Circadian disturbance induces erectile dysfunction by impairing endothelial function

In order to explore the impact of circadian disturbance on erectile function,we randomly divided 24 adult male rats into groups of control(light on at 8:00 a.***.and off at 8:00 p.m.),dark/dark(DD;constant dark),light/light(LL;constant light),and shift dark/light(DL;light off at 8:00 a.***.and on at 8:0o p.m.).Four weeks later,erectile function was measured and corpora cavernosa were harvested for analysis.The maximum intracavernous pressure(mlcP)and mCP/mean arterial pressure(MAP)ratio in the DD,LL,and DL groups were significantly lower than that in the control group.The LL and DL groups showed significantly attenuated endothelial nitric oxide synthase(eNOS),while DD,LL,and DL showed reduced neuronal nitric oxide synthase(nNOS)at both mRNA and protein levels.The production of nitric oxide(NO)and cyclic guanosinemonophosphate(cGMP)was inhibited by altered light/dark cycles to varying degrees.Circadian disturbance impaired endothelial function and contributed to erectile dysfunction.For the core circadian elements,mRNA expression of circadian locomotor output cycles kaput(Clock)and brain/muscle aryl-hydrocarbon receptor nuclear translocator-like protein 1(Bmal1)was elevated in the DL group,but their protein expression was not significantly changed.DD,LL,and DL increased period 1(Per1)and Per3 levels,while LL and DL increased PER1 levels.No significant difference was found for Per2levels,and PER2 and PER3 concentrations were not significantly changed.Moreover,LL and DL significantly increased cryptochrome-1(CRY1)and CRY2 at both mRNA and protein levels.The altered light/dark rat model showed that circadian disturbance contributed to erectile dysfunction probably by impairing endothelial function.Meanwhile,the core circadian elements were detected in the corpora cavernosa,but these were disrupted.However,which circadian element regulates erectile function and how it works need further analysis.

Distribution and bioavailability of mercury in size-fractioned atmospheric particles around an ultra-low emission power plant in Southwest China

Ultra-low emission(ULE)technology retrofits significantly impact the particulate-bound mercury(Hg)emissions from coal-fired power plants(CFPPs);however,the distribution and bioavailability of Hg in size-fractioned particulate matter(PM)around the ULE-retrofitted CF-PPs are less understood.Here,total Hg and its chemical speciation in TSP(total suspended particles),PM_(10)(aerodynamic particle diameter≤10μm)and PM_(2.5)(aerodynamic particle diameter≤2.5μm)around a ULE-retrofitted CFPP in guizhou province were quantified.Atmospheric PM_(2.5)concentration was higher around this ULE-retrofitted CFPP than that in the intra-regional urban cities,and it had higher mass Hg concentration than other sizefractioned PM.Total Hg concentrations in PM had multifarious sources including CFPP,vehicle exhaust and biomass combustion,while they were significantly higher in autumn and winter than those in other seasons(P40%).Mass concentrations of exchangeable,HCl-soluble,elemental,and residual Hg in PM_(2.5)were higher than those in other size-fractioned PM,and were markedly elevated in autumn and winter(P<0.05).In PM_(2.5),HCl-soluble Hg presented a significantly positive relationship with elemental Hg(P<0.05),while residual Hg showed the significantly positive relationships with HCl-soluble Hg and elemental Hg(P<0.01).Overall,these results suggested that atmospheric PM-bound Hg around the ULE-retrofitted CFPP tends to accumulate in finer PM,and has higher bioavailable fractions,while has potential transformation between chemical speciation.

Three rare anti-inflammatory sesquiterpene lactones from Magnolia grandiflora

Four new sesquiterpene lactones(SLs)(1–4),along with a biosynthetically related SL(5),have been isolated from the leaves of Magnolia grandiflora.Magrandate A(1)is notable as the first C18 homogemarane type SL,featuring a unique 1,7-dioxaspiro[4.4]nonan-6-one core.Compounds 2 and 3,representing the first instances of chlorine-substituted gemarane-type SL analogs in natural products,were also identified.The structures of these isolates were elucidated through a combination of spectroscopic data analysis,electronic circular dichroism calculations,and X-ray single-crystal diffraction analysis.All isolates demonstrated anti-inflammatory activity in lipopolysaccharide-stimulated RAW264.7 cells.Notably,3–5 showed a significant inhibitory effect on nitric oxide production,with IC50 values ranging from 0.79 to 4.73μmol·L^(−1).Additionally,4 and 5 exhibited moderate cytotoxic activities against three cancer cell lines,with IC50 values between 3.09 and 11.23μmol·L^(−1).

