Beyond glycemic control, applications of glucagon-like peptide-1 receptor (GLP-1r) agonists (GLP-1 RAs) inhibit inflammationand plaque development in murine atherosclerotic models. However, whether they modulate hematopoietic stem/progenitor cells(HSPCs)to prohibit skewed myelopoiesis in hypercholesteremia remains unknown. In this study, GLP-1r expression in fluorescenceactivated cell sorting (FACS)-sorted wild-type HSPCs was determined by capillary western blotting. Bone marrow cells (BMCs)of wild-type or GLP-1r−/− mice were transplanted into lethally irradiated low-density lipoprotein receptor deficient (LDLr−/−)recipients followed by high-fat diet (HFD) for chimerism analysis by FACS. In parallel, LDLr−/− mice were placed on HFD for 6weeks and then treated with saline or Exendin-4 (Ex-4) for another 6 weeks. HSPC frequency and cell cycle were analyzed byFACS, and intracellular metabolite levels were assessed by targeted metabolomics. The results demonstrated that HSPCs expressedGLP-1r and transplantation of GLP-1r−/− BMCs resulted in skewed myelopoiesis in hypercholesterolemic LDLr−/− *** vitro, Ex-4 treatment of FACS-purified HSPCs suppressed cell expansion and granulocyte production induced by LDL. In vivo, Ex-4treatment inhibited plaque progression, suppressed HSPC proliferation, and modified glycolytic and lipid metabolism in HSPCs ofhypercholesteremic LDLr−/− mice. In conclusion, Ex-4 could directly inhibit HSPC proliferation induced by hypercholesteremia.

The emergence of Omicron has gained global attention since November 2021. Of the Omicron strains, Omicron XBB.1.5 is the most recently identified. It carries deletions and more than 30 mutations, which have modified the biological features of severe acute respiratory syndrome coronavirus-2(SARS-CoV-2) for enhanced transmissibility and immune escape capacity. People living with human immunodeficiency virus(PLWH) are featured with acquired cluster of differentiation(CD)4+ T cell deficiency and at a higher risk of progression to severe coronavirus disease 2019(COVID-19). Vaccines are the most protective tools against SARS-CoV-2 infection. Nevertheless, the extent to which booster vaccination induces neutralizing antibody(NAb) production against Omicron XBB.1.5 in PLWH is not well defined.

The outbreak of coronavirus disease 2019(COVID-19)caused by the new virus SARS-CoV-2 has been announced as a public health emergency of international concern.1–3 The clinical features of patients with COVID-19 range from common fever and cough to other rare symptoms,such as diarrhea and *** disease can progress quickly,and 2–3%of patients die within a short time,which is generally due to multiple organ failure.4–7 Clinically,COVID-19 patients are classified into mild,moderate,severe,and critical cases.5–7 The immune response against SARS-CoV-2 is probably linked to the severity of ***,Zheng et al.8 showed that elevated levels of T-cell exhaustion and reduced functional diversity of T cells in peripheral blood may predict severe progression in COVID-19 patients;however,a more comprehensive understanding of the pathology of SARS-CoV-2 infection remains to be ***,we profiled immune cellular components using mass cytometry(CyTOF)to analyze the peripheral blood mononuclear cells(PBMCs)from patients with differences in disease progression by comparing with the PBMCs from healthy donors(HDs).

The COVID-19 pandemic has emerged as a global health emergency due to its association with severe pneumonia and relative high ***,the molecular characteristics and pathological features underlying COVID-19 pneumonia remain largely *** characterize molecular mechanisms underlying COVID-19 pathogenesis in the lung tissue using a proteomic approach,fresh lung tissues were obtained from newly deceased patients with COVID-19 *** virus inactivation,a quantitative proteomic approach combined with bioinformatics analysis was used to detect proteomic changes in the SARS-CoV-2-infected lung *** identified significant differentially expressed proteins involved in a variety of fundamental biological processes including cellular metabolism,blood coagulation,immune response,angiogenesis,and cell microenvironment *** inflammatory factors were upregulated,which was possibly caused by the activation of NF-κB *** dysregulation of the lung proteome in response to SARS-CoV-2 infection was *** results systematically outlined the molecular pathological features in terms of the lung response to SARS-CoV-2 infection,and provided the scientific basis for the therapeutic target that is urgently needed to control the COVID-19 pandemic.

随着中国经济社会快速发展以及旅游日益融入中国普通民众生活,住宿业发展迅速,但目前酒店管理专业学生的高离职率困扰着酒店管理实践。以在酒店实习的该专业学生为研究样本,利用扎根理论研究方法详细采集数据,并对其逐级分类编码,在理论饱和的状态下完成理论抽样。研究认为,在同样条件下,那些具有与酒店行业特征不匹配职业定位的酒店管理专业学生更易离职。从心理学角度深入剖析,可以从学生专业选择、学校人才培养、酒店人事资源管理等方面寻求有效对策。
