The miR157-SPL-CNR module acts upstream of bHLH101 to negatively regulate iron deficiency responses in tomato

Iron(Fe)homeostasis is critical for plant growth,development,and stress *** levels are tightly controlled by intricate regulatory networks in which transcription factors(TFs)play a central role.A series of basic helix-loop-helix(b HLH)TFs have been shown to contribute to Fe homeostasis,but the regulatory layers beyond b HLH TFs remain largely ***,we demonstrate that the SQUAMOSA PROMOTER-BINDING PROTEIN-LIKE(SPL)TF Sl SPL-CNR negatively regulates Fe-deficiency responses in tomato(Solanum lycopersicum)*** deficiency rapidly repressed the expression of Sl SPL-CNR,and Fe deficiency responses were intensified in two clustered regularly interspaced palindromic repeats(CRISPR)/CRISPRassociated protein 9-generated Sl SPL-CNR knockout lines compared to the *** transcriptome analysis identified 47 Fe deficiencyresponsive genes the expression of which is negatively regulated by Sl SPL-CNR,one of which,Slb HLH101,helps regulate Fe uptake *** SPLCNR localizes the nucleus and interacts with the GTAC and BOX 4(ATTAAT)motifs in the Slb HLH101 promoter to repress its *** of Sl SPL-CNR expression in response to Fe deficiency was well correlated with the expression of the micro RNA Slymi *** R157-overexpressing tomato lines displayed enhanced Fe deficiency responses,as did Sl SPL-CNR loss-of-function *** propose that the Slymi R157-Sl SPL-CNR module represents a novel pathway that acts upstream of Slb HLH101 to regulate Fe homeostasis in tomato roots.

CG hypermethylation of the bHLH39 promoter regulates its expression and Fe deficiency responses in tomato roots

Iron(Fe)is an essential micronutrient for all organisms,including plants,whose limited bioavailability restricts plant growth,yield,and nutritional *** the transcriptional regulation of plant responses to Fe deficiency have been extensively studied,the contribution of epigenetic modulations,such as DNA methylation,remains poorly ***,we report that treatment with a DNA methylase inhibitor repressed Fe deficiency-induced responses in tomato(Solanum lycopersicum)roots,suggesting the importance of DNA methylation in regulating Fe deficiency *** changes in the DNA methylome in tomato roots responding to short-term(12 hours)and long-term(72 hours)Fe deficiency identified many differentially methylated regions(DMRs)and DMR-associated *** DMRs occurred at CHH sites under short-term Fe deficiency,whereas they were predominant at CG sites following long-term Fe ***,no correlation was detected between the changes in DNA methylation levels and the changes in transcript levels of the affected genes under either short-term or long-term ***,one exception was CG hypermethylation at the bHLH39 promoter,which was positively correlated with its transcriptional *** agreement,we detected lower CG methylation at the bHLH39 promoter and lower bHLH39 expression in MET1-RNA interference lines compared with wild-type ***-induced gene silencing of bHLH39 and luciferase reporter assays revealed that bHLH39 is positively involved in the modulation of Fe ***,we propose that dynamic epigenetic DNA methylation in the CG context at the bHLH39 promoter is involved in its transcriptional regulation,thus contributing to the Fe deficiency response of tomato.

Investigating the reactivation of historical landslides during the 2022 Luding M_(S)6.8 earthquake

On September 5,2022,a strong earthquake with a magnitude of MS6.8 struck Luding County in Sichuan Province,China,triggering thousands of landslides along the Dadu River in the northwest-southeast(NW-SE)*** investigated the reactivation characteristics of historical landslides within the epicentral area of the Luding earthquake to identify the initiation mechanism of earthquake-induced *** of the two newly triggered and historical landslides were analyzed using manual and threshold methods;the spatial distribution of landslides was assessed in relation to topographical and geological factors using remote sensing *** study sheds light on the spatial distribution patterns of landslides,especially those that occur above historical landslide *** results revealed a similarity in the spatial distribution trends between historical landslides and new ones induced by *** landslides tend to be concentrated within a range of 0.2 km from the river and 2 km from the ***,both rivers and faults predominantly influenced the reactivation of historical ***,the reactivated landslides are characterized by their small to medium size and are predominantly situated in historical landslide *** number of reactivated landslides surpassed that of previously documented historical landslides within the study *** provide insights into the critical factors responsible for historical landslides during the 2022 Luding earthquake,thereby enhancing our understanding of the potential implications for future co-seismic hazard assessments and mitigation strategies.

