Objective This study aimed to investigate the potential relationship between urinary metals copper(Cu),arsenic(As),strontium(Sr),barium(Ba),iron(Fe),lead(Pb)and manganese(Mn)and grip *** We used linear regression models,quantile g-computation and Bayesian kernel machine regression(BKMR)to assess the relationship between metals and grip *** In the multimetal linear regression,Cu(β=−2.119),As(β=−1.318),Sr(β=−2.480),Ba(β=0.781),Fe(β=1.130)and Mn(β=−0.404)were significantly correlated with grip strength(P1.007(95%confidence interval:−1.362,−0.652;P1)when each quartile of the mixture of the seven metals was *** kernel function regression model analysis showed that mixtures of the seven metals had a negative overall effect on grip strength,with Cu,As and Sr being negatively associated with grip strength *** the total population,potential interactions were observed between As and Mn and between Cu and Mn(P_(interactions) of 0.003 and 0.018,respectively).Conclusion In summary,this study suggests that combined exposure to metal mixtures is negatively associated with grip ***,Sr and As were negatively correlated with grip strength levels,and there were potential interactions between As and Mn and between Cu and Mn.

Fluoropyrimidine-based combination chemotherapy plus targeted therapy is the standard initial treatment for unresectable metastatic colorectal cancer(mCRC),but the prognosis remains *** phase 3 trial(***:NCT03950154)assessed the efficacy and adverse events(AEs)of the combination of PD-1 blockade-activated DC-CIK(PD1-T)cells with XELOX plus bevacizumab as a first-line therapy in patients with mCRC.A total of 202 participants were enrolled and randomly assigned in a 1:1 ratio to receive either first-line XELOX plus bevacizumab(the control group,n=1.2)or the same regimen plus autologous PD1-T cell immunotherapy(the immunotherapy group,n=100)every 21 days for up to 6 cycles,followed by maintenance treatment with capecitabine and *** main endpoint of the trial was progression-free survival(PFS).The median follow-up was 19.5 *** PFS was 14.8 months(95%CI,11.6-18.0)for the immunotherapy group compared with 9.9 months(8.0-11.8)for the control group(hazard ratio[HR],0.60[95%CI,0.40-0.88];p=0.009).Median overall survival(OS)was not reached for the immunotherapy group and 25.6 months(95%CI,18.3-32.8)for the control group(HR,0.57[95%CI,0.33-0.98];p=0.043).Grade 3 or higher AEs occurred in 20.0%of patients in the immunotherapy group and 23.5%in the control groups,with no toxicity-associated deaths *** addition of PD1-T cells to first-line XELOX plus bevacizumab demonstrates significant clinical improvement of PFS and OS with well tolerability in patients with previously untreated mCRC.

BACKGROUND Acute-on-chronic liver disease(AoCLD)accounts for the majority of patients hospitalized in the Department of Hepatology or Infectious *** To explore the characterization of AoCLD to provide theoretical guidance for the accurate diagnosis and prognosis of *** Patients with AoCLD from the Chinese Acute-on-Chronic Liver Failure(ACLF)study cohort were included in this *** clinical characteristics and outcomes,and the 90-d survival rate associated with each clinical type of AoCLD were analyzed,using the Kaplan-Meier method and the log-rank *** A total of 3375 patients with AoCLD were enrolled,including 1679(49.7%)patients with liver cirrhosis acute decompensation(LC-AD),850(25.2%)patients with ACLF,577(17.1%)patients with chronic hepatitis acute exacer-bation(CHAE),and 269(8.0%)patients with liver cirrhosis active phase(LC-A).The most common cause of chronic liver disease(CLD)was HBV infection(71.4%).The most common precipitants of AoCLD was bacterial infection(22.8%).The 90-d mortality rates of each clinical subtype of AoCLD were 43.4%(232/535)for type-C ACLF,36.0%(36/100)for type-B ACLF,27.0%(58/215)for type-A ACLF,9.0%(151/1679)for LC-AD,3.0%(8/269)for LC-A,and 1.2%(7/577)for *** HBV infection is the main cause of CLD,and bacterial infection is the main precipitant of *** most common clinical type of AoCLD is *** diagnosis and timely intervention are needed to reduce the mortality of patients with LC-AD or ACLF.

