Icariin plus curcumol enhances autophagy through the mTOR pathway and promotes cathepsin B-mediated pyroptosis of prostate cancer cells

Objective:To examine the effect of icariin plus curcumol on prostate cancer cells PC3 and elucidate the underlying ***:We employed the Cell Counting Kit 8 assay and colony formation assay to assess cell viability and *** expression was analyzed using monodansylcadaverine *** and Western blot analyses were used to evaluate protein expressions related to autophagy,pyroptosis,and the mTOR *** damage was examined using the lactate dehydrogenase ***,cathepsin B and NLRP3 were detected by ***:Icariin plus curcumol led to a decrease in PC3 cell proliferation and an enhancement of *** levels of LC3-Ⅱ/LC3-Ⅰand beclin-1 were increased,while the levels of p62 and mTOR were decreased after treatment with icariin plus *** changes were reversed upon overexpression of ***,3-methyladenine resulted in a decrease in inflammatory cytokines,pyroptosis-related protein levels,and lactate dehydrogenase concentration,compared to the icariin plus curcumol *** cathepsin B reversed the regulatory effects of icariin plus ***:Icariin plus curcumol demonstrates great potential as a therapeutic agent for castration-resistant prostate cancer by enhancing autophagy via the mTOR pathway and promoting pyroptosis mediated by cathepsin *** findings provide valuable insights into the molecular mechanisms underlying the therapeutic potential of icariin and curcumol for prostate cancer treatment.

Background:Acute gouty arthritis(AGA)is an inflammatory joint disease with a high *** medical interventions,including nonsteroidal anti-inflammatory drugs,colchicine and glucocorticoids,can have serious adverse *** Granule(HZG),a compound chinese herbal medicine,has been used to treat AGA for more than 30 years with satisfactory effects and no significant adverse ***,the efficacy and safety of HZG in AGA patients remains ***:The present investigation was designed to examine the efficacy and safety profile of HZG in managing AGA ***,setting,participants and interventions:The current study was conducted as a noninferiority,randomized controlled clinical trial on 180 eligible enrolled *** were randomly assigned into the HZG and etoricoxib *** were administered for 5 d,during which the HZG group received HZG and placebo etoricoxib,while the etoricoxib group received etoricoxib and placebo HZG in the same ratio(1:1).Main outcome measures:The primary outcome was pain experienced by the patient in the gout-afflicted joint from days 2 to 5 of the treatment *** pain level was measured via a visual analogue scale,ranging from 0 mm to 100 *** secondary outcomes comprised joint tenderness and swelling,reduction of inflammatory biomarkers,and the patient’s and investigator’s global evaluations of therapeutic ***:The mean reduction in pain was-51.22 mm(95%confidence interval[CI],[-53.42,-49.03]mm)for the HZG and-52.00 mm(95%CI,[-54.06,-49.94]mm)for the etoricoxib *** mean difference between the two groups was 0.78 mm(95%CI,[-2.25,3.81]mm).All additional efficacy endpoints,covering decreased inflammation and pain relief,yielded compelling proof of *** in the HZG group exhibited a comparatively lower rate of adverse events compared to those in the etoricoxib group(4.44%vs 13.33%;P≤0.05).Conclusion:HZG and etoricoxib groups demons

Emodin Ameliorates High Glucose-Induced Podocyte Apoptosis via Regulating AMPK/mTOR-Mediated Autophagy Signaling Pathway

Objective To investigate the effect of emodin on high glucose(HG)-induced podocyte apoptosis and whether the potential anti-apoptotic mechanism of emodin is related to induction of adenosine-monophosphate-activated protein kinase(AMPK)/mammalian target of rapamycin(mTOR)-mediated autophagy in podocytes(MPC5 cells)in *** MPC5 cells were treated with different concentrations of HG(2.5,5,10,20,40,80 and 160 mmol/L),emodin(2,4,8µmol/L),or HG(40 mmol/L)and emodin(4µmol/L)with or without rapamycin(Rap,100 nmol/L)and compound C(10µmol/L).The viability and apoptosis of MPC5 cells were detected using cell counting kit-8(CCK-8)assay and flow cytometry analysis,*** expression levels of cleaved caspase-3,autophagy marker light chain 3(LC3)Ⅰ/Ⅱ,and AMPK/mTOR signaling pathway-related proteins were determined by Western *** changes of morphology and RFP-LC3 fluorescence were observed under *** HG at 20,40,80 and 160 mmol/L dose-dependently induced cell apoptosis in MPC5 cells,whereas emodin(4µmol/L)significantly ameliorated HG-induced cell apoptosis and caspase-3 cleavage(P<0.01).Emodin(4µmol/L)significantly increased LC3-Ⅱ protein expression levels and induced RFP-LC3-containing punctate structures in MPC5 cells(P<0.01).Furthermore,the protective effects of emodin were mimicked by rapamycin(100 nmol/L).Moreover,emodin increased the phosphorylation of AMPK and suppressed the phosphorylation of *** AMPK inhibitor compound C(10µmol/L)reversed emodin-induced autophagy *** Emodin ameliorated HG-induced apoptosis of MPC5 cells in vitro that involved induction of autophagy through the AMPK/mTOR signaling pathway,which might provide a potential therapeutic option for diabetic nephropathy.

