d-psicose intake exacerbates dextran sulfate sodium-induced colitis in mice through alteration in the gut microbiota and dysfunction of mucosal barrier

d-psicose,as a low-calorie rare sugar,has attracted a lot of attention in recent years for alternating to *** anti-obesity effect of d-psicose has been extensively confirmed in previous studies,however,the impact of d-psicose on colitis remains ***,we firstly evaluated the effect of the d-psicose prophylactic intervention on dextran sulfate sodium-induced colitis in C57BL/6 *** pathological symptoms,inflammatory cytokines levels,gut microbiota composition,short chain fatty acids(SCFAs)production and colonic barrier integrity were comprehensively *** results confirmed that d-psicose intervention aggravated colitis,characterized by the exacerbation of colon shortening,increase of colonic inflammatory infiltration,and marked exaltation of disease activity indices and IL-6,IL-1βand TNF-α***,the dysfunction of gut microbiota was identified in the psicose *** abundance of pro-inflammatory bacteria Lachnospiraceae_NK4A136_group was significantly up-regulated while the abundance of probiotics Akkermansia and Lactobacillus were significantly down-regulated in the psicose group compared to the model ***,the production of SCFAs was suppressed in the psicose group,accompanied by a decrease in the level of mucin 2(Muc-2).Collectively,the underlying mechanism of the exacerbation of colitis by d-psicose intervention might be attributed to microbiota dysfunction accompanied by the reduction of SCFAs,which leads to the damage of the mucosal barrier and the intensifi cation of inflammatory invasion.

Bioproduction of d-psicose using permeabilized cells of newly isolated Rhodobacter sphaeroides SK011

d-psicose exits in an extremely small amount in nature and is difficult to be chemically *** three bacteria have been used in the biotransformation of d-psicose from *** this paper,another bacterium which could convert d-fructose to d-psicose was isolated and identified as Rhodobacter *** process parameters of d-psicose production using permeabilized cells of Rhodobacter sphaeroides SK011 were optimized,including the permeabilization procedure:0.1%(w/v)CTAB,10 min,and reaction conditions:cell concentration,30 g dry cell wt/L;concentration of substrate,50 g/L;40℃,pH 9.0;reaction time,8 *** the optimized conditions,the permeabilized cells produced approximately 6.5 g/L d-psicose with a dpsicose productivity of 0.82 g·L^(-1)·h^(-1).This is the first report of bioproduction of d-psicose using permeabilized cells of Rhodobacter sphaeroides.

The effects of 90-day feeding of d-psicose syrup in male wistar rats

d-psicose is a rare sugar present in small quantities in natural products. In a previous study, we showed that d-psicose suppresses increase in plasma glucose and reduces body fat accumulation in rats. Based on acute and chronic toxicity testing in rats, d-psicose is classified as an ordinary and safe substance. Recently, we developed a high d-psicose syrup (PS) made from high fructose corn syrup (HFCS) by the alkaline isomerization method. However, the safety of PS as a food additive has not been demonstrated. In this study, we investigated the effects of 90-day feeding of PS in male Wistar rats. The rats were fed diets containing 3% d-psicose (control) or 4.3% PS for 90 days. The body weight gain and intra-abdominal adipose tissue weight did not differ between the control and PS group. The weights of the tissues did not differ between the two dietary groups. In clinical chemistry and hematological analyses, no differences were found between the control and PS groups. No gross pathological findings were evident at dietary doses of 4.3% PS. Therefore, the present study found no adverse effects of PS in rats fed a diet containing 4.3% PS for 90 days.