Metallogenesis of the Baidi Au-Sb deposit, southwest guizhou province, China: mineralogical and geochemical evidence from sulfur-bearing minerals

The Baidi Au-Sb deposit, which contains 8 t of Au and 10,979 Mt of Sb, is a typical and rare paragenetic deposit located in southwestern guizhou province, China.Previous studies have focused on individual ores, but have not combined them to identify their paragenetic mechanism or metallogenic regularity. Therefore, we used field investigations, microscopic observations, and in situ analyses to identify the spatial distribution, mineral paragenesis, compositional evolution, and metallogenic material sources of the ore bodies. We also determined the Au and Sb paragenetic characteristics and the metallogenesis of the deposit. The main Au-bearing minerals in the deposit were early(Apy1–2) and late(Apy3) stage arsenopyrites, as well as pre-mineralization(Py1), mineralization(Py2–5), and late mineralization(Py6–7) stage pyrites. The main Sb-bearing minerals were stibnite(Snt), skinnerite, bournonite,and valentinite. The minerals formed in the order of Py1,Py2–3 + Apy1, Py4–5 + Apy2, Snt, and Py6–7 + Apy3.The δ34S values of the arsenopyrites and pyrites ranged from-5 to 5‰, while those of stibnite were mostly less than-5‰ in the later mineralization stages. Sulfur was provided by deep magmatic hydrothermal fluids, but sedimentary sulfur was added in the later stages. Moreover,the trace elemental contents fluctuated and eventually became similar to those of the sedimentary strata. By comprehensively considering the ores along with the geological characteristics of the deposit, we determined that deep magma provided the Au during ore formation. Later tectonic changes provided Sb from the sedimentary strata,which precipitated along fault expansion areas and produced Au and Sb paragenesis.

MicroR-9c-5p and novel-mir50 co-target Akt to regulate Lasioderma serricorne reproduction

High fecundity is a common characteristic of insect pests which increases the difficulty of population control.Serine/threonine kinase Akt is an indispensable component of the insulin signaling pathway.Silencing of LsAkt severely hinders reproduction in Lasioderma serricorne,a stored product insect pest.However,the post-transcriptional pathway of LsAkt in L.serricorne remains unknown.This study identified 2 binding sites of miR-9c-5p and novel-mir50 in the coding sequences of LsAkt.The expression profiles of 2 microRNAs(miRNAs)and LsAkt displayed an opposite pattern during the adult stages.Luciferase reporter assay showed that novel-mir50 and miR-9c-5p could downregulate the expression of LsAkt.Overexpression of miR-9c-5p and novel-mir50 by injection of mimics inhibited the expression of LsAkt and reduced oviposition,decreased egg hatchability,and blocked ovarian development.It also decreased the expression of genes involved in ovarian development(LsVg and LsVgR)and the nutritional signaling pathway(LsTOR,LsS6K,and Ls4EBP),and reduced the phosphorylation of Akt.Conversely,injection of miR-9c-5p and novel-mir50 inhibitors induced the expressions of LsAkt,LsVg,LsVgR,LsTOR,LsS6K,and Ls4EBP,enhanced Akt phosphorylation level,and accelerated ovarian development.Injection of bovine insulin downregulated the expression of miR-9c-5p and novel-mir50 and upregulated the LsAkt expression.It also rescued the reproductive development defects associated with miR-9c-5p/novel-mir50 overexpression,forming a positive regulatory loop of insulin signaling.These results indicate that miR-9c-5p/novel-mir50 regulates the female reproduction of L.serricorne by targeting Akt in response to insulin signaling.The data also demonstrate the effects of the insulin/miRNA/Akt regulatory axis in insect reproduction.

Evolutionary game dynamics of combining two different aspiration-driven update rules in structured populations

In evolutionary games,most studies on finite populations have focused on a single updating mechanism.However,given the differences in individual cognition,individuals may change their strategies according to different updating mechanisms.For this reason,we consider two different aspiration-driven updating mechanisms in structured populations:satisfied-stay unsatisfied shift(SSUS)and satisfied-cooperate unsatisfied defect(SCUD).To simulate the game player’s learning process,this paper improves the particle swarm optimization algorithm,which will be used to simulate the game player’s strategy selection,i.e.,population particle swarm optimization(PPSO)algorithms.We find that in the prisoner’s dilemma,the conditions that SSUS facilitates the evolution of cooperation do not enable cooperation to emerge.In contrast,SCUD conditions that promote the evolution of cooperation enable cooperation to emerge.In addition,the invasion of SCUD individuals helps promote cooperation among SSUS individuals.Simulated by the PPSO algorithm,the theoretical approximation results are found to be consistent with the trend of change in the simulation results.

