Transcriptome analysis reveals long noncoding RNAs involved in fiber development in cotton (Gossypium arboreum)

Long noncoding RNAs （lncRNAs） play important roles in various biological regulatory processes in yeast, mammals, and plants. However, no systematic identification of lncRNAs has been reported in Gossypium arboreum. In this study, the strand-specific RNA sequencing （ssRNA-seq） of samples from cotton fibers and leaves was performed, and lncRNAs involved in fiber initiation and elongation processes were systematically identified and analyzed. We identified 5,996 lncRNAs, of which 3,510 and 2,486 can be classified as long intergenic noncoding RNAs （lincRNAs） and natural antisense transcripts （IncNAT）, respectively. LincRNAs and lncNATs are similar in many aspects, but have some differences in exon number, exon length, and transcript length. Expression analysis revealed that 51.9% of lincRNAs and 54.5% of lncNATs transcripts were preferentially expressed at one stage of fiber development, and were significantly highly expressed than protein-coding tran- scripts （21.7%）. During the fiber and rapid elongation stages, rapid and dynamic changes in lncRNAs may contribute to fiber development in cotton. This work describes a set of lncRNAs that are involved in fiber development. The characterization and expression analysis of lncRNAs will facilitate future studies on their roles in fiber development in cotton.

Introduction:Genome sequence plays an important role in both basic and applied *** raimondii,the putative contributor of the D subgenome of upland cotton(***,highlights the need to improve the genome quality rapidly and ***:We performed Hi-C sequencing of *** and reassembled its genome based on a set of new Hi-C data and previously published *** also compared the reassembled genome sequenee with the previously published G raimondii genomes for gene and genome sequence ***:A total of 9842%of scaffold sequences were clustered successfully,among which 99.72%of the clustered sequences were ordered and 99.92%of the ordered sequences were oriented with *** evaluation of results by heat-map and collinearity analysis revealed that the current reassembled genome is significantly improved than the previous one(Nat Genet 44:98-1103,2012).Conclusion:This improvement in G raimondii genome not only provides a better reference to increase study efficiency but also offers a new way to assemble cotton ***,Hi-C data of *** may be used for 3D structure research or regulating analysis.

Background： Virescence, as a recognizable phenotype available for research on chloroplast development and in the early development stage of cotton, is not only photosynthesis but also for heterosis exploitation in cotton Methods： In current study, for fine mapping of virescent-1 （V1） in cotton, three populations with a total of 5 678 individuals were constructed using T582 which has the virescent trait. Tobacco rattle virus, TRV1 and TRV2 （pYL156）, were used as vectors for the virus-induced gene silencing （VlGS） assay. Results： The V1 gene was fine-mapped to a 20 kb interval on chromosome 20 of tetraploid cotton. We identified only one candidate gene with four single nucleotide polymorphisms between parents, among which the single nucleotide polymorphism at the position of 1 082 base pair caused the change of amino acid residue from Arg （3-79） to Lys （]-582）. The relative expression of the candidate gene in virescent plants was extensively lower than that in normal plants. Nullification of the gene by VlGS significantly turned the green leaf of normal cotton plants into yellow. We named this candidate gene as GhRVL. Conclusions： This study will facilitate the further research on virescent formation, and will be useful for breeding of hybrid cottons.

Regulating planting density and nitrogen(N)fertilization could delay chlorophyll(Chl)degradation and leaf senescence in maize *** study measured changes in ear leaf green area(GLA_(ear)),Chl content,the activities of Chl a-degrading enzymes after silking,and the post-silking dry matter accumulation and grain yield under multiple planting densities and N fertilization *** dynamic change of GLA_(ear)after silking fitted to the logistic model,and the GLA_(ear) duration and the GLAearat 42 d after silking were affected mainly by the duration of the initial senescence period(T_(1))which was a key factor of the leaf *** average chlorophyllase(CLH)activity was 8.3 times higher than pheophytinase activity and contributed most to the Chl content,indicating that CLH is a key enzyme for degrading Chl a in *** density increased the CLH activity and decreased the Chl content,T1,GLAear,and GLA_(ear) *** high density,appropriate N application reduced CLH activity,increased Chl content,prolonged T1,alleviated high-density-induced leaf senescence,and increased post-silking dry matter accumulation and grain yield.

