Aim: The present study aimed to examine whether apipuncture (stimulation of acupuncture points with bee venom)at ST36 and GV3 acupoints promotes neuroprotection and reduces neuroinflammation by modulating M1 and M2 phenotype ***: Wistar rats were treated with bee venom (BV) (0.08 mg/kg) injection at acupoints ST36 and GV3 [BV (ST36 + GV3)-spinal cord injury (SCI)] or BV injection at non-acupoints [BV (NP)-SCI] or no treatment (CTL-SCI)after SCI by compression. The spinal cord mRNA expression of iNOS, Arg-1 and TGF-β was measured by real time PCR and the levels of IBA-1;BCL-2;NeuN e CNPase was measured by western blotting. Locomotor performance was measured by Basso, Beattie, and Bresnahan (BBB) and grid-walking ***: Apipuncture treatment was able to (1) ameliorate locomotor performance;(2) reduce inflammatory markers (Cox-2 levels) and activation of microglia and macrophages;(3) reduce the polarization of the M1 phenotype marker (iNOS) and increase M2 (Arg-1 and TGF-β) phenotypic markers;(4) promote neuroprotection by reducing the death of neurons and oligodendrocytes;and (5) increase the expression of the anti-apoptotic factor ***: Apipuncture treatment induces locomotor recovery and neuroprotection after the compression model of spinal cord injury. Further, it reduces neuroinflammation by decreasing M1 polarization and increasing M2 phenotype.

＜正＞Using tissue immunohistochemistry technique,the present study investigated the effects of subcutaneous（s.c.） bee venom（BV） administration on the expression of three major NMDA receptor subunits（NR1,NR2A,NR2B） in rat lumbar spinal cord,S1 area and *** to *** injection（100 ug/50 ul） into the hind paw,the animal exhibited a typical behavioral expression of persistent pain and hyperalgesia,whereas *** of equal volume of normal saline produced little *** coronal sections of lumbar spinal cord and brain were sequentially prepared at 4 h after BV or saline *** was then performed and positive labeling was visualized by treatment with diaminobenzidine（DAB） and hydrogen *** of NMDA receptor subunits expression were finished by comparisons between two sides of the hindpaw or comparisons between B V-treated and saline-treated groups. As shown in our results,expression of the NR2A subunit of NMDA receptor was significantly exaggerated in ipsilateral lumbar spinal cord,S1 area and hippocampus following *** but not saline *** significant changes were detected in the expression of NR1 and NR2B subunits in each ***,the number of NR2A-positive dendric tree also markedly increased in ipsilateral hippocampus of BV-treated ***,these data suggest that the NR2A subunit of NMDA receptor might behave as a candidate molecular detector in mediating nociceptive signal processing in the lumbar spinal cord,S1 area and hippocampus following BV-induced persistent pain and *** is also speculated that BV-evoked augmentation of NR2A-positive dendric tree in the hippocampus may bear some associative relationships with the synaptic plasticity occurred in this area.

In Vitro Study on the Effect of bee venom on Some Cell Lines and Lumpy Skin Disease Virus

bee venom （BV） was used from long time ago in the medical field as treatment of chronic joint affections. In the recent decades, the screening process of new sources of antimicrobials discovers its high advantageous characteristics for combating various types of microbes, as well as trials to discover its anti-cancer medicinal fields. Lumpy skin disease virus （LSDV） causes disease in cattle of economic importance, and this work aimed to find treatment as well as alternative inactivant for LSDV. The use of bee venom as antiviral was experimented in this work and exhibited satisfied inhibitory effects on LSDV, meanwhile, the antigenic properties was still intact. The viability of virus was tested in tissue culture cells lines and in embryonated chicken eggs. According to doses and time of exposure, the cell lines of Hep-2 （human larynx carcinoma） and MCF7 （breast carcinoma cell line） were treated with different concentrations of BV and examined after 24 h post-inoculation. The Hep-2 and MCF7 cell lines were treated with various concentrations of BV in descending doses as follow： 25, 20, 15, 10, 5 and 0.5 ug/mL of BV. Then bee venom pathological effects on Hep-2 cells and MCF7 cells were observed, such as apoptosis, retarded growths and cytolysis. The results indicate the possibilities of using bee venom as anti-neoplastic and antiviral.

＜正＞Recent advances have given us a much more detailed understanding of the roles played by MAPKs in the pathophysiogenesis of clinically-relevant pathological *** MAPK family has three major members,extracellular signal-regulated kinase（ERK）,p38 and c-Jun N-terminal kinase（JNK）,representing three different signal transduction *** with the enormous amounts of literature reporting activation and involvement of ERK,p38 or JNK in the process of inflammatory of neuropathic pain,fewer study has been performed to investigate possible regional differences in activation of different MAPK isoforms following peripheral noxious ***,our lab has conducted series of preliminary work to address this issue using Western blot technique in the bee venom tonical pain *** found that,in bee venom-treated rats,phosphorylation of two ERK isoforms（ERK 1 and ERK2） was significantly enhanced for a long period（lasting for at least 48 h） in either the spinal cord dorsal horn or the primary somatosensory cortex（S1 area） without any significant changes in total expression ***,isoform-dependent differences were also detected between these two ***,ERK2 exhibited stronger activation than ERK1 in the spinal cord,whereas ERK1 was more remarkably phosphorylated than ERK2 in the S1 *** above-described results indicate that,under pain state, either ERK1 or ERK2 can be effectively phosphorylated with a long-term duration,but their response patterns differ from each other across distinct *** another set of immunolabeling studies,we evaluated potential different activation patterns of two JNK isoforms（JNK 46 KDa and JNK 54 KDa） in the spinal cord and S1 area of cortex following subcutaneous （s.c.） bee venom-induced persistent ***,we found that *** venom injection markedly elevated the phosphorylation level of both JNK isoforms in either region for a long time course（lasting for at least 48 h） without affecting their tota

