BACKGROUND & OBJECTIVE:Anti-cancer effect of 5-Fluorouracil (5-FU) is mediated mainly by inhibition of the thymidylate synthase (TS),while dihydropyrimidine dehydrogenase (DPD) is an initial and a rate-limiting catabolic enzyme of *** this study, the mRNA expression profiles of TS,DPD were investigated in both healthy volunteers and Nasopharyngeal Carcinoma (NPC) *** data will be useful to select special schedules of chrono-therapy for nasopharyngeal carcinoma ***:A total of 14 healthy volunteers and 13 NPC patients were involved in this *** venous blood was sampled every 4 hours during one 2.-hour period starting at 12.00 *** circadian gene expression profiles of DPD,TS in PBMC were measured using the real-time quantitative RT-PCR *** whole blood reduced glutathione (GSH) concentration was determined by high performance liquid *** plasma cortisol concentration was determined by ***:Plasma cortisol levels of both groups displayed clear and similar circadian *** both groups,the plasma cortisol level peaked in the morning and was lowest at *** GSH concentrations showed statistically significant differences according to sampling time (ANOVA for Repeated Measures,F=5.182.P=0.02.) in both *** present study,the Mesor (rhythm-adjust mean) level of DPD gene copies was 1.31×10±5.74×10 copy/μg RNA (mesor±SEM) in healthy volunteers,and it was 3.2.×10±8.05×10 copy/μg RNA in NPC *** Mesor level of TS gene copies was 2.2.×10±4.90×10 copy/μg RNA in healthy volunteers, and it was 8.2.×10±1.66×10 copy/μg RNA in NPC *** was of statistical significance for DPD mRNA expression according to sampling time (ANOVA for Repeated Measures,F=8.2.5,P=0.002.,but not fit for the cosinor curve(Cosinor,P>0.05).It was of no statistical significance for TS gene (ANOVA for Repeated Measures,F=3.489,P= 0.083;Cosinor,P>0.05).CONCLUSION:a clear circadian rhythm was not displayed

Aimed at exploring the potential antitumor activities of xanthone derivatives, twenty-six synthetic hydroxylxanthones and benzoxanthones as well as their structurally perturbed analogs were examined for their cytotoxic activities against eight kinds of human cancer cell lines, HepG2.Lovo,KB,CNE2.HeLa,Capan2.K562.and *** of them exhibited higher cytotoxicity on HepG2.than the other seven cancer cell *** furthermore,six xanthone derivatives with the highest cytotoxicity against HepG2.showed excellent tumor specificity comparing with their effects on the normal liver cell line(L02..Several valuable structure-activity relationships were elicited from the cytotoxic activity *** pre{2.t results indicated that these xanthone derivatives may be exploitable as lead compounds to develop into a kind of high selectivity and low toxicity antitumor agents specific to liver cancer.

Objective:To analyze the efficacy and side-effect of teniposide plus cisplatin regimen on malignant glioma ***:A total of 56 patients with histologically confirmed malignant gliomas（WHO grade III/IV） were *** patients were received radiotherapy following *** were 2. patients received adjuvant chemotherapy and the other 32.patients received salvage *** chemotherapy regimen comprised of VM-2. 300mg/m2.plus DDP 80 mg/m2.which were divided for 3-5 days intravenous administration,every 3-4 weeks as a *** clinical efficacy and survival time of the patients were evaluated if the patients who were completed more than two cycles ***:Forty-nine patients were included for evaluation:complete remission（CR） was achieved in 2.cases（4.1%）and 6 cases（12.2.） were partial remission（PR）,while stable disease（SD）were found in 30 patients（61.3%）,and progressive disease（PD） were in 11 cases（2..4%）.Objective response rate（CR+PR） was 16.3%,and disease control rate（CR+PR+SD） was 77.6%.The progression-free survival（PFS） rate at 6 months was 55.1%.The survival rate at 1 year,2.years and 3 years was 63.3%,34.7%,and 2..4%,*** median overall survival time（OS） was 2..8 *** side-effect of the regimen was mainly hematological,with grade 3 or 4 neutropenia in twelve patients（2..5%）,and which was recovered in several days after receiving G-CSF ***:VM-2. plus DDP chemotherapy regimen can achieve fairly high response rate and disease control rate for patients with malignant gliomas,and the toxicity is acceptable.

