Mechanism of peripheral basophils decrease in systemic lupus erythematosus

摘      要:Objective:To investigate the mechanisms of peripheral basophils decreasing in SLE patients and further clarify the role of basophils in the pathogenesis of ***:Fifty SLE patients were selected and divided into active group(SLEDAI score,10,n=27)and inactive group(SLEDAI score10,n=23).Normal health as control *** Caspase-3 and Fas expression of peripheral basophils were detected by flow *** levels of anti-dsDNA antibody were assayed by enzyme-linked immunosorbent assay(ELISA).Additionally,the migration of basophils in skin lesions of SLE patients was immunohistochemically analyzed using basophilspecific antibody 2D7,and the migration of basophils to lymph nodes of MRL/lpr mice were detected by flow cytometry(CD49bIgE).Results:Expression of Cleaved Caspase-3 in peripheral basophils from SLE group were significantly higher than that of control group,while there were no significant difference between SLE active group and SLE inactive *** with control group,the rates of Fas expression sigificantly increased in patients of active SLE;Expression of Cleaved Caspase-3 in peripheral basophils were negatively correlated with serum levels of anti-dsDNA antibody in active SLE patiens;Basophils were found in skin lesions of SLE patients staining with 2D7 positive which appears most in the superficial dermis and the inflammatory sites and exhibited both discrete cellular localization and diffuses extracellular staining,while it was not seen in ***,the migration of basophils to lymph nodes were found in MRL/lpr mice,but not in C57BL/6 ***:Migration of basophils to lymph nodes or skin lesions and acceleration of basophils apoptosis participated in the decrease of peripheral basophils in SLE patients,but the role of migrating of basophils in SLE need to be further studied.