A novel peptide from Apis mellifera and solid-phase synthesis of its analogue

A novel peptide designated secapin-1, was purified and characterized from Apis mellifera. The molecular weight of 25 amino acid peptide secapin-1 was found to be 2821.5625 Da by ESI-FTICR-MS. It showed high identity to secapin. The sequence of secapin-1 was determined to be YIINVPPRCPPGSKFVKNKCRVIVP by automatic Edman degradation. A disulfide bond was formed between Cys9 and Cys20 residues. In addition, an analogue of secapin-1 was synthesized by solid phase peptide synthesis method. The synthesis product was successfully purified and identified to homogeneity by using a combination of SEC, IEC, and RP-HPLC techniques.

Effect of natural extract eye drops in dry eye disease rats

AIM:To investigate the therapeutic effect of natural extract eye drops containing bee venom,musk,and deer antlers in dry eye disease(DED)animal ***:Scopolamine-injected DED rats and lacrimal gland-excised rats were allocated into control,saline,and natural extract groups respectively and a normal group(lacrimal gland excision was not performed)in lacrimal gland-excised *** eye drop instillation 4 times a day for 5 d,corneal fluorescein staining(CFS)scores,tear MUC5AC levels,and tear lactic dehydrogenase(LDH)levels were ***:In scopolamine-injected rats,the natural extract-treated group had significantly lower CFS scores(1.7±0.5,4.7±1.4,3.8±1.9,P=0.006)and tear LDH levels(0.10±0.01,0.19±0.01,0.16±0.08 OD,P=0.014)but higher tear MUC5AC levels(12.9±3.7,7.9±2.0,9.7±3.6 ng/m L,P=0.041)compared with the control and saline-treated *** were no significant differences between the control and saline-treated *** lacrimal gland-excised rats,the natural extract-treated group also had lower CFS scores(4.3±1.2,11.5±2.3,9.0±1.9,Pbee venom,musk,and deer antlers may have effectiveness in DED treatment by restoring the damaged ocular surface,increasing tear volume,and recovering the tear mucin layer in DED rats.

＜正＞Pain is a multi-dimentional experience consisting of sensory-discriminative,affective-motivational,and cognitive -evaluative *** the present presentation,we report how persistent inflammatory pain on one hindpaw can cause plastic changes in network synaptic responses in the dentate gyrus（DG） obtained by multi-electrode array（Med64 system,Alpha-Med Sciences Co.,Ltd.） recordings on acute hippocampal slices of rats receiving subcutaneous injection of bee venom which mimics honeybee sting on *** naive or saline-control rats,electrical test stimulation（40-200 mA） at molecular layer of the DG where the perforant path fibers terminate could evoke widespread network synaptic responses shown as field excitatory postsynaptic potentials（fEPSPs）.The evoked network fEPSPs were mostly confined to the region of DG spatially with either positive- or negative-going fast PS wave followed by a small and slow oppositegoing *** network responses were intensity-dependent and could be eliminated by perfusion with TTX,high magnesium-low calcium and CNQX,respectively,but not AP5,suggesting involvement of both pre-synaptic transmitter release and postsynaptic activation of glutamate non-NMDA receptors in production of network ***,in rats suffering from persistent pain stimulation,the second small and slow wave of the evoked network fEPSPs was becoming sharp and bigger in amplitude which were becoming more sensitive to AP5 and bicuculline than naive and saline-control rats,suggesting involvement of glutamate NMDA and GAB AA receptors in the change of *** long-term potentiation of network synaptic responses was also significantly enhanced by persistent pain *** together,the present results add a new line of evidence showing strong impact of persistent pain experience on the network synaptic functions of the hippocampal formation.

＜正＞Objective:The present study was designed to investigate the effects of subcutaneous saline or bee venom injection on responses of the motor cortical neurons and those of behaviors in conscious,behaving ***:We examined the motor cortical neuronal activation with an in vivo multi-channel recording technique before and after saline or bee venom was delivered subcutaneously（s.c.） into the plantar surface of the hind paw contralateral to the recording ***:A total of 81 single neurons were recorded in the motor *** first analyzed the spatial sequence *** saline-induced phasic pain state,29（36%） neurons showed significant excitatory responses,whereas 19（23%） displayed significant inhibitory *** bee venom-induced tonic pain state,52（64%） indicated significant excitatory responses,however,7（9%） showed inhibitory *** then analyzed the temperoal sequence *** terms of saline-induced phasic nociception,a sharp increase of spike discharges at the start of saline injection was *** terms of bee venom-induced tonic pain state,the increase of spike discharges was during the initial half hour after *** venom ***:The present result,a microcosmic view of the brain pain-related responses,accorded well with the results of neuroimaging studies,a macroscopic view of the brain pain-related responses,indicating the role of the motor cortex in the information processing of pain.