星点设计-响应曲面法优化IMP-RH60/mPEG-PLLA胶束的处方工艺及其体外评价

目的:通过星点设计-响应曲面法优化欧前胡素-聚氧乙烯氢化蓖麻油/甲氧基聚乙二醇聚乳酸[imperatorin micelles loaded with Cremophor?RH 60/monomethoxy poly(ethylene glycol)-block-poly(L-lactide),IMP-RH60/mPEG-PLLA]胶束的处方工艺。方法:在单因素考察的基础上,以RH60用量、药物与载体比例、水相体积为自变量,包封率和载药量为因变量,进行三因素三水平的星点设计-响应面法实验,分析结果得到IMP-RH60/mPEG-PLLA胶束的最优处方,并对胶束的释放和摄取进行评估。结果:确定最优处方工艺条件为:RH60的投药量为28 mg,IMP投药量为1.2 mg,mPEG-PLLA投药量为10 mg,水化体积为6 mL,在此条件下得到的胶束粒径为(25.48±0.92)nm,Zeta电位为-(5.50±1.06)mV,平均包封率为(85.78±0.84)%,平均载药量为(2.66±0.03)%。体外评价结果表明胶束释放效果优于原料药,并增强了药物的摄取。结论:星点设计-响应曲面法所建立的模型精密度高,可用于IMP-RH60/mPEG-PLLA胶束的处方工艺优化。

As an atomic emission spectroscopy technique, laser-induced breakdown spectroscopy (LIBS), first proposed by Breech and Cross in 1962 [1], has attracted much academic and industrial interest for its unique properties. By focusing a pulsed laser onto the surface of a target, laser-induced plasmas can be generated. The simplicity of sample preparation of the laser ablation process enables LIBS to directly analyze solid, liquid, or gaseous materials in the atmosphere, making it a convenient tool for chemical analyses. The unique features of LIBS such as requiring little or no sample preparation, allowing remote detection, and facilitating multiple elemental analyses have contributed to its rapid development in recent years.

Potential etiopathogenesis of seventh day syndrome following living donor liver transplantation:ischemia of the graft?

BACKGROUND: Seventh-day syndrome (7DS) is an early serious complication following liver transplantation with a high mortality because of its unknown etiopathogenesis. This study aimed to analyze the potential etiopathogenesis of 7DS. METHODS: A retrospective analysis of 98 consecutive living donor liver transplants performed from 2001 to 2007 at our center revealed that 5 patients had suffered from 7DS; their age, MELD score, portal vein inflow and other parameters were compared with those of the other recipients. RESULTS: The 5 patients showed common features: (a) initial uneventful recovery, and a dramatic rise of serum liver enzyme levels I to 2 weeks later; (b) decreased inflow in the portal vein accompanied by augmentation of serum creatinine and urea nitrogen level; and (c) serial liver biopsy findings of apoptosis and ischemic necrosis of hepatocytes. Four of the 5 patients died. Age, waiting time to transplantation, MELD score, operation time, cold ischemic time, portal interceptive time and diameter of the portal vein were not significantly different between the 2 groups, but a difference was found in the flow rate of the portal vein (t=3.234, P1). CONCluSIONS: The 5 patients suffered from a decreased portal vein inflow, ischemic liver necrosis, and renal failure. Hence, hypoperfusion of the liver graft was considered to be the etiopathogenesis of 7DS, for which, however sufficient evidence is lacking. More studies of 7DS are needed.