全域旅游是新时代旅游产业高质量发展的核心战略，对全域旅游发展水平进行评价有助于我们认清产业演进规律和发展现状，改变只强调数量和规模的产业发展思路，树立产业综合质量发展观，加快旅游产业的全面优化升级。基于现有的统计数据基础，对全域旅游发展水平进行全面衡量和评价仍较为困难，文章在总结分析全域旅游内涵的基础上，从产业域、空间域和管理域三个维度选取一些有代表性的评价指标，采用全局主成分分析和熵值法相结合的方式，对我国31个省（市、区）的全域旅游发展水平进行了试评价。研究表明：31个省（市、区）的全域旅游发展水平稳步提升，东部地区领先于其他地区；从各维度看，产业域水平发展稳定，空间域水平发展迅猛，而管理域发展水平波动性比较明显。我国全域旅游发展存在着显著的全局空间自相关性，主要表现为空间集聚效应，且具有增强的趋势；大多数省份在地理分布上多处于高高集聚区和低低集聚区，全域旅游发展在空间上存在较为明显的交互影响关系。研究认为，产业高质量发展、完善的空间设施共享和综合治理创新是推进全域旅游发展、促进区域旅游产业转型升级的重要内容，在此基础上，要鼓励区域旅游产业统筹协调发展。

Protective mechanism of Paeoniae Radix Alba against chemical liver injury based on network pharmacology,molecular docking,and in vitro experiments

Objective:To explore and validate the potential targets of Paeoniae Radix Alba(***,Bai Shao)in protecting against chemical liver injury through network pharmacology,molecular docking technology,and in vitro cell ***:Network pharmacology was used to identify the common potential targets of *** and chemical liver *** docking was used to fit the components,which were subsequently verified in vitro.A cell model of hepatic fibrosis was established by activating hepatic stellate cell(HSC)-LX2 cells with 10 ng/mL transforming growth factor-β*** cells were exposed to different concentrations of total glucosides of paeony(TGP),the active substance of ***,and then evaluated using the cell counting kit-8 assay,enzyme-linked immunosorbent assay,and western ***:Analysis through network pharmacology revealed 13 key compounds of ***,and the potential targets for preventing chemical liver injury were IL-6,AKT serine/threonine kinase 1,jun protooncogene,heat shock protein 90 alpha family class A member 1(HSP90AA1),peroxisome proliferator activated receptor gamma(PPARG),PTGS2,and *** Ontology(GO)enrichment analysis indicated the involvement of response to drugs,membrane rafts,and peptide *** Encyclopedia of Genes and Genomes(KEGG)enrichment analysis revealed that the main pathways involved lipid and atherosclerosis and chemical carcinogenesis-receptor *** and albiflorin exhibited strong affinity for HSP90AA1,PTGS2,PPARG,and *** concentrations of TGP can inhibit the expression of COL-I,COL-III,IL-6,TNF-a,IL-1β,HSP-90a,and PTGS2 while increasing the expression of PPAR-γand CASP3 in activated HSC-LX2 ***:*** primarily can regulate targets such as HSP90AA1,PTGS2,PPARG,***,the main active compound of ***,protects against chemical liver injury by reducing the inflammatory response,activating apoptotic proteins,and promoting the apoptosis of activated HSCs.

Cotton is an important economic crop which could provide natural fiber,plant oil and ***,existence of pigment gland and toxic gossypol inhibit full use of cottonseed ***(miRNAs) play important roles in post-transcription,but the role of miRNAs in pigment gland formation remains ***,4 pairs of near-isogenic lines(NILs) consist of 3 glanded and 4 glandless cotton lines were used for small RNA *** total of 58 known and 58 novel miRNAs were *** them,10 and 6 miRNAs showed different expression between dominant glandless and glanded cotton,recessive glandless and glanded cotton,*** miRNAs potentially target 73 and 90 genes,respectively,including phytohormone-related genes and transcription factors,such as ARF,ERF,RGA and ***,two novel miRNAs Ghnmir1/Ghnmir9 and their targets bHLH-MYC transcription factors were differentially expressed between all glanded and glandless cotton,implying they probably take part in pigment gland *** on Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) analysis,plant hormone signal transduction,MAPK signaling pathway and phagosome may be relative to regulation of glandless *** study provides evidence for understanding miRNA potential regulatory roles in pigment gland development,which is helpful to clarify the molecular mechanisms of pigment gland formation and future low-gossypol cotton breeding.

文章通过使用各省经济距离空间权重矩阵、地理距离空间权重矩阵、邻接关系空间权重矩阵,检验中国知识密集型服务业集聚对区域创新的影响。结果表明,中国各省知识密集型服务业具有很强的空间关联性,知识密集型服务业集聚能显著促进区域创新,知识密集型服务业集聚通过技术溢出效应显著促进区域创新,知识密集型服务业集聚也通过规模经济效应促进区域创新。通过分解知识密集型服务业集聚影响区域创新的空间效应可发现,中国各省的知识密集型服务业集聚不能促进周边地区区域创新,各省资本劳动比也不能促进周边地区区域创新。
