Leukocyte cell-derived chemotaxin 2 inhibits development of atherosclerosis in mice

Leukocyte cell-derived chemotaxin 2 (LECT2), a multifunctional hepatokine, is involved in many pathological conditi ons. However, its role in atherosclerosis remains undefined. In this study, we admimistered vehicle or LECT2 to male Apoe^-/- mice fed a Western diet for 15 weeks. Atherosclerotic lesions were visualized and quantified with Oil-red O and hematoxylin staining. The mRNA expression levels of MCP-1, MMP-1, IL-8 IL-1β, and TNF-a were analyzed by quantitative real-time polymerase chain reaction. Serum TNF-a, IL-1β, IL-8, MCP-1, and MMP-1 concentrations were measured by en zyme-li nked immuno sorbent assay. CD68, CD31, and a-SMA, markers of macrophages, endothelial cells, and smooth muscle cells, respectively, were detected by immuno staining. Results showed that LECT2 reduced total cholesterol and low-density lipoprotein concentrations in serum and inhibited the development of atherosclerotic lesions, accompanied by reductions in inflammatory cytokines and lower MCP-1, MMP-1, TNF-a, IL-8, and IL-1β mRNA abundanee. Furthermore, LECT2 decreased CD68, but in creased cr SMA in atherosclerotic lesi ons, suggesting an in crease in smooth muscle cells and reduction in macrophages. In summary, LECT2 inhibited the development of atherosclerosis in mice, accompanied by reduced serum total cholesterol concentration and lower inflammatory responses.

Site selection of self-driving and recreational vehicle camps in China:An investigation using analytic hierarchy process and entropy

Self-driving and recreational vehicle(RV)camps are a new form of industry module with the integration of transportation and tourism in China,thus the scientific and reasonable site selection plays an important role in the success of camps’construction and *** terms of relying resources and development factors,camps can be divided into five categories:scenic-spot-based,transportation-based,environment-based,project-based and leisure and *** to whether it is of excludability and competitiveness,the camps in China mainly embody the attribute of private *** on the combination of subjective evaluation and objective calculation,the evaluation model of spatial site selection is constructed and the weight of each index is calculated by using analytic hierarchy process(AHP)method and entropy coefficient *** results show that traffic condition factor is the priority to the selection of campsite,and whether it is on the popular main self-driving route and the grade of trunk roads are the dominant *** second factor taken into consideration is the social factors,in which government policy supports and land cost play a key *** third factor is the market,in which the urban economic level,partnership with the government and tourist resource conditionsare of great *** fourth factor of the campsite selection includes natural elements,in which the quality of ecological environment and water source conditions are mostly *** the future,it is suggested that a camp pattern of"public goods"plus"private goods"should be built and the construction of camps in underdeveloped areas should be highly developed so as to form camp spatial network from individual points to a series of campsite and finally the campsite group in China will be set up.

Sob gene is critical to wing development in Bombyx mori and Tribolium castaneum

The development of insect appendages requires the expression of multiple genes in a strict spatial and temporal order. The odd-skipped family genes are vital transcriptional factors involved in embryonic development. The development and morphogenesis of the insect wing requires multiple transcription factors to regulate the expression of wing patterning genes at the transcriptional level. However, the function of odd-related genes in insect wing morphogenesis and development during postembryonic stages is unclear. We focused on the roles of the sister of odd and bowl (sob) gene, a member of odd-skipped family genes, during the wing morphopoiesis in Bombyx mori using the clustered regularly interspaced palindromic repeats (CRISPR)/CRISPR-associated protein 9 system and in Tribolium castaneum by RNA interference. The results showed that the wings were significantly smaller and degenerated, and wing veins were indistinct in the sob gene loss-of-function group in both B. mori and T. castaneum. Quantitative real-time polymerase chain reaction revealed that the Tcsob gene regulated the expression of wing development genes, such as the cht 7 and the vg gene. The findings suggest the importance of sob gene in insect wing morphology formation during postembryonic stages.

Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

In-situ measurement via the flow-through method and numerical simulations for radon exhalation during measurements of the radon exhalation rate

Small-scale measurements of the radon exhalation rate using the flow-through and closed-loop methods were conducted on the surface of a uranium tailing pond to better understand the differences between the two *** abnormal radon exhalation behavior was observed,leading to computational fluid dynamics(CFD)-based simulations in which dynamic radon migration in a porous medium and accumulation chamber was *** on the in-situ experimental and numerical simulation results,variations in the radon exhalation rate subject to permeability,flow rate,and insertion depth were quantified and *** in-situ radon exhalation rates measured using the flow-through method were higher than those measured using the closed-loop method,which could be explained by the negative pressure difference between the inside and outside of the chamber during the *** consistency of the variations in the radon exhalation rate between the experiments and simulations suggests the reliability of CFD-based techniques in obtaining the dynamic evolution of transient radon exhalation rates for diffusion and convection at the porous medium-air *** synergistic effects of the three factors(insertion depth,flow rate,and permeability)on the negative pressure difference and measured exhalation rate were quantified,and multivariate regression models were established,with positive correlations in most cases;the exhalation rate decreased with increasing insertion depth at a permeability of 1×10^(−11) m^(2).CFD-based simulations can provide theoretical guidance for improving the flow-through method and thus achieve accurate measurements.

Timely Policy Exit:Reducing Over-Investment and Driving High-Quality Firm Performance

Using data from the 11th to 14th Five-Year Plan periods(2006-2025),this study applies a Difference-in-Differences(DID)approach to assess the impact of industrial policy *** that have faced policy withdrawal for over a decade are categorized as the treatment group,while consistently supported industries form the control *** analysis examines how withdrawal affects firm total factor productivity(TFP)and investment *** results show that policy withdrawal boosts firm TFP by reducing over-investment and improving the efficiency of R&D *** effect is particularly evident in industries with strong,competitive leading ***,in regions with lower levels of marketization,timely policy withdrawal plays a key role in curbing *** study also highlights a dual effect of policy withdrawal:while it fosters corporate social responsibility,it may also encourage financial *** findings suggest that the implementation of industrial policy should provide“timely assistance”over a limited timeframe rather than long-term support to well-established *** industries mature,policy support should be gradually reduced or phased out to avoid over-investment and enhance firm efficiency.