[Objective]To investigate the circadian profiles of dihydropyrimidine dehydrogenase (DPD), thymidylate synthase (TS) in mRNA level on Chinese NPC patients. [Methods] In present study,total of 13 NPC patients were involved,9 males and 4 females. Peripheral veinal blood was collected at 6 time points (12.00,16:00,2.:00,and 2.:00,04:00, 08:00) during a single 2. hours from all the *** circadian gene expression profiles of DPD,TS in PBMC were measured using the real-time quantitative RT-PCR *** addition, the whole-blood reduced Glutathione (GSH),Dihydrouracil and uracil ratios (UH/U ratios), an indirect method for measuring DPD activity,HPLC method. [Results] In present study,Mesor (rhythm-adjust mean) of DPD gene copies was 6.2.×10± 7.76×10 copy/μg RNA (mesor±SEM),and it was 8.2.x10±8.33×10 copy/μg RNA for TS gene. It was of no statistical significance for DPD mRNA expression according to sampling time(ANOVA for Repeated Measures,F=1.067,P=0.369:Cosinor,P=0.078),and for TS (ANOVA for Repeated Measures,F=0.505,P=0.765,Cosinor,P=0.1068).The plasma cortisol displayed a clearly circadian rhythm and the peak time located at 09:14±01:11 (Corsinor,P＜ 0.0001).The mean value of UH/U was 4.80±0.2. and a significant circadian rhythm was not displayed (ANOVA for Repeated Measures,F=0.352.P=0.862.Corsinor,P=0.3053). The mesor value of GSH was 19.60±1.11 (nmol/L) and GSH displayed a possible circadian rhythm and the acrophase located at 05:02.(ANOVA for Repeated Measures,F=3.985,P=0.050: Corsinor,P=0.0645). [Conclusion]A clear circadian rhythm was not displayed for DPD gene and TS gene in mRNA level, also not found for DPD activity on Chinese NPC patients,although these patients still maintained an obvious circadian rhythm in plasma cortisol level and whole-blood reduced *** the chrono-modulated chemotherapy schedule using 5-Fu that based on the fixed rhythm of DPD activity needs more investigation for Chinese NPC patients.

AIM:To assess the effects of combined transcatheter arterial chemoembolization(TACE) and percutaneous ethanol ablation(PEA) in patients with large hepato-cellular carcinoma(HCC). METHODS:A total of 63 patients with unresectable large HCC were treated with TACE followed by *** largest dimension of the tumors ranged from 5.3 cm to *** survival rates,acute effects,toxicity and prognostic factors were analyzed. RESULTS:The cumulative survival rates at 1,3 and 5 years were 59.4%,2..4%and 15.8%,respectively(a median survival of 2..7 mo).Tumor area was reduced by more than 50%in 30(47.6%) *** 56 cases with increased a-fetoprotein(AFP) values,AFP level was declined by more than 75%.The combined thera-py was generally well *** two patients died from variceal bleeding associated with the *** Cox proportional hazards model showed that the num-ber of tumors,the tumor margin and the ethanol dose were independent prognostic factors. CONCLUSION:The combined TACE and PEA therapy is a promising approach for unresectable large HCC.

[Purpose]The purpose was to evaluate the safety and efficacy of CT-guided radioactive 12.I seeds permanent implantation in the treatment of extrahepatic extrapulmonary secondary malignancies.[Method]Twenty-four patients were included in this prospective *** primaries comprised 7 colorectal carcinomas,3 nasopharyngeal carcinomas,3 cervical carcinomas,2.thyroid carcinoma,and 1 vaginal melanoma,gastric carcinoma ,lung carcinima,ovary carcinoma,oropharynx carcinoma,primary hepatic carcinoma,duodenum carcinoma, pheochromocytoma,or breast *** single secondary solid carinoma locations included local lymph node, pelvic metastatic tumor,obturator internus,chest wall and local *** were 12.males,and 12.females,the median age was 56.5 years(range:31-76).The diameter of tumors was 1.5 6.5 cm,the average was 3.7 *** patients had undergone extensive ***-guided radioactive seeds permanent implantation employed a 12.-Iodine *** to Treatment Planning System(TPS),the matched peripheral dose(MPD) was 90～140Gy each local lession,and total number of the seeds implanted was 10～165 each *** or MRI follow-up was performed 2.months and every 2.or 3 months post *** endpoints were complications,local tumor control and progressive-free survival.[Results]Minor complications comprised GradeⅡradioactive rectum proctitis rectum morbidity(n=2.,pain(n=5),fever(n=2. and 2.seeds migration to lung in one *** median follow-up time for all 2. patients surviving was 6 months(range:1～2. months).One patient with oropharynx sqamous carcinima was in poor overall condition because of long time malnutrition,and died 2. days posttreatment due to progessive overall *** patient with vaginal melanoma died 3 months posttreatment due to acute cerebral hemorrhage which was assured by *** patient with right colon carcinoma died of bowel obstruction 4 months posttreatment due to long time *** patients
