Development of the SCNT Embryo from Butyrolactone Ⅰ Prematured bovine Oocytes

The somatic cell nuclear transfer （SCNT） technique has been applied successfully in a range of mammalian species giving rise to offspring, however, the efficiency of development to term has remained low. Oocyte meiotic arrest is an important option to make the SCNT more flexible and increase the number of cloned embryos produced. This paper showed that the use of butyrolactone I to arrest the meiotic division for 24 h prior to in vitro maturation （IVM） provided bovine oocytes capability of supporting development of blastocysts as efficiently as non arrested oocytes. In the SCNT group, cleavage rates were not affected by prematuration when adding butyrolactone I during IVM （57.9% vs. 62.4%; control vs. 10 μmol.L^-1 butyrolactone I）, developmental rates to biastocyst were unaffected （22.0% vs. 20.3%; control vs. 10 μmol.L^-1 butyrolactone I, p〉0.05） by the addition ofbutyrolactone I during IVM in cloned embryos. Moreover, the total cell numbers of blastocyst were not affected by butyrolactone I （total nucleus numbers, 132±16.5 vs. 128±19.4, p〉0.05）. In conclusion, the SCNT embryos from butyrolactone I- prematured bovine oocytes had the same developmental potential as the non treated one. The present study provided a method for laboratories of in vitro embryos production, mainly those working on the SCNT, where prematuration could be used to iiacrease the flexibility of the procedure.

Aim: Malnutrition increases the severity and mortality of HIV infection. Therefore, this field study was started and aimed to provide a major cohort of HIV-infected children with the colostrum-based food product ColoPlus (ColoPlus AB, Malmö, Sweden) and to investigate the effects of ColoPlus on the nutritional status and immunological capacity of children including tolerability and safety of the product. Methods: In this major field program comprising 850 malnourished HIV positive children, 50 grams of ColoPlus was administrated for 4 weeks as the first meal in selected health facilities in Northern Uganda. Forty-eight of these children (8 months - 14 years of age) were recruited into a descriptive prospective study and were followed for 12 weeks. At the start (week 0) and at weeks 4, 8 and 12, CD4+ cell counts, serum albumin and hemoglobin were analyzed. The remaining 802 patients were observed to register safety and tolerability of ColoPlus. Results: There was a general improvement of wellbeing of the children with increased body weight and decreased fatigue. In the descriptive study of the 48 children, there was a significant rise of the CD4+ values at week 4 (+15.4% ± 2.8%, p = 0.0001) compared to week 0, and at week 8 (+39.1% ± 3.9%, p < 0.0001), but a return towards the 0-values at week 12 (+2.1%, ±2.8%, NS). Hemoglobin and serum albumin showed an almost similar trend. ColoPlus was well tolerated by all the 850 children and no side effects or adverse events were seen. Conclusion: These results show that addition of a colostrum-based food product to the daily diet is beneficial in HIV-positive malnourished children. An improvement in nutritional status as well as in immune capacity was seen. These effects were prolonged and remained at least 4 weeks after cessation of ColoPlus administration.

Objective:To investigate the relationship between inactive foot and mouth disease virus (FMDV) vaccination and result of gamma interferon (IFN-γ) assay for tuberculosis in cattle negative for bovine tuberculosis. Methods:A total of 300 tuberculosis-negative cattles were subcutaneously vaccinated with inactive FMD vaccine. Nine hundred blood samples were collected from the same 300 cattles after 7, 15, and 30 days of vaccination. Then, all samples were tested with IFN-γ assay for tuberculosis. Results:A total of 900 blood samples were tested with IFN-γ assay for tuberculosis and all of the samples were found negative for bovine tuberculosis. Conclusions:Our results indicate that inactive FMD vaccination in cattle do not affect results of IFN-γ assay for tuberculosis.

Penile reconstruction following cancer resection has long been a challenge due to demand for both aesthetic and functional preservation. Although various methods have attempted to achieve both goals, none have succeeded entirely. In this report, we introduce a novel reconstructive technique on a 64-year-old patient with squamous cell carcinoma of the glans penis. The technique consisted of two staged procedures. First, a dermal substitute, consisting of bovine collagen, was sutured onto the defect after partial glansectomy and three weeks later, the integrated dermis was covered with autologous non-meshed partial thickness skin graft. Use of this dermal substitute in a staged fashion allowed for ability to await final pathologic margins prior to definitive reconstruction and increased dermal thickness. After 9-month follow-up, sexual function returned within 3 months and the patient remained healed. We found this technique to be useful in concurrently preserving penile function and cosmesis. Future studies should include more patients and longer follow-up.

AIM: To investigate gastric antisecretory and gastro- protective activity of bovine hemoglobin (B-Hb) in rats. METHODS: Adult Albino-Wistar rats were divided into groups of 6 animals each. B-Hb in doses of 100, 300 and 900 mg/kg body weight was tested for gastric acid secretion and antiulcer activity. Gastric secretions were measured 6 h after pylorus ligation in rats pretreated with B-Hb. The acidity was measured by titrating gastric contents against 0.01 mol/L NaOH to pH 7. Indomethacin ulcers were produced by oral administration of 30 mg/kg bw in the rats pretreated with B-Hb one hour before indomethacin. Six hours after indomethacin stomach removed and ulcer index was recorded. Ethanol ulcer were produced by 1 mL of ethanol in the rats pretreated with B-Hb 30 min before the *** hour after ethanol stomach were cut open to score ulcers. Histological examination and analysis of gastric wall mucus, non-protein sulfhydryl groups (NP-SH), and myeloperoxidase (MPO) were carried in gastric tissue following ethanol administration. RESULTS: In control rats pylorus ligation for 6 h resulted in the accumulation of 8.1 ± 0.61 mL of gastric secretion. The treatment of the rats with 100, 300 and 900 mg/kg of B-Hb produced a significant decrease in the volume of gastric secretion 5.6 ± 0.63, 5.5 ± 0.75 and 4.7 ± 0.58 mL respectively as compared to the control group [analysis of variance (ANOVA) F = 4.77, P < 0.05]. The lesion area in the control group was found to be 22.4 ± 3.2 mm2 six hours after the administration of indomethacin. Treatment of rats with B-Hb at doses of 100 mg/kg (24.3 ± 3.29 mm2), 300 mg/kg (16.2 ± 1.45 mm2) and 900 mg/kg (12.6 ± 1.85 mm2) produced a dose dependent decreased the lesion scores (ANOVA F = 4.50, P < 0.05). The ulcer index following one hour after 1 mL ethanol was 7.1 ± 0.31. Pretreatment of rats with B-Hb at the doses of 100 mg/kg (2.5 ± 0.42), 300 mg/kg (2.1 ± 0.4) and 900 mg/kg (0.7 ± 0.21) significantly inhibited the formation of gas

Fresh semen has a very limited life span after it leaves the gonad. The relative research shows that the epididymal plasma provides a special environment to spermatozoa for survival. In this study, the osmolarity and protein concentration of caput and caudal bull epididymis were analyzed. bovine caudal epididymal sperm were incubated in different protein and sorbitol combinations of CEP-2 diluents at 4 ℃ for 120 h and sperm viability was assessed. Under this condition, sperm viability was greater than 60% after 5 d. The osmolarity of caput and caudal epididymis were （287.0 ± 13.7） and （310.8 ± 17.0 ） mOsm, respectively. The protein concentration of caput and caudal epididymis were （ 37.43 ± 12.55 ） and （50.58±11.08） mg/mg, respectively. This may have an application in the animal production industry.