A robust tensor watermarking algorithm for diffusion-tensor images

Watermarking algorithms that use convolution neural networks have exhibited good robustness in studies of deep learning networks.However,after embedding watermark signals by convolution,the feature fusion eficiency of convolution is relatively low;this can easily lead to distortion in the embedded image.When distortion occurs in medical images,especially in diffusion tensor images(DTIs),the clinical value of the DTI is lost.To address this issue,a robust watermarking algorithm for DTIs implemented by fusing convolution with a Transformer is proposed to ensure the robustness of the watermark and the consistency of sampling distance,which enhances the quality of the reconstructed image of the watermarked DTIs after embedding the watermark signals.In the watermark-embedding network,Ti-weighted(Tlw)images are used as prior knowledge.The correlation between T1w images and the original DTI is proposed to calculate the most significant features from the T1w images by using the Transformer mechanism.The maximum of the correlation is used as the most significant feature weight to improve the quality of the reconstructed DTI.In the watermark extraction network,the most significant watermark features from the watermarked DTI are adequately learned by the Transformer to robustly extract the watermark signals from the watermark features.Experimental results show that the average peak signal-to-noise ratio of the watermarked DTI reaches 50.47 dB,the diffusion characteristics such as mean diffusivity and fractional anisotropy remain unchanged,and the main axis deflection angleαAc is close to 1.Our proposed algorithm can effectively protect the copyright of the DTI and barely affects the clinical diagnosis.

Mannitol inhibits the proliferation of neural stem cell by a p38 mitogen-activated protein kinase-dependent signaling pathway

Purpose:Mannitol is one of the first-line drugs for reducing cerebral edema through increasing the extracellular osmotic pressure.However,long-term administration of mannitol in the treatment of cerebral edema triggers damage to neurons and astrocytes.Given that neural stem cell(NSC)is a subpopulation of main regenerative cells in the central nervous system after injury,the effect of mannitol on NSC is still elusive.The present study aims to elucidate the role of mannitol in NSC proliferation.Methods:C57 mice were derived from the animal house of Zunyi Medical University.A total of 15 pregnant mice were employed for the purpose of isolating NSCs in this investigation.Initially,mouse primary NSCs were isolated from the embryonic cortex of mice and subsequently identified through immunofluorescence staining.In order to investigate the impact of mannitol on NSC proliferation,both cell counting kit-8 assays and neurospheres formation assays were conducted.Thein vitro effects of mannitol were examined at various doses and time points.In order to elucidate the role of Aquaporin 4(AQP4)in the suppressive effect of mannitol on NSC proliferation,various assays including reverse transcription polymerase chain reaction,western blotting,and immunocytochemistry were conducted on control and mannitol-treated groups.Additionally,the phosphorylated p38(p-p38)was examined to explore the potential mechanism underlying the inhibitory effect of mannitol on NSC proliferation.Finally,to further confirm the involvement of the p38 mitogen-activated protein kinase-dependent(MAPK)signaling pathway in the observed inhibition of NSC proliferation by mannitol,SB203580 was employed.All data were analyzed using SPSS 20.0 software(SPSS,Inc.,Chicago,IL).The statistical analysis among multiple comparisons was performed using one-way analysis of variance(ANOVA),followed by Turkey’’s post hoc test in case of the data following a normal distribution using a Shapiro-Wilk normality test.Comparisons between 2 groups were determined using Student’s t-te

Hepatocellular carcinoma(HCC),a common malignancy worldwide,still lacks effective clinical treatment.The study aimed to investigate the oncogenes that affect the progression of HCC and their possible mechanisms.In our study,we initially confirmed a higher level of PRDX2 in the bile of HCC patients compared to those with choledocholithiasis by 2-DE,LC-MS,and ELISA.Subsequently,we demonstrated the high expression of peroxiredoxin 2(PRDX2)in HCC based on the TCGA database and clinical sample analysis.Furthermore,PRDX2 overexpression enhanced the viability of HCC cells.And PRDX2 silencing induced senescence of HCC cells.In vivo,knockdown of PRDX2 significantly reduced the weight of xenograft tumors.PRDX2 also was found to activate the Wnt/β-catenin pathway by inducingβ-catenin nuclear translocation.Consequently,we proved that silencing PRDX2 could inhibit proliferation and Wnt/β-catenin pathway while promoting senescence in HCC cells.

Damage Characteristics Analysis and Fractal Study of Shale With Prefabricated Fractures under Thermal-mechanical Coupling

To study the damage and failure of shale with different fracture inclination angles under uniaxial compression loading,in this work,RFPA2D-Thermal,a two-dimensional real failure process analysis software,was used for numerical simulation.Numerical simulation results show that quartz in shale mainly affects the tensile and compressive strength of shale by increasing rock brittleness.The coupling of temperature and pressure will cause lateral and volume destruction of shale,which enables the shale body to be more easily broken.Fracture inclination is the key factor affecting shale damage patterns.The failure mode of shale with low-and high-angle fractures is mainly shear failure,and the compressive strength does not vary with crack inclination.The damage mode of obliquely intersecting fractured shale is slip damage along the fracture face,the compressive strength decreases and then increases with the fracture inclination,and a minimum value exists.The acoustic emission simulation results of the damage process effectively reflect the accumulated internal damage and macroscopic crack appearance until fracture instability when the prefabricated fractured shale is subjected to uniaxial compressive loading.The crack inclinations of 0°and 120℃ corresponds to the most complex"N"shape damage mode.The crack inclinations of 30°and 60°,and the damage mode is an inverted"λ"shape.