Discovery of Kalmane Diterpenoids as Potent Analgesics from the Flowers of Rhododendron dauricum

Kalmane diterpenoids,featuring a 5/8/5/5 tetracyclic carbon skeleton,are very rare in *** flowers of Rhododendron dauricum L.(Ericaceae)were phytochemically investigated for the first time,leading to the isolation of eight kalmane diterpenoids(1-8)including four new ones,named rhodokalmanols A-D(1-4).The structures of 1-8 were elucidated by comprehensive spectroscopic methods and ic NMR-DP4+analysis,and the absolute configurations of 1,2,4,and 5 were defined by single-crystal X-ray diffraction analysis with Cu Kα*** A(1)represents the first 5,8-epoxykalmane diterpenoid and also the first kalm-15(16)-ene *** BD(2-4)are the first examples of kalm-7(8)-ene,kalm-16(17)-ene,and 8α-methoxykalmane diterpenoids,*** the isolated kalmane diterpenoids 1-8 exhibited significant analgesic effects,and rhodokalmanol C(3)and kalmanoi(8)expressed more potent analgesic activity than morphine at doses of 0.2 and 0.04 mg/*** preliminary structure-activity relationships of kalmane diterpenoids as potent analgesics are discussed.

Effects of Jiawei Buzhong Yiqi Decoction on the Endometrial Receptivity of Rats with Polycystic Ovary Syndrome

[Objectives]The study aims to discuss the effects of Jiawei Buzhong Yiqi Decoction on the endometrial receptivity of rats with polycystic ovarian syndrome(PCOS).[Methods]Adult female SD rats were selected and randomly divided into the model group and control *** the model group,the PCOS model was established by using the method of Poretsky *** in the control group were given physiological saline intervention at the same *** successfully modeled rats were randomly divided into the PCOS group and traditional chinese medicine *** them,rats in the chinese medicine group were given Jiawei Buzhong Yiqi Decoction by gavage;rats in the model group and the control group were given normal saline for 14 *** rats were killed by anesthesia after the last administration,and serum and uterine specimens were *** sex hormones(including T,E_(2),and P),glucose and insulin levels were detected;the equivalent diameter and area of endometrial gland and gland cavity,and the endometrial thickness of rats were measured;the expression of endometrial leukemia inhibitory factor(LIF),integrinαvβ3 protein and mRNA was detected by using Elisa and RT-PCR methods.[Results]The serum T,glucose,and insulin levels of rats in the PCOS group were significantly higher than those of the control group and the traditional chinese medicine group,and there was no significant difference in the above indicators between the control group and the traditional chinese medicine group;there was no significant difference in the E_(2) and P levels between the three *** equivalent diameter and area of endometrial gland and gland cavity,and the endometrial thickness of rats in the PCOS group were lower than those of the control group and the traditional chinese medicine group;there was no significant difference in the above indicators between the traditional chinese medicine group and the control *** expression of LIF,integrinαvβ3 protein and mRNA in the endometrium of rats

Rhodauricanol A, an analgesic diterpenoid with an unprecedented5/6/5/7 tetracyclic system featuring a unique16-oxa-tetracyclo[11.2.1.0^(1,5).0^(7,13)]hexadecane core from Rhododendron dauricum

A novel diterpenoid with an unprecedented 5/6/5/7 tetracyclic system, rhodauricanol A(1), five new grayanane-derived diterpenoids, dauricanols A-E(2-6), and five known ones(7-11) were isolated from the flowers of Rhododendron dauricum. Rhodauricanol A(1) possesses a unique 5/6/5/7 tetracyclic ring system featuring a 16-oxa-tetracyclo[11.***.1.0^(1,5).0^(7,13)]hexadecane core. Dauricanols A-C(2-4) are the first1,3-dioxolane conjugates of grayanane diterpenoids and 5-hydroxymethylfurfural and vanillin, respectively, and dauricanols D(5) and E(6) represent the first examples of 6-deoxy-1,5-seco-grayanane diterpenoids. Their structures were determined by spectroscopic methods, quantum chemical calculation including ^(13)C NMR-DP4+ analysis and ECD calculation, and single-crystal X-ray diffraction analysis. Plausible biosynthetic pathways for 1-4 were proposed. All the isolates showed significant analgesic activities,and dauricanols B(3) and C(4) showed more potent analgesic activities than the positive control, morphine.

Natural locomotion such as walking,crawling,and swimming relies on spatially controlled deformation of soft tissues,which could allow efficient interaction with the external *** one of the ideal candidates for biomimetic materials,hydrogels can exhibit versatile bionic ***,it remains an enormous challenge to transfer these insitu deformations to locomotion,particularly above complex ***,inspired by the crawling mode of inchworms,an isotropic hydrogel with thermoresponsiveness could evolve to an anisotropic hydrogel actuator via interfacial diffusion polymerization,further evolving to multisection structure and exhibiting adaptive deformation with diverse degrees of ***,a dynamic mortise-and-tenon interlock could be generated through the interaction between the self-deformation of the hydrogel actuator and rough terrains,inducing continual multidimensional locomotion on various artificial rough substrates and natural sandy ***,benefiting from the powerful mechanical energy transfer capability,the crawlable hydrogel actuators could also be utilized as hydrogel motors to activate static cargos to overstep complex terrains,which exhibit the potential application of a biomimetic mechanical discoloration ***,we believe that this design principle and control strategy may be of potential interest to the field of deformable materials,soft robots,and biomimetic devices.
