic and cerebral perfusion pressure,oxygenation,and metabolism.Meningioma resection can involve massive intraoperative blood loss due to an abundant blood supply in structures.Moreover,the routine use of mannitol during meningioma resection may affect hemodynamic stability.Fluid overload is correlated to neurological,pulmonary,or gastrointestinal(GI)edema,which has unfavorable outcomes.[1]Thus,neurosurgeries,particularly meningioma resection,require individualized Fluid therapy to achieve a balance between inadequate Fluid and Fluid overload.
