Genetic benefits of female from extra-pair paternity are context dependent within the socially monogamous Tree Sparrow

Females actively seek extra-pair paternity(EPP)to acquire a fitness advantage for their ***“contextdependence hypothesis”posits that female extra-pair mate choice has plasticity in response to environmental conditions,and therefore magnitude of female genetic benefits from EPP depends on the environmental ***,chronic heavy metal pollution can cause adverse effects on fitness-related traits of wild ***,few studies were available on the interactions between heavy metal pollution and *** selected an area that was contaminated by heavy metals for more than 60 years(Baiyin,BY),a relatively unpolluted area(Liujiaxia,LJX),and Tree Sparrows(Passer montanus)as study species to explore the response of female extrapair mate choice and genetic benefits from EPP to heavy metal pollution in socially monogamous *** relatedness between social mates and extra-pair mates was ***,we compared the body size and heterozygosity of extra-pair offspring(EPO),within-pair offspring(WPO),social males and extra-pair males from the two Tree Sparrow *** found that at BY,female Tree Sparrows tended to choose extra-pair males with larger body size and lower genetic similarity,thereby producing higher heterozygosity and larger body size of EPO compared with those of ***,no similar phenomenon was observed in the Tree Sparrow population from *** addition,there was a significant interaction between population and paternity in the analyses of the fitness-related traits,suggesting that environmental variation could affect female genetic benefits from *** study confirmed the existence of presumed interactions between environmental pollution and EPP within the natural population of socially monogamous Tree *** findings shed lights on the possible effects of long-term environmental stress on mating system in wild birds.

An alternative splice variant of human thioredoxin

An alternative splice variant of human thioredoxin (hTRX) has been identified by using reverse transcription_polymerase chain reaction, cloning and expressing of hTRX_encoding gene, and DNA sequencing. This variant contains hTRX exons 1, 2, 4 and 5. The entire protein encoding region of this variant, named hTRXδ3, is identical to hTRX except for the omission of exon 3. Human TRX and TRXδ3 cDNA has been expressed in \%***\%. The catalytic activity of rhTRXδ3 on DTT_dependent insulin reduction is lower than that of rhTRX, although hTRXδ3 still contains the active site.

Optimal analysis conditions for sperm motility parameters with a CASA system in a passerine bird, Passer montanus

Background:Sperm motility parameters,which can be measured objectively and repeatedly by a computer-assisted sperm analysis(CASA)system,are important indicators of sperm ***,the sperm motility parameters assessed by a CASA system can be affected by various factors,including instrument components and settings,sperm preparation or analysis *** date,no standardized protocol is available that would permit to assess sperm kinetic characteristics in passerine birds and this lack precludes any comparison of sperm swimming ability and sperm quality across ***:In this study,we chose the Tree Sparrow(Passer montanus)as the object to evaluate sperm motility parameters,including sperm motility,sperm velocity and sperm movement trajectory,at different analysis time,temperatures and pH using the WLJY-9000 CASA ***:Sperm motility parameters remained statistically unchanged at 1‒9 *** motility was similar at 38℃ and 40℃,but a greater percentage of slow progressive sperm was detected at 38℃ compared to 40℃ and 42℃.Additionally,progressive motility was lower and immotility was higher at 42℃than 38℃and/or 40℃(close to the body temperature of the Tree Sparrow).The percentages of rapid progressive sperm,progressive sperm and immotile sperm were statistically similar at pH 7.0,7.5 and 8.0 with the exception of lower percentage of progressive sperm at pH 7.0 compared to pH *** addition,slower sperm velocity and worse sperm movement trajectory were found at pH 6.0 and 9.0 than those at pH 8.0,7.5 or ***:Our study indicates that the ideal conditions for sperm motility parameters assessment in Tree Sparrow are obtained between 1 and 9 min after dilution,an environment at body temperature(40℃)and a pH around *** results of this study provide a reference for the evaluation of sperm characteristics and sperm quality using a CASA system in passerine birds.

