Characterization of Salmonella Enterica Serotype Typhimurium from Outpatients of 28 Hospitals in Henan Province in 2006

Objective To characterize the diarrheal patients with Salmonella typhimurium （S. typhimurium） infections and to set up the first baseline for S. typhimuri_um pulsed-field gel electrophoresis （PFGE） patterns in Henan province, thus laying a foundation for comprehensive surveillance of Salmonella in human as well as foods. Methods S. typhimurium isolates recovered from outpatients with diarrhea in Henan province from May to October of 2006 were characterized. Antimicrobial susceptibility tests of 8 antimicrobial agents and PFGE were carried out to analyze the S. typhimurium isolates. results Twenty-four （0.9%） S. typhimurium isolates were identified from 2661 stool specimens of diarrheal cases. Eighty-eight percent of isolates showed resistance to at least one antimicrobial agent. The resistance to chloramphenicol （79%） was most common. Fifty-eight percent of isolates were resistant to ciprofloxacin. All the 14 ciprofloxacin-resistant isolates were resistant to more than five antimicrobial agents. Thirty-three percent of S. typhimurium isolates were resistant to ampicillin, chloramphenicol, streptomycin, sulfamethoxazole, and tetracycline （R-type ACSSuT）. Eight antimicrobia-resistant phenotypes were found among the 24 isolates in 16 PFGE patterns. Conclusion The rate of multidrug-resistant S. typhimurium is relatively high in S. typhimurium PFGE patterns of Henan province. Multidrug-resistant S. typhimurium should be considered a public health threat.

Introduction:Human noroviruses are the leading cause of acute viral gastroenteritis(AGE)worldwide in all age ***.4 strains have been the predominant genotype circulating globally over the last 2 decades and since ***.4 Sydney viruses have emerged and caused the majority of AGE outbreaks ***:Data from norovirus outbreaks from the laboratory-based surveillance of norovirus outbreaks in china(CaliciNet china)between October 2016–December 2020 were ***:During October 2016–December 2020,1,954 norovirus outbreaks were reported,and positive fecal samples from 1,352(69.19%)outbreaks were ***.4 Sydney[P31]viruses accounted for 2.1%(October 2016–August 2017),5.5%(September 2017–August 2018),3.3%(September 2018–August 2018),26.6%(September 2019–August 2020),and and 1.1%(September 2020–December 2020)of GII outbreaks,*** to reference strains of GII.4 Sydney[P31]from 2012 to 2013,7 amino acid mutations in epitopes[A(297,372 and 373),B(333),E(414),and H(309 and 310)]and 1 in human histo-blood group antigens binding site at site II 372 were found by analyzing 9 GII.4 Sydney[P31]complete genomic ***:This report identified the genomic variation of GII.4 Sydney[P31]from CaliciNet *** surveillance with prompt genotyping and genetic analysis is necessary to monitor the emergence of novel GII.4 variants.

A Novel reassortant H3N8 Influenza Virus Isolated from Drinking Water for Duck in a Domestic Duck Farm in Poyang Lake Area

Objective To conduct a full genome sequence analysis for genetic characterization of an H3N8 influenza virus isolated from drinking water of a domestic duck farm in Poyang Lake area in 2011. Methods The virus was cultivated by specific pathogen free （SPF） chicken embryo eggs and was subtyped into hemagglutinin （HA） and neuraminidase （NA） by real-time PCR method. Eight gene segments were sequenced and phylogenetic analysis was conducted. results The NA gene of this virus belongs to North American lineage; other seven genes belong to Eurasian lineage. Compared with the viruses containing NA gene, the PB2 and PB2 gene came from different clades. And this indicates that the virus was a novel reassortant genotype. The HA receptor binding preference was avian-like and the cleavage site sequence showed a low pathogenic feature. There was no drug resistance mutation of M2 protein. The mutations of Asn30Asp, and Thr225Ala of the M1 protein implied the potential of pathogenicity increase in mice. Conclusion The finding of novel genotype of H3N8 virus in drinking water in this duck farm near Poyang Lake highlighted the importance of strengthening the surveillance of avian influenza in this region, which could contribute to pinpointing the influenza ecological relations among avian, swine, and human.

Current status of severe fever with thrombocytopenia syndrome in china

Severe fever with thrombocytopenia syndrome(SFTS) is an emerging infectious disease caused by SFTS virus(SFTSV). SFTSV is associated with a high mortality rate and has been reported in china, South Korea and Japan. SFTSV undergoes rapid changes owing to evolution, gene mutations, and reassortment between different strains of SFTSV. In this review, we summarize the recent cases and general properties of SFTS, focusing on the epidemiology, genetic diversity,clinical features, and diagnostics of SFTSV in china. From 2010 to October 2016, SFTS cases were reported in 23 provinces of china, with increased numbers yearly. Infection and death cases are mainly found in central china, where the Haemaphysalis longicornis ticks are spread. The national average mortality rate of SFTS infection was 5.3%, with higher risk to elder people. The main epidemic period was from May to July, with a peak in May. Thus, SFTS reminds a significant public health problem, and development of prophylactic vaccines and effective antiviral drugs will be highly needed.

