Dietary Methionine Affect Meat Qulity and Myostatin Gene Exon 1 Region Methylation in Skeletal Muscle Tissues of Broilers

Dietary amino acids imbalance will result in stunted broiler performance and deteriorated meat quality, which are involved in various biochemical cycles in vivo. In this study, the effects of dietary methionine on meat quality and methylation of myostatin exon 1 were investigated. Drip loss of the broilers fed with diet of high methionine levels （0.2%） increased from （6.3 ± 0.1）% （control group） to （10.1 ± 1.0）%, and the muscle shearing force increased from （22.8 ± 1.9） N （control group） to （26.3 ±2.3） N. Moreover, many CpG sites were found at the myostatin exon 1 region （nucleotides 2 360-2 540 bp）. To further understand the regulation of broiler myostatin expression, the methylation status of broiler myostatin exon 1 and its mRNA expression were analyzed. At the myostatin exon 1 region where CG enriches （nucleotides 2 360-2 540 bp）, the percentages of methylation were 46 and 84% in low Met and high Met content groups after 55-d feeding, respectively. In skeletal muscle tissues, the exon 1 hypermethylation status of myostatin gene was found to be negatively correlated with the gene expression. These results suggested that methylation of this gene is a dynamic process, which plays a dominant role in regulating gene expression for development of individuals.

Cloning of a down-regulated gene encoding small GTP binding protein in hybrid wheat

Previous studies showed that differential gene expression between wheat hybrids and their parents was responsible for the heterosis. To provide an insight into the molecular basis of wheat heterosis, one cDNA, designated TaRab, was identified from the cDNA library of wheat seedling leaves. The sequence comparison in GenBank revealed that TaRab is homologous to a group of genes encoding Rab-GTP binding protein. Semi-quantitative RT-PCR analysis indicated that TaRab was expressed in all plant tissues examined, but at slightly higher level in leaves. Further analysis exhibited that TaRab displayed lower expression in hybrid than in its patents in both roots and leaves, which was in agreement with the original results of suppression subtractive hybridization. TaRab was located on chromosome 7B and C-7DS5-0.36 by in silico mapping. The relationship between differential expression of TaRab and the molecular basis of wheat heterosis was also discussed.

Effect of hormonal status and metabolic changes of restricted ewes during late pregnancy on their fetal growth and development

This study investigated the effects of hormonal status; metabolic changes of restricted ewes during late pregnancy on the ovine fetus growth; development. One hundred Mongolian ewes, synchronized for oestrus; mated, were divided into three groups; offered 0.175 MJME·kgw[0.75]·d[1] (Restricted group 1, RG1), 0.33 MJME·kgw[0.75]·d[1] (Restricted group 2, RG2); ad libitum access to feed (Control group, CG) during their late pregnancy respectively. The results suggested that with the supply of exogenous energy decreasing during late pregnancy, maternal body weight; net body weight loss in RG2; RG1 were lower than those of CG (Ps0.01). The insulin; IGF-1 concentrations of ewes in RG2; RG1 tended to be lower than those of CG (P>0.05), but the GH concentrations in RG2; RG1 were enhanced; there was significant difference between RG1; CG on d 120 of gestation (P0.05). However, the NEFA; FAA concentrations of ewes in RG1 were reduced from d 90 to d 120 of gestation, then enhanced from d 120 to d 140 of gestation. During the late pregnancy, with the supply of nutrition decreasing, the negatively physiological; biochemical maternal reactions to restriction became worse, which significantly reduced the average lamb birth weight; daily growth rate of fetus in RG2 (P<0.05); RG1 (P<0.01).

Strategies on Sample Size Determination and Qualitative and Quantitative Traits Integration to Construct Core Collection of Rice (Oryza sativa)

The development of a core collection could enhance the utilization of germplasm collections in crop improvement programs and simplify their management. Selection of an appropriate sampling strategy is an important prerequisite to construct a core collection with appropriate size in order to adequately represent the genetic spectrum and maximally capture the genetic diversity in available crop collections. The present study was initiated to construct nested core collections to determine the appropriate sample size to represent the genetic diversity of rice landrace collection based on 15 quantitative traits and 34 qualitative traits of 2 262 rice accessions. The results showed that 50-225 nested core collections, whose sampling rate was 2.2%-9.9%, were sufficient to maintain the maximum genetic diversity of the initial collections. Of these, 150 accessions （6.6%） could capture the maximal genetic diversity of the initial collection. Three data types, i.e. qualitative traits （QT1）, quantitative traits （QT2） and integrated qualitative and quantitative traits （QTT）, were compared for their efficiency in constructing core collections based on the weighted pair-group average method combined with stepwise clustering and preferred sampling on adjusted Euclidean distances. Every combining scheme constructed eight rice core collections （225, 200, 175, 150, 125, 100, 75 and 50）. The results showed that the QTT data was the best in constructing a core collection as indicated by the genetic diversity of core collections. A core collection constructed only on the information of QT1 could not represent the initial collection effectively. QTT should be used together to construct a productive core collection.

Microwave-assisted Solid-phase Synthesis, Biological Evaluation and Molecular Docking of Angiotensin I-converting Enzyme Inhibitors

Short peptides based on the tripeptides, Leu-Arg-Pro and Leu-Lys-Pro, were synthesized by microwave assisted solid-phase synthesis method, in order to make a search for potential inhibitors for angiotensin I-converting enzyme（ACE） with minimum side effects in the treatment of hypertension. One peptide with the sequence Leu-Arg-Pro-Phe-Phe shows the strongest inhibition towards ACE with an IC50 value of 0.26 μmol/L in vitro. The study of structure-activity relationship shows that the introduction of a bulky group into the N-terminal of this series of inhibitors may enlarge steric hindrance, resulting in the poor inhibitory activity towards ACE. The inhibitory activity decreased in turn when L-Pro, D-Pro or Ac6c was at the C-terminal respectively. The binding interaction between each of these inhibitors and testicular ACE（tACE） was performed by molecular docking. The results suggest that Leu-Arg-Pro-Phe-Phe mainly occupied the S1 subsite of tACE, and made contact with tACE via seven H-bonds. It appeared that the site on the peptide that bound with tACE was influenced by the configuration of the amino acid, L or D-form, at the C-terminal of the peptide.

We studied the allelic frequency distributions and statistical forensic parameters of 21 new short tandem repeat(STR)loci and the amelogenin locus,which are not included in the combined DNA index system(CODIS),in a Russian ethnic minority group from the Inner Mongolia Autonomous Region,china.A total of 114 bloodstain samples from unrelated individuals were extracted and co-amplified with four fluorescence-labeled primers in a multiplex polymerase chain reaction(PCR)*** capillary electrophoresis,the PCR products of the 21 STR loci were separated and genotyped.A total of 161 alleles were observed in the Russian ethnic minority group,and corresponding allelic frequencies ranged from 0.0044 to *** 21 non-CODIS STR loci of the Russian ethnic minority group were characterized by high genetic diversity and therefore may be useful for elucidating the population's genetic background,for individual identification,and for paternity testing in forensic practice.
