Signaling transduction pathways involved in basophil adhesion and histamine release

Background Little is known about basophil with respect to the different signaling transduction pathways involved in spontaneous, cytokine or anti-IgE induced adhesion and how this compares to IgE-dependent and IgE-independent mediator secretion. The purpose of the present study was to investigate the roles of β1 and β2 integrins in basophil adhesion as well as hosphatidylinositol 3-kinase （PI3K）, src-kinases and extracellular signal regulated kinase （ERK）1/2 in basophil adhesion and histamine release （HR）. Methods basophils （purity of 10%-50%） were preincubated with anti-CD29 or anti-CD 18 blocking antibodies before used for adhesion study. basophils were preincubated with the pharmacological inhibitors wortmannin, PP1, PD98059 before used for adhesion and HR study. Cell adherence to bovine serum albumin （BSA） or fibronectin （Fn） was monitored using cell associated histamine as a basophil marker and the histamine was measured by the glass fiber assay. Results basophil spontaneous adhesion to Fn was inhibited by anti-CD29. Interleukin （IL）-3, granulocyte/macrophage colony stimulating factor （GM-CSF） induced adhesion to BSA was inhibited by anti-CD18. Wortmannin at 1 μmol/L and PP1 at 20 μmol/L strongly interfered with, whereas PD98059 at 50μmol/L weakly inhibited basophil spontaneous adhesion to Fn. One μmol/L wortmannin strongly inhibited IL-3, IL-5, GM-CSF and anti-IgE induced adhesion to BSA. PP1 at 20 μmol/L partly inhibited anti-IgE induced adhesion. Fifty μmol/L PD98059 marginally inhibited IL-5, weakly inhibited anti-IgE, partly inhibited GM-CSF induced adhesion. Wortmannin, PP1 and PD98059 inhibited anti-IgE （1：100 or 1：1000） induced basophil HR in a dose dependent manner. They inhibited calcium ionophore A23187 （10 μmol/L, 5 μmol/L） induced basophil HR in a dose dependent manner, but to different extend with PP1 being the most efficient. Objective:To investigate the activation state of peripheral basophils from RA patients and its mechanism,and the relationship between disease activity and imbalance of Th1/Th2 ***:In this study,flow cytometry was employed to test the ratio of the number of Th1 to Th2 cell and levels ofbasophil and its activation state in peripheral blood mononuclear cells(PBMC)cells from rheumatoid arthritis(RA).ELISA method was used to measure serum levels of IFN-γand *** Blot was employed to detect serum circulating immune complexes containing Ig E(Ig E-CIC).Results:The ratio of the number of Th1 to Th2 cell have a low linear correlation with the DAS28 score;Serum levels of IFN-γin patients with RA was significantly higher than the control group,and correlated with the disease activity,but not IL-4;Levels of basophils of RA patients was significantly reduced compared with the control group,and highly activated(high expression of CD203c and CD62L);The presence of high levels of Ig E-CIC in RA patients,but not in control ***:Peripheral basophil of RA patients was highly activated and significantly reduced,may migrate to secondary lymphoid tissues and sites of inflammation,and against the imbalance of Th1 response,which may provide new clues for the prevention and treatment of RA.

Th1/Th2 imbalance plays a crucial role in the development of rheumatoid *** is well known that basophil can affect Th1/Th2 balance by enhancing Th2 response and impairing Thl response then be involved in the development of many *** little is known the role of basophil in rheumatoid *** circulating basophil and Th1response dominated in adult rheumatoid arthritis patients have been reported,while children with juvenile rheumatoid arthritis have been found mainly increased circulating basophil and Th2 response ***,circulating basophil in both of them are activated and associated with disease *** addition,longitudinal study found Th2 response dominatedin the early stage of rheumatoid arthritis,while Thl response dominated in long-term chronic rheumatoid *** finding suggested that basophil maybe involved in the development of rheumatoid arthritis by affect Thl/Th2 balance,especially in the early stage of RA,maybe a new therapeutic strategy in RA and need further investigation.

Background: Allergic diseases are orchestrated by dysregulated Th2-type immune responses involving multiple immune effector cells. basophils are one of the key effector cells in mediating allergic immune responses through releasing cytokines and mediators e.g. IL-4, IL-13 and histamine. Short chain fatty acids(SCFAs) produced by commensal intestinal microbiota via fermentation of indigestible dietary fibers are reported to suppress allergic immune reactions by targeting mast cells, eosinophils and type 2 innate lymphoid cells(ILC2). However, the role of SCFAs on basophils are not known yet. In this study, we aimed to elucidate the function of SCFAs on human ***: Human basophils were purified from healthy donors by FACS sorting. The expression of surface receptors and the apoptosis were analyzed by flow cytometric analysis. The cytokines IL-4 and IL-13 and histamine release were measured by ***: Human basophils express SCFAs receptor GPR41, which was enhanced by IL-3 rather than other established modulators e.g. anti-IgE, fMLP or C5a. Acetate but not propionate and butyrate induced calcium influx via GPR41. However, SCFA propionate and butyrate rather than acetate induced the apoptosis of basophils. In vitro cytokine release assay showed that propionate and butyrate suppressed the expression of IL-4 while acetate, propionate and butyrate enhanced the expression of IL-13. Moreover, propionate and butyrate enhanced the histamine release associated with increased expression of the basophil degranulation marker ***: Our findings demonstrate that a complicated role of SCFAs in mediating the biology of basophils. The effects of SCFAs in regulating basophils-associated allergic disorders need to be further justified.

