BACKGROUND Pulmonary lymphoepithelioma-like carcinoma(PLELC)is a rare type of nonsmall-cell lung *** lymphoepithelioma-like carcinoma(LELC)metastasis secondary to PLELC has not been reported *** SUMMARY A 64-year-old female was admitted to our hospital for a regular gastroscopy examination with a 6-year history of surgical resection for left *** emission tomography/computed tomography suggested high accumulation of 18F-fludeoxyglucose in the gastric cardia *** gastrointestinal endoscopy confirmed a large mass at the stomach ***(IHC)of the biopsy suggested metastatic stomach *** gastrectomy showed that this 6.5 cm×5.0 cm mass was located in the stomach fundus near the *** examination showed a poorly differentiated carcinoma with prominent lymphoplasmacytic *** demonstrated that the tumor was positive for CK(AE1/AE3),p63,p40,p53,Ki-67(70%),and EGFR(3+)and negative for CK7,CK20,Her2,and *** situ hybridization analysis showed positive staining Epstein-Barr virus-encoded *** programmed cell death ligand 1(PD-L1)expression score was 98%,and the combined positive score was 100,with no evidence of microsatellite ***,the patient was unequivocally diagnosed with metastatic stomach LELC secondary to pulmonary *** discharge,this patient underwent PD-1 inhibitor treatment(toripalimab,240 mg)every 3 wk for ten cycles,and she has had no tumor *** For gastric LELC metastasis,PD-1 inhibitor therapy could become a new therapeutic approach,though there is still no evidence from large data sets to support this.

Hepatic ischemia-reperfusion injury(HIRI)is the cause of postoperative hepatic dysfunction and failure,and even *** an important biological effector molecule,hydrogen sulfide(H_(2)S)of mitochondria as a gasotransmitter that is usually used to protect against acute HIRI ***,the exact relationship between HIRI and mitochondrial H_(2)S remains tangled due to the lack of an effective analytical ***,we have fabricated a mitochondria-targeted H_(2)S-activatable fluorogenic probe(Mito-GW)to explore the stability of mitochondrial H_(2)S and track the changes of mitochondrial H_(2)S during the *** virtue of pyridinium electropositivity and its amphiphilicity,Mito-GW could accumulate in *** goes through an analyte-prompted immolation when reacts with H_(2)S,resulting in the releasing of the fluorophore(GW).Therefore,the extent of Mito-GW conversion to GW can be used to evaluate the changes of mitochondrial H_(2)S level in living cells and *** proof-of-principle,we have used MitoGW to demonstrate the mitochondria H_(2)S-levels increase and then decrease during HIRI in vitro and in *** research highlights the tremendous potential of Mito-GW as a mitochondrial H_(2)S fluorogenic probe in elucidating the pathogenesis of HIRI,providing a powerful tool for promoting future research on hepatology.

The patterns of material accumulation in buildings and infrastructure accompanied by rapid urbanization offer an important,yet hitherto largely missing stock perspective for facilitating urban system engineering and informing urban resources,waste,and climate ***,our existing knowledge on the patterns of built environment stocks across and particularly within cities is limited,largely owing to the lack of sufficient high spatial resolution *** study leveraged multi-source big geodata,machine learning,and bottom-up stock accounting to characterize the built environment stocks of 50 cities in china at 500 m fine-grained *** per capita built environment stock of many cities(261 tonnes per capita on average)is close to that in western cities,despite considerable disparities across cities owing to their varying socioeconomic,geomorphology,and urban form *** is mainly owing to the construction boom and the building and infrastructure-driven economy of china in the past ***’s urban expansion tends to be more“vertical”(with high-rise buildings)than“horizontal”(with expanded road networks).It trades skylines for space,and reflects a concentration-dispersion-concentration pathway for spatialized built environment stocks development within cities in *** results shed light on future urbanization in developing cities,inform spatial planning,and support circular and low-carbon transitions in cities.