Spatial-temporal patterns and evolution of carbon storage in China's terrestrial ecosystems from 1980 to 2020

Analyzing the changes in carbon storage in terrestrial ecosystems caused by land use changes is a crucial part of exploring the carbon cycle. In addition, enhancing carbon storage in terrestrial ecosystems is an effective and environmentally friendly measure to sequester anthropogenic carbon emissions, which is significant for achieving carbon neutrality and curbing global climate change. This paper uses land use data and carbon density tables with the InVEST model to obtain a carbon storage distribution map of China. It further applies land use response elasticity coefficients, Theil index multi-stage nested decomposition, and spatial autocorrelation analysis to examine the spatial-temporal patterns, causes of changes, and evolution characteristics of carbon storage in terrestrial ecosystems from 1980 to 2020. The results show that the temporal changes in China’s carbon storage generally present an inverted S-curve, with an initial rapid decline followed by a slower decrease. Spatially, it features high levels in the northeast, low levels in the northwest, and a uniform distribution in the central and southern *** disturbance of land use type changes on terrestrial ecosystem carbon storage has been effectively mitigated. The significant reduction in grassland area in the Southwest region is the main source of carbon storage loss during the study period, and the encroachment of construction land on arable land in large urban agglomerations is one of the important causes of carbon storage loss. The Theil index multi-stage nested decomposition results indicate that the overall difference in carbon storage in China has decreased, while differences among cities within provinces and among counties within cities have increased. The influence of natural factors on the distribution of carbon storage is weakening, whereas the impact of human activities is becoming more profound, enhancing its influence on the spatial distribution of carbon storage in China. With the application of resins in various fields, numerous waste resins that are difficult to treat have been produced. The industrial wastewater containing Cr(Ⅵ) has severely polluted soil and groundwater environments, thereby endangering human health. Therefore, in this paper, a novel functionalized mesoporous adsorbent PPR-Z was synthesized from waste amidoxime resin for adsorbing Cr(Ⅵ). The waste amidoxime resin was first modified with H3PO4 and ZnCl_(2), and subsequently, it was carbonized through slow thermal decomposition. The static adsorption of PPR-Z conforms to the pseudo-second-order kinetic model and Langmuir isotherm, indicating that the Cr(Ⅵ) adsorption by PPR-Z is mostly chemical adsorption and exhibits single-layer adsorption. The saturated adsorption capacity of the adsorbent for Cr(Ⅵ) could reach 255.86 mg/g. The adsorbent could effectively reduce Cr(Ⅵ) to Cr(Ⅲ) and decrease the toxicity of Cr(Ⅵ) during adsorption. PPR-Z exhibited Cr(Ⅵ) selectivity in electroplating wastewater. The main mechanisms involved in the Cr(Ⅵ) adsorption are the chemical reduction of Cr(Ⅵ) into Cr(Ⅲ) and electrostatic and coordination interactions. Preparation of PPR-Z not only solves the problem of waste resin treatment but also effectively controls Cr(Ⅵ) pollution and realizes the concept of “treating waste with waste”.

Artificial ion channels that enable high-efficiency ion transport have important implications in nanofluidics and biomedical applications such as drug ***,we show a simulation-based chemical design of a biomimetic sodium channel that possesses permeation rate and selectivity potentially higher than those of the state-of-the-art natural vertebrate voltage-gated sodium ***,our theoretical findings have undergone empirical testing,aligning well with the Arrhenius law as derived from a diverse range of experimental *** high-efficiency ion transport is achieved by anchoring the carboxylate functional groups within the channel filter.A key chemical guiding principle underlying the ion channel design is that the free-energy barrier for the Na+passage across the channel should be comparable to typical thermal energy at room *** the implementation of the chemical design,we found that the relatively low free-energy barrier can be attributed to the compensation effect of the carboxylate groups to the partially lost oxygen shell of the ion within the ion channel,as well as to the consonant vibration of the ions inside and outside the *** mechanistic understanding brings new insight,at the molecular level,into the high-efficiency ion transport across the designed membrane *** proof of principle achieved from the simulations will stimulate future experimental confirmation and potential applications of the high-performance artificial channels in nanofluidics and in bioinspired iontronics.

Fully endoscopic far-lateral supracerebellar infratentorial approach for trigeminal neuralgia: illustrative case reports

Background Trigeminal neuralgia (TN) is a common cause of craniofacial pain. The retrosigmoid approach is usually used to treat TN, but no cases of endoscopic far-lateral supracerebellar infratentorial approach (EF-SCITA) were used to undergo operation for *** presentation Two patients were presented with severe facial pain and preliminary diagnosis was TN. Preoperative magnetic resonance imaging revealed that a superior cerebellar artery (SCA) compressed the trigeminal nerve in case 1, and a tumor located in the petrous apex extending into the Meckel’s cave compressed the trigeminal nerve in case 2. Operations were achieved through the EF-SCITA. The pain was totally relieved with no postsurgical complications in both *** We present the first two case reports of EF-SCITA to relieve classical and secondary TN successfully. The EF-SCITA can be a promising approach for treating TN.
