Application of PCR and real-time PCR for monitoring cyanobacteria, Microcystis spp. and Cylindrospermopsis raciborskii in Macao freshwater reservoir

Freshwater algal blooms have become a growing concern world-wide. They are caused by a high level ofcyanobacteria, predominantly Microcystis spp. and Cylindrospermopsis raciborskii, which can produce microcystin and cylindrospermopsin, respectively. Longtime exposure to these cyanotoxins may affect public health, thus reliable detection, quantification, and enumeration of these harmful algae species has become a priority in water quality management. Traditional manual enumeration of algal bloom cells primarily involves microscopic identification which limited by inaccuracy and time-consumption. With the development of molecular techniques and an increasing number of microbial sequences available in the Genbank database, the use of molecular methods can be used for more rapid, reliable, and accurate detection and quantification. In this study, multiplex polymerase chain reaction （PCR） and real-time quantitative PCR （qPCR） techniques were developed and applied for monitoring cyanobacteria Microcystis spp. and C. raciborskii in the Macao Storage Reservoir （MSR）. The results showed that the techniques were successful for identifying and quantifying the species in pure cultures and mixed cultures, and proved to be a potential application for water sampling in MSR. When the target species were above 1 million cells/L, similar cell numbers estimated by microscopic enumeration and qPCR were obtained. Further quantification in water samples indicated that the ratio of the estimated number of cell by microscopy and qPCR was 0.4-12.9 for cyanobacteria and 0.2-3.9 for C. raciborskii. However, Microcystis spp. was not observed by manual enumeration, while it was detected at low levels by qPCR, suggesting that qPCR is more sensitive and accurate. Thus the molecular approaches provide an additional reliable monitoring option to traditional micro- scopic enumeration for the ecosystems monitoring program.

Inulin-type fructooligosaccharides（FOS） purified from Morinda officinalis,with degrees of polymerization（DP） from 3 to 9, has been approved as the prescribed oral drug by CFDA in 2012 with the indications for mild and moderate depressive episode, while the bioprocess of this FOS mixtures are largely unknown. This work aimed to illustrate this bioprocess related to gut microbiota and intestine construction. The depression-like rat model was built by chronic unpredictable mild stress（CUMS） procedure for seven weeks, then verified by open field test（OFT） and sucrose preference test（SPT）. Under the three weeks’ treatment period, the fecal sample were collected at day 0, 15 and 22, and further analyzed the microbiota DNA composition using ERIC-PCR method. Hematoxylin and eosin（H＆E） staining was applied to investigate the gut epithelial morphology. Results showed that, rats with depression-like behaviors presented low SPT value（＜50%, p＜0.01） and low total travel distance in OFT test（p＜0.01）. Conversely, SPT value was increased（＞65%, p＜0.05） after treatments with FOS, indicating recovering the pleasant sensation. Meanwhile, in partial least square data analysis（PLS-DA） plot, differential modulations of normal and depressive rat model were observed at 0-day microbiota composition. Furthermore, while compared the 22-day plot, there were greatly changed the microbiota composition after treatments（fluoxetine, FOS-DP5 and FOS mixture）. In other side, the colon mucosa, intestinal villus（height and integrity） were severe damaged in intestine of depressive rat model. It was the intestinal inflammatory and severe damage happened in depression-like rats. However, intestinal micro-architecture abnormalities were restored and reversed by FOS treatment. Inferior to FOS, fluoxetine aroused intestinal irritation（cell necrosis, exfoliation and deep staining） on parts of treated rats. CAR designs for solid tumors: overcoming hurdles and paving the way for effective immunotherapy

Chimeric antigen receptor T cell(CAR-T) therapy has revolutionized immunotherapy by modifying patients’ immune cells genetically. By expressing CARs, these modified cells can specifically identify and eliminate tumor cells. The success of CAR-T therapy in hematological malignancies, such as leukemia and lymphoma, has been remarkable. Numerous studies have reported improved patient outcomes and increased survival rates. However, the application of CAR-T therapy in treating solid tumors faces significant challenges. Solid tumors possess complex microenvironments containing stromal cells, extracellular matrix components, and blood vessels. These factors can impede the infiltration and persistence of CAR-T cells within the tumor. Additionally, the lack of target antigens exclusively expressed on tumor cells raises concerns about off-target effects and potential toxicity. This review aims to discuss advancements achieved by CAR-T therapy in solid tumors and the clinical outcomes in the realm of solid tumors.