Serological investigation of vector-borne disease in dogs from rural areas of china

Identification of Orientia tsutsugamushi,spotted fever group and typhus group Rickettsia by duplex and nested PCR methods

Objective:To identify members of genera of rickettsia and *** ***:Rapid and duplex and nested PCR methods have been established by designing primers based on the conserved regions of heat shock protein GroEL gene.345 mouse viscera samples including liver,spleen and kidney,96 Xenopsylla cheopis and 32 chiggers collected from Hongta areas of Yuxi city,Yunnan province were tested by the new PCR ***:The result of the study showed that the new PCR methods could identify most members of genera -Rickettsia and Orientia simultaneously with 100%specificity and its sensitivity could test one copy per *** results of detection prevalence of rickettsioses in mouse,flea and mites DNA samples showed that the total rickettsia infection rate in mouse was 34.78%(120/345).The total infection rates in ***,O.t Karp and *** of mouse samples were 28.12%(97/345),19.71%(68/345) and O. 29%(1/345) ***-infection rates in *** and 0.t Karp of mouse samples were 13.33%(46/ 345).O.t Karp type has been the main epidemic strain in these ***:We concluded that this PCR method could be used to detect multi-genera rickettsia *** evidences provided in this and previous studies strongly support that Hongta areas of Yuxi city are a natural focus for typhus and scrub typhus with the common occurrence of their confection.

External quality assessment of the detection of rickettsioses in china

Objective:To report a training course on the laboratory diagnoses of rickettsioses that 10 provincial/city CDCs participated in laboratory external quality assurance(EQA) based on the serological specific antibodies detection and rapid PCR amplifying targeted genes of rickettsiae. Methods:An EQA program to evaluate the following laboratory procedures was developed to detect rickettsiae:(1) immunofluorescent assay(IFA) to detect specific antibodies of A. phagocytophilum,*** and *** respectively.(2) Two sets of nested PCR were used amplifying groEL genes for most members of the family Rickettsiaceae and amplifying 16SrRNA genes for the most members of family anaplasmae,respectively.A scoring scheme based on the distribution of the median antibody titer values of the serologic assays was designed and a ranking list of the scores of the PCR results based on the detected minimal copy numbers of reference DNA was ***:Among nine laboratories who reported the results on time,eight laboratories gave acceptable serologic results,the other one provided an unacceptable antibody titer(1:2 vs median 1:64) results for *** limits of detection(LOD) for the PCR amplifying for five references DNA ranged from 1copy/μL to 10~6 copy/μ***:We successfully trained and popularized modern diagnostic methods of rickettsiae in 10 provincial CDCs in china and first conducted the EQA projects and evaluated the results.

regulatory effect of miRNA 320a on expression of aquaporin 4 in brain tissue of epileptic rats

Objective:To study the expression of miRNA 320 a in the brain tissue of epileptic rats and analyze its effect on the expression of aquaporin 4(AQP4).Methods:All rats were performed with the intraperitoneal injection of lithium chloride(3 mmol/kg) and then the intraperitoneal injection of pilocarpine(30 mg/kg) 24 h later(injected twice) to prepare the epileptic model of Wistar *** in the control group were injected with the equal volume of normal *** to the Racine scale,rats with over stage 3 of epilepsy were chosen and the brain tissue was separated quickly and then stored at-80 ℃.The immunohistochemistry was used to detect the expression of aquaporin in the brain tissue of epileptic model and the real-time PCR was employed to determine the difference in the expression of miRNA 320 a and AQP4 in the brain tissue of rats between the epileptic model group and control *** 5-day neonatal Wistar rats were chosen to collect the cerebral cortex and their primary astrocytes were separated and *** were transfected with miRNA mimic and imitated to the endogenous miRNA 320 a to up-regulate the expression of miRNA 320 ***:In the model group,the expression of AQP4 was significantly higher than the control group(Pression of miRNA 320 a in the model group was lower than control group(Prelated to *** the primary astrocytes,the transfection of miRNA 320 a mimic could significantly reduce the expression of AQP4,while its inhibitor could up-regulate the expression of AQP4,which indicated that miRNA 320 a could reduce the expression of ***:In the primary astrocytes of rats,the miRNA 320 a could inhibit the expression of AQP4 and after adding the inhibitor of miRNA 320 a,the expression of AQP4 was up-regulated.

目的分析中国首次分离的基因Ⅰ型流行性乙型脑炎病毒(JEV)全基因组的基因组特征。方法设计JEV全基因组扩增引物,RT-PCR扩增片断,PCR产物直接测序,拼接后获得全基因组序列。采用生物学软件进行同源性和系统进化分析。结果 1977和1982年分离自云南蚊虫的M28和BN82215病毒株基因组全长分别为10 969bp和10 970bp,5′非编码区均含有96个核苷酸,3′非编码区含有573和574个核苷酸。它们的开放阅读框(ORF)都从97到10 396位,共10 299个核苷酸,编码3 433个氨基酸。M28和BN82215株与来自GenBank的5个基因型JEV株的全基因组核苷酸和氨基酸同源性分别为78.4%~96.3%和91.4%~99.6%,与近几年国内外基因Ⅰ型流行株的同源性最高,进化关系最接近,都属于基因Ⅰ型。这两株病毒与JEV疫苗株SA14-14-2相比,有56个氨基酸差异,其中E基因有11个氨基酸差异,但都不属抗原关键位点。结论本研究阐明了中国首次分离的基因Ⅰ型JEV的全基因组分子特征,决定病毒抗原和毒力的E蛋白关键位点无明显变化。证实20世纪70和80年代云南省就有基因Ⅰ型JEV流行。