Effects of heavy metals Cd^(2+),Pb^(2+)and Zn^(2+)on DNA damage of loach Misgurnus anguillicaudatus

The effects of heavy metals Cd^(2+),Pb^(2+)and Zn^(2+)at 0.05,0.5 and 5.0 mg/L level and their interactions at 0.5 mg/L level on DNA damage in hepatopancreas of loach Misgurnus anguillicaudatus for 1-35 days exposure were examined by single cell gel electrophoresis(SCGE).For each test group,20 loaches with similar body size(5.17-7.99 g;11.79-13.21 cm)were selected and kept in aquaria with dechlori-nated water at(22±1)℃and fed a commercial diet every 48 *** to the percentage of damaged DNA with tail and its TL/D(tail length to diameter of nucleus)value,the relationship between DNA damage degree and heavy metal dose and exposure time was *** showed that the percentage of damaged DNA and the TL/D value were increased with the prolonged exposure *** highest percentage(84.85%)of damaged DNA was shown in 5.0 mg/L Zn^(2+)group after 28 days exposure and the biggest TL/D value(2.50)in all treated groups after 35 days *** the first treated week,the damnification of DNA was mainly recognized as the first level,after that time,the third damaged level was mostly observed and the percentage of damaged DNA was beyond 80%.The joint toxic effects among Cd^(2+),Pb^(2+)or Zn^(2+)revealed much complexity,but it generally displayed that the presence of Cd^(2+)could enhance the genotoxicity of Pb^(2+)or Zn^(2+).In conclusion,the results suggestedthattherewasasignificanttime-anddose-depended relationship between the heavy metal and DNA damage in hepatopancreas of loach,and SCGE could represent a useful means to evaluate the genotoxicity of environmental contamination on aquatic organisms.

Secretory expression of the fusion protein composed of human IL-2 and PreS antigen of hepatitis B virus in mammalian cells

In order to make entire HBV preSAg secrete from mammalian cells, we constructed an eukaryotic expression vector by using leader sequence of human interleukin-2 (IL-2) as secretory signal peptide, and using high hydrophilic amino acids as the linker between IL-2 C end and preSAg N end. As a result, the IL-2preS fusing protein could be secreted from mammalian cells transfected with the reconstructed vector and the expression efficiency was identical to that of natural IL-2. It was considered that the retentive effect of preS1Ag could be successfully bypassed. The results not only laid a theoretical and practical foundation for constructing specific gene vaccine against HBV persistent infection, but also supplied experimental evidence for studying modulation of protein secretory expression.

A low-jitter low-power monolithically integrated optical receiver for SDH STM-16

A high-scale integrated optical receiver including a preamplifier, a limiting amplifier, a clock and data recovery (CDR) block, and a 1:4 demultiplexer (DEMUX) has been realized in a 0.25 μm CMOS technology. Using the loop parameter optimization method and the low-jitter circuit design technique, the rms and peak-to-peak jitter of the recovered 625 MHz clock are 9.4 and 46.3 ps, respectively, which meet the jitter specifications stipulated in ITU-T recommendation G.958. In response to 2.5 Gb/s PRBS input data (2 23 1), the recovered and frequency divided 625 MHz clock has a phase noise of 83.8 dBc/Hz at 20 kHz offset and the 2.5 Gb/s PRBS data has been demultiplexed into four 625 Mb/s data. The power dissipation is only 0.3 W under a single 3.3 V supply (excluding output buffers).

FAM 19A4 is an abbreviation for family with sequence similarity 19 （chemokine （C-C motif）-Iike） member A4, which is a secretory protein expressed in low levels in normal tissues. The biological functions of FAM19A4 remain to be determined, and its potential receptor（s） is unclarified. In this study, we demonstrated that FAM 19A4 was a classical secretory protein and we verified for the first time that its mature protein is composed of 95 amino acids. We found that the expression of this novel cytokine was upregulated in lipopolysaccharide （LPS）-stimulated monocytes and macrophages and was typically in polarized M 1. FAM 19A4 shows chemotactic activities on macrophages and enhances the macrophage phagocytosis of zymosan both in vitro and in vivo with noticeable increases of the phosphorylation of protein kinase B （Akt）. FAM 19A4 can also increase the release of reactive oxygen species （ROS） upon zymosan stimulation. Furthermore, based on receptor internalization, radio ligand binding assays and receptor blockage, we demonstrated for the first time that FAM19A4 is a novel ligand of formyl peptide receptor 1 （FPR1）. The above data indicate that upon inflammatory stimulation, monocyte/macrophage-derived FAM 19A4 may play a crucial role in the migration and activation of macrophages during pathogenic infections.