Objective:To investigate the mechanisms of peripheral basophils decreasing in SLE patients and further clarify the role of basophils in the pathogenesis of ***:Fifty SLE patients were selected and divided into active group(SLEDAI score,10,n=27)and inactive group(SLEDAI scorebasophils were detected by flow *** levels of anti-dsDNA antibody were assayed by enzyme-linked immunosorbent assay(ELISA).Additionally,the migration of basophils in skin lesions of SLE patients was immunohistochemically analyzed using basophilspecific antibody 2D7,and the migration of basophils to lymph nodes of MRL/lpr mice were detected by flow cytometry(CD49bIgE).Results:Expression of Cleaved Caspase-3 in peripheral basophils from SLE group were significantly higher than that of control group,while there were no significant difference between SLE active group and SLE inactive *** with control group,the rates of Fas expression sigificantly increased in patients of active SLE;Expression of Cleaved Caspase-3 in peripheral basophils were negatively correlated with serum levels of anti-dsDNA antibody in active SLE patiens;basophils were found in skin lesions of SLE patients staining with 2D7 positive which appears most in the superficial dermis and the inflammatory sites and exhibited both discrete cellular localization and diffuses extracellular staining,while it was not seen in ***,the migration of basophils to lymph nodes were found in MRL/lpr mice,but not in C57BL/6 ***:Migration of basophils to lymph nodes or skin lesions and acceleration of basophils apoptosis participated in the decrease of peripheral basophils in SLE patients,but the role of migrating of basophils in SLE need to be further studied.

We studied the RSV specific IgE antibody, histamine and basophil from infants with RSV bronchiolitis and found during the acute phase either the titers of RSV-IgE or the concentration of histamine increased significantly, the number of basophil and basophil degranulation in the presence of RSV antigen also increased. In vitro studies revealed hypersensitivity participates in the pathogenesis of RSV bronchiolitis. We also found that infants with RSV bronchiolitis, the RSV-IgE persisted for a long time presumably this plays an important role in recurrent wheezing after RSV infection for years.

Staining Effect of Pomegranate Flower Extract on Human Blood Cells： First Results

Pomegranate flower plant, popularly known using for the treatment of various diseases, was not investigated as a source of dye for cytological studies using human blood cells. The importance of this study is to appear dyeing result of pomegranate flower extract on human blood cells. The natural dye source was pomegranate flower known as roselle and potassium aluminum sulfate （alum = KAISO4＇ 12H20） was used as mordant or metal salt. Distilled water was used as solvent. Fresh, clean and air-dried flowers were extracted with distilled water at 100℃ for 30 minutes and then filtered. One drop blood from a healthy 20-year woman was spread as a peripheral on to ten plates and dried at nearly 25℃. These slides were stained by soaking in pomegranate flower extract with/without alum （KA1SO4·12H2O） at 100℃ for 60 minutes. Slides were washed with distilled water, dried and done microscobic examination. The different blood cells dyed dark orange in alum mordant media at 100 ℃. As a result, pomegranate flower has the capacity to use dyeing human blood cells such as eosinophil, basophil and neutrophil.

2019 coronavirus disease(COVID-19) presents as a newly recognized pneumonia that has brought about a global pandemic and is increasingly considered as a systemic *** investigated the clinical and laboratory features of recovered COVID-19 patients without pre-existing hematologic diseases at Wuhan No.1 ***-nine male and68 female Chinese patients were included with the median age at 64 years in the present ***(37.80%),monocytosis(51.97%),lymphocytopenia(25.20%),and anemia(51.97%) were the most common hematologic findings in our cohort,particularly in severe or critically ill *** levels of changes in leukocytes,neutrophils,lymphocytes,monocytes,eosinophils,basophils,platelets,hemoglobin levels,mean corpuscular volume(MCV),and mean cell hemoglobin concentration(MCHC) are overall associated with lung involvement,oxygen demand,and disease ***,changes of eosinophils(end hospitalization-baseline)(coefficients=10.32;95% CI=1.03-19.60,P=0.03) and basophils(Max-Min)(coefficients=71.43;95% CI=8.55-134.31,P=0.03) were independent predictors of delayed recovery in the hospital by the multivariate analysis in this recovered population.A variety of hematologic changes are associated with the severity and clinical outcome of recovered COVID-19 patients,which warrants further exploration of their underlying mechanisms.