20（S）-protopanaxatriol saponins（PTS）and 20（S）-protopanaxadiol saponins（PDS）are well known as main bioactive components of Panax genus,including ***,*** and *** composition and proportion of PTS and PDS varied greatly according to *** the facts that PTS and PDS exhibit different and even antagonistic pharmacological effects,it is meaningful to develop an approach to rapidly quantify PTS and PDS in Panax *** the present study,a novel strategy for qualitative and quantitative determination of PTS and PDS in ***,*** and ***,was proposed,based on the overlapping peaks of main components of PTS（calibrated by ginsenoside Rg1） and PDS（calibrated by ginsenoside Rb1）.The analysis was performed by high performance liquid chromatography coupled with evaporative light scattering detection（HPLC-ELSD）.Under the specific chromatographic conditions,all samples showed two overlapping peaks containing several main ginsenosides belongs to PTS and PDS,*** overlapping peaks were also identified by *** on sum and ratio of PTS and PDS,60 tested Panax samples were divided into three main clusters according to *** findings suggested that this strategy provides a simple and rapid approach to quantify PTS and PDS in Panax herbs.

A dual mediated liver tumor targeting polymer constituted with glycyrrhetinic acid(GA)-modified poly(ethylene glycol)-poly-L-histidine-poly(lactic-co-glycolic acid)(GA-PEG-PHIS-PLGA)was synthesized,in which GA contributed as liver target ligand and PHIS devoted as the trigger of tumor slight acid ***(ADG),a diterpenoid lactone isolated from Andrographis paniculata,which exhibited anti-liver cancer activities while extremely hydrophobic character,was chosen as the model *** features ofliver targeted nanoparticles(NPs)via self-assembly of amphipathic copolymers including its morphology,diameter,size distribution and in vitro drug release in different pH value buffer were *** relative changes following pH 7.4 to 5.0 buffer would mean this polymer could be suitable as the drug delivery carriers for the slight acid micro-environment of solid ***,the results of in vitro cell cytotoxicity and cellular uptake studies showed the introduction of GA to the micelles could significantly increase the affinity for human hepatic carcinoma Hep3B cells,which induced a 2.6-fold higher endocytosis than it for human breast carcinoma MAD-MB-231 *** results indicated that this copolymer would be greatly promising in the chemotherapy of liver cancer due to its GA ligand and acid micro-environment response.

Objective:To investigate the pro-angiogenic effects of paeoniflorin(PF)in a vascular insufficiency model of zebrafish and in human umbilical vein endothelial cells(HUVECs).Methods:In vivo,the pro-angiogenic effects of PF were tested in a vascular insufficiency model in the Tg(fli-1:EGFP)y1 transgenic *** 24 h post fertilization(hpf) embryos were pretreated with vascular endothelial growth factor(VEGF) receptor tyrosine kinase inhibitor II(VRI) for 3 h to establish the vascular insufficiency model and then post-treated with PF for 24 *** formation of intersegmental vessels(ISVs) was observed with a fluorescence *** mRNA expression of fins-like tyrosine kinase-1(flt-1),kinase insert domain receptor(kdr),kinase insert domain receptor like(fdrl) and von Willebrand factor(vWF) were analyzed by real-time polymerase chain reaction(PCR).In vitro,the pro-angiogenic effects of PF were observed in HUVECs in which cell proliferation,migration and tube formation were ***:PF(6.25- 100μmol/L) could rescue VRI-induced blood vessel loss in zebrafish and PF(25 to 100μmol/L),thereby restoring the mRNA expressions of flt-1,Mr,kdr1 and vWF,which were down-regulated by VRI *** addition,PF(0.001- 0.03 μmol/L) could promote the proliferation of HUVECs while PF stimulated HUVECs migration at 1.0-10 umol/L and tube formation at 0.3 umol/***:PF could promote angiogenesis in zebrafish in vivo and in HUVECs in vitro.
