Impact of intra-operative cholangiography and parenchymal resection to donor liver function in living donor liver transplantation

BACKGROUND: Living donor liver transplantation(LDLT)has been widely accepted over the past decade, and hepatic dysfunction often occurs in the donor in the early stage after liver donation. The present study aimed to evaluate the effect of intraoperative cholangiography(IOC) and parenchymal resection on liver function of donors in LDLT, and to assess the role of IOC in influencing the biliary complications and improving the overall ***: Data from 40 patients who had donated their right lobes for LDLT were analyzed. Total bilirubin(TB), alanine aminotransferase(ALT), aspartate aminotransferase(AST),alkaline phosphatase(ALP) and γ-glutamyl transpeptidase(GGT)at different time points were compared, and the follow-up data and the biliary complications were also ***: The ALT and AST values were significantly increased after IOC(P0.05) or parenchymal resection(P>0.05). The biochemical changes caused by IOC or parenchymal resection were not correlated with the degree of post-operative liver injury or the recovery of liver function. The liver functions of the donors after operation were stable, and none of the donors suffered from biliary stenosis or leakage during the ***: IOC and parenchymal resection may induce a transient increase in liver enzymes of donors in LDLT, but do not affect the recovery of liver function after operation. Moreover,the routine IOC is helpful to clarify the division line of the hepatic duct, thus reducing the biliary complication rate.

Microstructure and mechanical properties of a compound reinforced Mg95Y2.5Zn2.5 alloy with long period stacking ordered phase and W phase

The microstructure evolution of Mg100-2xYxZnx （x=2, 2.5, 3, 3.5） alloys was investigated. Results show that the Mg100-2xYxZnx alloys are composed of a-Mg, long period stacking ordered （LPSO） phase and eutectic structure phase （W phase）, and the Mg95Y2.5Zn2.5 alloy has the best comprehensive mechanical properties. Subsequently, the microstructure evolution of the optimized alloy Mg95Y2.5Zn2.5 during solidification and heat treatment processes was analyzed and discussed by means of OM, SEM, TEM, XRD and DTA. After heat treatment, the lamellar phase 14H-LPSO precipitated in a-Mg and W phase transforms into particle phase （MgyZn2）. Due to the compound reinforcement effect of the particle phase and LPSO phase （18R＋14H）, the mechanical properties of the alloy are enhanced. The tensile strength and elongation of the Mg95Y2.5Zn2.5 alloy is improved by 9.1% and 31.3% to 215 MPa and 10.5%, respectively, after solid-solution treatment.

Quercetin in combating H2O2 induced early cell apoptosis and mitochondrial damage to normal human keratinocytes

Background Oxidative stress plays an important role in the pathogenesis of epidermal diseases. This study aimed to investigate the effects of quercetin on the anti-oxidative response and on mitochondrial protection in cultured normal human keratinocytes. Methods Cultured HaCaT cells were treated with different concentrations of H202 （0, 50, 100, 250, 500 pmol/L） for different periods of time （0.5, 1, 2, 4 hours） to establish an oxidative stress model. The cultured HaCaT cells were randomly assigned to control, H2O2, and quercetin＋H2O2 groups. For the quercetin groups, the cells were treated with different concentrations of quercetin （0, 10, 25, 50 umol/L） before exposure to H2O2. Morphological changes of the cells were observed under an inverted microscope and an electron microscope. The cell viability was detected by the MIF method. The cell apoptosis （AnnexinV/propidium iodide double stain） and mitochondrial membrane potential （△ψm） changes were detected by flow cytometry. Results An oxidative stress model of HaCaT cells was established under a suitable concentration （250 umol/L） and treated time of H2O2 （2 hours）. The cell viability and △ψm decreased in a concentration-dependent and time-dependent manner while the percentage of apoptotic cells significantly increased in the H2O2 groups compared with the control group （P 〈0.05）. The cell viability and △ψm of the quercetin treated group increased （P 〈0.05） and the percentage of apoptotic cells decreased at concentrations of 1-50 umol/L quercetin （P 〈0.01） compared with H2O2 treated group. Conclusion Quercetin can relieve the cell damage and apoptosis from H2O2 induced injury to HaCaT cells by anti-oxidation and mitochondrial protection.

Transcriptome-wide evolutionary analysis on essential brown algae(Phaeophyceae)in China

Brown algae （Chromista, Ochrophyta, Phaeophyceae） are a large group of multicellular algae that play im-portant roles in the ocean＇s ecosystem and biodiversity. However, poor molecular bases for studying their phylogenetic evolutions and novel metabolic characteristics have hampered progress in the field. In this study, we sequenced the de novo transcriptome of 18 major species of brown algae in China, covering six orders and seven families, using the high-throughput sequencing platform Illumina HiSeq 2000. From the transcriptome data of these 18 species and publicly available genome data of Ectocarpus siliculosus and Phaeodactylum tricornutum, we identified 108 nuclear-generated orthologous genes and clarified the phy-logenetic relationships among these brown algae based on a multigene method. These brown algae could be separated into two clades：Clade Ishigeales-Dictyotales and Clade Ectocarpales-Laminariales-Desmares-tiale-Fucales. The former was at the base of the phylogenetic tree, indicating its early divergence, while the latter was divided into two branches, with Order Fucales diverging from Orders Ectocarpales, Laminariales, and Desmarestiale. In our analysis of taxonomy-contentious species, Sargassum fusiforme and Saccharina sculpera were found to be closely related to genera Sargassum and Saccharina, respectively, while Petalonia fascia showed possible relation to genus Scytosiphon. The study provided molecular evidence for the phylo-genetic taxonomy of brown algae.