Experiments and sensitivity analyses for heat transfer in a meter-scale regularly fractured granite model with water flow

Experiments of saturated water flow and heat transfer were conducted for a meter-scale model of regularly fractured granite. The fractured rock model （height 1502.5 ram, width 904 mm, and thickness 300 mm）, embedded with two vertical and two horizontal fractures of pre-set apertures, was constructed using 18 pieces of intact granite. The granite was taken from a site currently being investigated for a high-level nuclear waste repository in China. The experiments involved different heat source temperatures and vertical water fluxes in the embedded fractures either open or filled with sand. A finite difference scheme and computer code for calculation of water flow and heat transfer in regularly fractured rocks was developed, verified against both the experimental data and calculations from the TOUGH2 code, and employed for parametric sensitivity analyses. The experiments revealed that, among other things, the temperature distribution was influenced by water flow in the fractures, especially the water flow in the vertical fracture adjacent to the heat source, and that the heat conduction between the neighboring rock blocks in the model with sand-filled fractures was enhanced by the sand, with larger range of influence of the heat source and longer time for approaching asymptotic steady-state than those of the model with open fractures. The temperatures from the experiments were in general slightly smaller than those from the numerical calculations, probably due to the fact that a certain amount of outward heat transfer at the model perimeter was unavoidable in the experiments. The parametric sensitivity analyses indicated that the tem- perature distribution was highly sensitive to water flow in the fractures, and the water temperature in the vertical fracture adjacent to the heat source was rather insensitive to water flow in other fractures.

The proton-proton(pp)fusion chain dominates the neutrino production in the *** uncertainty of the predicted pp neutrino flux is at the sub-percent level,whereas that of the best measurement is O(10%).In this study,for the first time,we measure solar pp neutrinos in the electron recoil energy range from 24 to 144 keV using the PandaX-4T commissioning data with 0.63 tonne×year *** pp neutrino flux is determined as(8.0±3.9(stat)±10.0(syst))×1010 s^(-1)cm^(-2),which is consistent with the Standard Solar Model and existing measurements,corresponding to an upper flux limit of 23.3×10^(10)s^(-1)cm^(-2)at 90%C.L..

研究焦棓酸对猪链球菌的体外抗菌活性及对生物被膜的抑制作用,并初步探讨其抑菌机制。采用96孔微量稀释法测定常用抗菌药物及焦棓酸对猪链球菌的最小抑菌浓度和最小杀菌浓度;平板活菌计数法和分光光度计测定焦棓酸对猪链球菌作用的时间-杀菌曲线;采用结晶紫染色法检测焦棓酸对猪链球菌生物被膜形成的影响;通过荧光定量PCR从转录水平分析焦棓酸对猪链球菌生长与生物被膜形成相关基因表达的情况。结果显示,猪链球菌2型HB菌株对磺胺异恶唑、磺胺甲恶唑耐药严重,焦棓酸对猪链球菌2型HB菌株的最小抑菌浓度为32μg/mL,最小杀菌浓度为64μg/mL,并且焦棓酸对猪链球菌的抑制作用呈剂量依赖性;焦棓酸在16μg/mL能显著抑制猪链球菌2型HB菌株生物被膜形成的能力;另外,经焦棓酸处理后,发现猪链球菌的生长、生物被膜形成、毒力相关基因ftsZ、stK、mrp、fbps、gapdh、ccpA、luxS下调表达。综上所述,焦棓酸可为耐药猪链球菌在临床上感染的治疗提供新思路。

Two new monosaccharide steroidal saponins,named ypsilandroside S(1)and ypsilandroside T(2),have been isolated from the whole plants of Ypsilandra *** structures were elucidated as heloniogenin 3-O-b-D-apiofuranoside(1)and pregna 5,16-dien-3b,12a-diol-20-one-3-O-b-D-apiofuranoside(2)by spectroscopic techniques(1D and 2D NMR,MS).Compounds 1 and 2 were tested for their inhibitory effects on lipopolysaccharide-induced nitric oxide production in RAW 264.7 cells.