Background: Hepatic ischemia-reperfusion injury (HIRI) stands as an unavoidable complication arising from liver surgery, profoundly intertwined with its prognosis. The role of lysine methyltransferase SET domain bifurcated 1 (SETDB1) in HIRI remains elusive, despite its confirmation as a potential therapeutic target for diverse diseases. Here, we investigated the mechanism by which SETDB1 regulated HIRI. Methods: RNA sequencing data were used to identify the expression and potential targets of SETDB1 through bioinformatics analysis. To elucidate the impact of SETDB1 on HIRI, both an in vivo model of HIRI in mice and an in vitro model of hepatocyte hypoxia/reoxygenation were established. Biochemical and histological analyses were used to investigate the influence of SETDB1 on liver damage mediated by HIRI. Chromatin immunoprecipitation and coimmunoprecipitation were implemented to explore the in-depth mechanism of SETDB1 regulating HIRI. Results: We confirmed that hepatocellular SETDB1 was up-regulated during HIRI and had a close correlation with HIRI-related inflammation and apoptosis. Moreover, inhibition of SETDB1 could mitigate HIRI-induced liver damage, inflammation, and apoptosis. Through our comprehensive mechanistic investigation, we revealed that SETDB1 interacts with apoptosis-signal-regulating kinase 1 (ASK1) and facilitates the methylation of its lysine residues. Inhibition of SETDB1 resulted in reduced phosphorylation of ASK1, leading to a marked suppression of downstream c-Jun N-terminal kinase (JNK)/p38 signaling pathway activation. The therapeutic effect on inflammation and apoptosis achieved through SETDB1 inhibition was nullified by the restoration of JNK/p38 signaling activation through ASK1 overexpression. Conclusions: The findings from our study indicate that SETDB1 mediates lysine methylation of ASK1 and modulates the activation of the ASK1–JNK/p38 pathway, thus involved in HIRI-induced inflammation and apoptosis. Colorectal cancer(CRC)is the second leading cause of cancer mortality *** initial diagnosis,approximately 20%of patients are diagnosed with metastatic CRC(mCRC).Although the APC-Asef interaction is a well-established target for mCRC therapy,the discovery and development of effective and safe drugs for mCRC patients remains an urgent and challenging *** this study,we identified a novel structural scaffold based on MAI inhibitors,the first-in-class APC-Asef inhibitors we reported *** model-driven optimizations of the N-terminal cap and experimental evaluations of inhibitory activity were performed,and 24-fold greater potency was obtained with the best inhibitor compared to the parental *** addition,the cocrystal structure validated that the two-layerπ-πstacking interactions were essential for inhibitor stabilization in the bound ***,in vitro and in vivo studies have demonstrated that novel inhibitors suppressed lung metastasis in CRC by disrupting the APC-Asef *** results provide an intrinsic structural basis to further explore drug-like molecules for APC-Asef-mediated CRC therapy.

Carbendazim,a widely used fungicide in china,has been found to have reproductive toxicity and mutagenic ***,information on the spatial-temporal variations of carbendazim residues in food in china is ***,we investigated the presence of carbendazim in china's plant-based foods from 2011 to 2020,evaluated the spatial-temporal characteristics,and assessed the associated exposure risks in the Chinese *** results revealed a high detection frequency of carbendazim in fruits(26.4%)and high concentrations in vegetables(~110 mg kg^(-1)),indicating widespread misuse of the *** acute risks of consuming certain vegetables and cereals exceeded the recommended limits by up to 12 and 5 times,*** there has been a decline in carbendazim residue levels in food since the implementation of the Chinese government's action plan for zero growth of pesticide use in 2015,some provinces still exhibited high levels of carbendazim in multiple food categories,which were positively correlated with annual pesticide *** highlight that carbendazim contamination reflects the broader issue of pesticide use in *** emphasizes the need for committed and targeted national policies to reduce carbendazim residues in food and suggests that such measures could also regulate the use of other pesticides,given that pesticide abuse in china is not limited to specific *** call for the re-evaluation of maximum residue limits of carbendazim,particularly in highly consumed foods such as cereals.

AIM:To repor t the clinical profile,endoscopic dacryocystorhinostomy(En-DCR)management,and acute dacryocystitis(AD)outcomes in ***:Clinical data of 554 adult AD patients(554 eyes)who presented in 7 tertiary eye care centers for 10y from Jan 2010 to Mar 2020 were retrospectively *** profile,En-DCR management,and outcomes of all cases were *** anatomical and functional success were evaluated for 12mo ***:The analysis included 149 males and 368 females with a median age of 55.2y(range:18-84y).There were 459 eyes with a history of epiphora or purulent *** time between a symptom of lacrimal duct obstruction and acute onset was 1 to 540(66.1±58.2)***-nine eyes had a history of the previous acute ***-four eyes developed a cutaneous fistula,while 11 eyes had post septal cellulitis ***-DCR with an anatomical success of 91.7%and functional success of 90.1%.The success rate of the patients with a history of acute episodes and the preoperative fistula was lower than the overall success ***:En-DCR can be performed during an acute episode in AD with a success rate of over 90%.