Decreased expression of HDAC8 indicates poor prognosis in patients with intrahepatic cholangiocarcinoma

Background: Intrahepatic cholangiocarcinoma (ICC) is a highly malignant primary tumor in the liver, and the rates of incidence and mortality are rapidly increasing globally. Histone deacetylase 8 (HDAC8) is a transcriptional regulator and is associated with tumorigenesis of several tumor types. This study aimed to evaluate the correlation between HDAC8 expression and clinicopathological parameters in ICC patients. Methods: ICC tissues and corresponding nonmalignant bile duct tissues were obtained from 60 patients. HDAC8 and Ki-67 expression were evaluated by immunohistochemistry staining. HDAC8 expression and the clinicopathological features and prognosis of the patients were analyzed. The mRNA level of HDAC8 in ICC was further analyzed using data from The Cancer Genome Atlas (TCGA). Results: The expression of HDAC8 were lower in ICC tissues (39/60, 65%) than in the corresponding nonmalignant bile duct tissues (54/60, 90%)( P = 0.001). Low HDAC8 expression in ICC was significantly associated with lymph node metastases (47.6% vs. 17.9%, P = 0.015). In addition, the positive cells rate of HDAC8 was statistically and negatively correlated with the Ki-67 index in ICC lesions ( r =?0.7660, P < 0.001). Importantly, the overall survival rate and recurrence-free survival rate in ICC patients with low HDAC8 expression were lower than those with high HDAC8 expression ( P = 0.008 and P = 0.011, respectively). Conclusions: Decreased HDAC8 expression in ICC is related to poor prognosis, and HDAC8 may be an independent prognostic indicator of ICC patients after curative resection.

Ester-based anti-freezing electrolyte achieving ultra-low temperature cycling for sodium-ion batteries

With the continuous advancement of industrialization,sodium-ion batteries(SIBs)need to operate in various challenging circumstances,particularly in extremely cold ***,at ultra-low tem-peratures,the reduced ionic conductivity and sluggish Na+migration of commonly carbonate-based elec-trolytes will inevitably lead to a sharp decrease in the capacity of ***,we design a carboxylate ester-based electrolyte with excellent ultra-low temperature performance by straightforward cosolvent *** to the low viscosity,melting point,and sufficient ionic conductivity of the designed elec-trolyte,the resulting Na||Na_(3)V_(2)(PO_(4))_(2)O_(2)F can achieve the capacity retention of 96%(100 cycles at 0.1 C)at-40℃ and can also operate stably even at-50℃.Besides,galvanostatic intermittent titration tech-nique(GITT),ex-situ X-ray photoelectron spectroscopy(XPS),and high-resolution transmission electron microscopy(TEM)tests are employed to analyze and confirm that the carboxylate ester-based electrolyte promotes robust and uniform cathode/electrolyte interface layer formation and accelerates ion diffusion kinetics,which collectively facilitates the better low-temperature *** addition,the assembled hard carbon||NVPOF full cells further prove the practicability of the carboxylate ester-based electrolyte at low-temperature,which delivers high discharge capacity of 108.4 and 73.0 mAh g^(-1) at-25 and-40℃.This work affords a new avenue for designing advanced low-temperature electrolytes for SIBs.

Iodoacetic acid(IAA) is an unregulated disinfection byproduct in drinking water and has been shown to exert cytotoxicity, genotoxicity, tumorigenicity, and reproductive and developmental toxicity. However, the effects of IAA on gut microbiota and its metabolism are still unknown, especially the association between gut microbiota and the metabolism and toxicity of IAA. In this study, female and male Sprague–Dawley rats were exposed to IAA at 0 and 16 mg/kg bw/day daily for 8 weeks by oral gavage. Results of 16S r RNA gene sequencing showed that IAA could alter the diversity, relative abundance and function of gut microbiota in female and male rats. IAA also increased the abundance of genes related to steroid hormone biosynthesis in the gut microbiota of male rats. Moreover, metabolomics profiling revealed that IAA could significantly disturb 6 and 13 metabolites in the feces of female and male rats, respectively. In female rats, the level of androstanediol increased in the IAA treatment group. These results were consistent with our previous findings, where IAA was identified as an androgen disruptor. Additionally, the perturbed gut microbiota and altered metabolites were correlated with each other. The results of this study indicated that IAA could disturb gut microbiota and its metabolism. These changes in gut microbiota and its metabolism were associated with the reproductive and developmental toxicity of IAA.
