A novel bioartificial pancreas fabricated via islets microencapsulation in anti-adhesive core-shell microgels and macroencapsulation in a hydrogel scaffold prevascularized in vivo

Islets transplantation is a promising treatment for type 1 diabetes mellitus. However, severe host immune rejection and poor oxygen/nutrients supply due to the lack of surrounding capillary network often lead to transplantation failure. Herein, a novel bioartificial pancreas is constructed via islets microencapsulation in core-shell microgels and macroencapsulation in a hydrogel scaffold prevascularized in vivo. Specifically, a hydrogel scaffold containing methacrylated gelatin (GelMA), methacrylated heparin (HepMA) and vascular endothelial growth factor (VEGF) is fabricated, which can delivery VEGF in a sustained style and thus induce subcutaneous angiogenesis. In addition, islets-laden core-shell microgels using methacrylated hyaluronic acid (HAMA) as microgel core and poly(ethylene glycol) diacrylate (PEGDA)/carboxybetaine methacrylate (CBMA) as shell layer are prepared, which provide a favorable microenvironment for islets and simultaneously the inhibition of host immune rejection via anti-adhesion of proteins and immunocytes. As a result of the synergistic effect between anti-adhesive core-shell microgels and prevascularized hydrogel scaffold, the bioartificial pancreas can reverse the blood glucose levels of diabetic mice from hyperglycemia to normoglycemia for at least 90 days. We believe this bioartificial pancreas and relevant fabrication method provide a new strategy to treat type 1 diabetes, and also has broad potential applications in other cell therapies.

Azomethine-H as a Highly Selective Fluorescent Probe for Fe^3＋ Detection in 100% Aqueous Solution and Its Application in Living Cell Imaging

A simple assay for the detection of Fe^3＋ in water by means of fluorescence spectroscopy was developed based on a commercially available reagent, Azomethine-H（A-H）, allowing sensing trace levels of Fe^3＋ with high selectivity over other cations. A significant fluorescence quenching of A-H at 424 nm was found after its binding with Fe^3＋ in 100% aqueous solution at pH=7.0, while other physiologically relevant metal ions posed little interference. The fluorescence responses can be well described by the modified Stern-Volmer equation. A good linear relationship（Re=0.9904） was observed up to 1.6x10 5 mol/L Fe^3＋ ions. The detection limit, calculated via the 3tr IUPAC（international union of pure and applied chemistry） criteria, was 1.95x10-7 mol/L. Moreover, the colorimetric and fluorescent response of A-H to Fe^3＋ can be conveniently detected by the naked eye, providing a facile method for visual detection of Fe^3＋. The proposed method was used to determine Fe^3＋ in water samples. Moreover, inverted fluorescence microscopy imaging using human umbilical vein endothelial cells shows that A-H can be used as an effective fluorescent probe for detecting Fe^3＋ in living cells.

Low temperature Si/Si wafer direct bonding using a plasma activated method

Manufacturing and integration of micro-electro-mechanical systems （MEMS） devices and integrated circuits （ICs） by wafer bonding often generate problems caused by thermal properties of materials. This paper presents a low temperature wafer direct bonding process assisted by 02 plasma. Silicon wafers were treated with wet chemical cleaning and subsequently activated by 02 plasma in the etch element of a sputtering system. Then, two wafers were brought into contact in the bonder followed by annealing in N2 atmosphere for several hours. An infrared imaging system was used to detect bonding defects and a razor blade test was carried out to determine surface energy. The bonding yield reaches 90%--95% and the achieved surface energy is 1.76 J/m2 when the bonded wafers are annealed at 350 ~C in N2 atmosphere for 2 h. Void formation was systematically observed and eli-mination methods were proposed. The size and density of voids greatly depend on the annealing temperature. Short O2 plasma treatment for 60 s can alleviate void formation and enhance surface energy. A pulling test reveals that the bonding strength is more than 11.0 MPa. This low temperature wafer direct bonding process provides an efficient and reliable method for 3D integration, system on chip, and MEMS packaging.

Effects of Fermentation Product Containing Phytase on Productive Performance,Egg Quality,and Phosphorous Apparent Metabolism of Laying Hens Fed Different Levels of Phosphorus

This study investigated the effects of fermentation product containing phytase （FPP） that was fermented using waste vinegar residue （WVR） as substrate from Aspergillusficuum NTG-23 on productive performance, egg quality, and phosphorus apparent metabolism of laying hens. First, 375 22-wk-old Jinghong hens were allocated into 5 treatments （5 replicates of 15 hens each） in an 8-wk experiment for evaluating the parameters of productive performance, egg quality, serum, and tibia. Experimental diets contained 4% FPP and 96% corn-soybean diet. The levels of dicalciurn phosphate （DCP） were 1.34, 1.01, 0.67, 0.34 and 0%. Next, thirty 31-wk-old Jinghong hens were fed 5 types of diets for evaluating phosphorous apparent metabolism rate. Egg productive rate, egg weight, feed conversion ratio, Haugh unit, egg albumen height, serum calcium, tibia ash, tibia ash calcium and tibia breaking strength were not different significantly among 5 treatments. The significant difference of average daily feed intake was not appeared when the DCP content of corn-soybean-FPP diet was reduced to 0.67%; the eggshell hardness, eggshell thickness and serum phosphorus were not reduced significantly until the DCP content of corn-soybean-FPP diet was reduced to 0.34%. The yolk color was improved when the laying hens fed deficient DCP corn-soybean-FPP diet. A 22.14% reduction in excreta phosphorus was observed when the laying hens fed low phosphorus （0.67% DCP） corn-soybean-FPP diet. A 30% elevation of phosphorus apparent metabolism rate was obtained when the DCP content of corn-soybean-FPP diet was decreased from 1.34 to 1.01%. The reducing cost of layer diet was totalized about 120 CNY 1000 kg1 diet when the content of DCP was 0.67% in corn-soybean-FPP diet. These results indicated that FPP could be applied in laying hen as a potential, cost-effective and rational application of WVR.

Total laparoscopic liver resection in 78 patients

AIM： To summarize the clinical experience of laparoscopic hepatectomy at a single center. METHODS： Between November 2003 and March 2009, 78 patients with hepatocellular carcinoma （n = 39）, metastatic liver carcinoma （n = 10）, and benign liver neoplasms （n = 29） underwent laparoscopic hepatectomy in our unit. A retrospective analysis was done on the clinical outcomes of the 78 patients. RESULTS： The lesions were located in segments Ⅰ （n = 3）, Ⅱ （n = 16）, Ⅲ （n = 24）, Ⅳ （n = 11）, Ⅴ （n = ii）, Ⅵ （n = 9）, and Ⅷ （n = 4）. The lesion sizes ranged from 0.8 to 15 cm. The number of lesions was three （n = 4）, two （n = 8） and one （n = 66） in the study cohort. The surgical procedures included left hemi-hepatectomy （n = 7）, left lateral lobectomy （n = 14）, segmentectomy （n = 11）, local resection （n = 39）, and resection of metastatic liver lesions during laparoscopic surgery for rectal cancer （n = 7）. Laparoscopic liver resection was successful in all patients, with no conversion to open procedures. Only four patients received blood transfusion （400-800 mL）. There were no perioperative complications, such as bleeding and biliary leakage. The liver function of all patients recovered within 1 wk, and no liver failure occurred. CONCLUSION： Laparoscopic hepatectomy is a safe and feasible operation with minimal surgical trauma. It should be performed by a surgeon with sufficient experience in open hepatic resection and who is proficient in laparoscopy.
