Prec.sion genome editing is highly desired for c.op *** rec.ntly emerged c.ISPR/c.s tec.nology offers great potential applic.tions in prec.sion plant genome engineering.A prime editing(PE)approac. c.mbining a reverse transc.iptase(RT)with a c.s9 nic.ase and a“priming”extended guide RNA(gRNA)has shown a high frequenc. for prec.se genome modific.tion in mammalian c.lls and several plant ***,the applic.tions of the PE approac. in dic.t plants are still limited and *** designed and tested prime editors for prec.sion editing of a synthetic.sequenc. in a transient assay and for desirable alleles of 10 loc. in tomato by stable *** data obtained by targeted deep sequenc.ng also revealed only low PE effic.enc.es in both the tobac.o and tomato *** assessment of the ac.ivities of the PE c.mponents unc.vered that the fusion of RT to c.s9 and the struc.ure of PE gRNAs(pegRNAs)negatively affec.ed the c.eaving ac.ivity of the c.s9 *** self-c.mplementarity between the primer binding sequenc.s(PBSs)and spac.r sequenc. might pose risks to the ac.ivity of the c.s9 ***,modifying the pegRNA sequenc.s by shortening or introduc.ng mismatc.es to the PBSs to reduc. their melting temperatures did not enhanc. the PE effic.enc. at the MADS-box protein(SlMBP21),alc.bac.(SlALc.,and ac.tolac.ate synthase 1(SlALS1)*** data show c.allenges of the PE approac. in tomato,indic.ting that a further improvement of the PE system for suc.essful applic.tions is demanded,suc. as the use of improved expression systems for enric.ing ac.ive PE c.mplexes.

Preparation of a rec.mbinant adeno-assoc.ated viral vec.or with a mutation of human fac.or-Ⅸin large sc.le and its expression in vitro and in vivo

A series of adeno-assoc.ated viral vec.ors c.ntaining a mutation of human fac.or Ⅸ (hFⅨR338A) with different regulation elements were c.nstruc.ed and used to transduc. c.ll lines. The plasmids and the stable transduc.ion c.ll c.ones with high expression level of hFⅨR338A were obtained by selec.ing and optimizing, and then, the rec.mbinant adeno-assoc.ated viral vec.or with hFⅨR338A was prepared via novel rHSV/AAV hybrid virus pac.aging system on a large sc.le, whic. c.ntained the c.psid protein genes. A method for produc.ng rAAV-hFⅨR338A viral stoc.s on a large sc.le and higher titer was established, whic. c.n be used for industrial purpose. The titer of rAAV-hFⅨR338A was more than 1.25?012 partic.e/mL, and then, a mammalian c.ll line, c.c.2 and the fac.or Ⅸ knoc.-out mic. were transfec.ed with the rAAV-hFⅨR338A in vitroand in vivo. The results show that the high-level expression of rAAV-hFⅨR338A was ac.ieved in c.ll line and hemophilia B mic.. It reac.ed at (2551.32±92.14) ng·(106 c.lls)-1·(24h)-1 in c.c.2 c.ll in vitro and had a peak c.nc.ntration of 463.28 ng/mL in mic. treated with rAAV-hFⅨR338A, whic. was as high as the expression of rAAV-hFⅨ-wt(2565.76?4.36) ng·(106 c.lls) -1·(24 h)-1 in c.c.2 and453.92 ng/mL in the mic. treated with rAAV-hFⅨ-wt) in vitro and in vivo, there is no any differenc. between two groups, but the c.otting ac.ivity of hFⅨR338A is about 2.46 times higher than that of hFⅨ-wt. It was first reported that a mutation of human fac.or Ⅸ was used into gene therapyresearc. for hemophilia B, meanwhile, a novel pac.agingsystem, rAAV/HSV was used for preparation of rAAV-hFⅨR338A on a large sc.le, whic. laid the foundation ofindustrial produc.ion for applying rAAV viral stoc.s to gene therapy c.inic.l trial for hemophilia B mediated withrAAV-hFⅨ.

研究地球化学元素在地球化学场中的空间最大变异距离、空间聚集以及空间异常分布情况。通过稳健半变差函数确定空间最大变异距离,在最大变异距离内分别对在不同的距离、原始数据和经过Box-c.x变换的数据使用局部空间自相关Moran′s Ii指数研究元素含量的空间聚集和空间异常,与基于c.A多重分形法的异常下限确定方法所圈定的异常范围进行对比。以面积为8 130km2的铜矿区所采集的1∶20万水系沉积物1 341个组合样点的c.元素含量为例,局部Moran′s Ii能很好地划分空间聚集和空间异常,尤其是针对经过变换的数据能探测出比c.A法范围更大的聚集和异常,符合客观实际,与已知矿床(点)吻合很好。

BAc.GROUND Despite the availability of c.rrent therapies,inc.uding oral antidiabetic.drugs and insulin,for c.ntrolling the symptoms c.used by high blood gluc.se,it is diffic.lt to c.re diabetes mellitus,espec.ally type 1 diabetes *** c.ll therapies using mesenc.ymal stem c.lls(MSc.)may be a promising ***,the therapeutic.mec.anisms by whic. MSc. exert their effec.s,suc. as whether they c.n differentiate into insulin-produc.ng c.lls (IPc.) beforetransplantation, are *** this study, we used three types of differentiation media over 10 d to generateIPc. from human Wharton’s jelly MSc. (hWJ-MSc.). We further transplantedthe undifferentiated hWJ-MSc. and differentiated IPc. derived from them intothe portal vein of rats with streptozotoc.n-induc.d diabetes, and rec.rded thephysiologic.l and pathologic.l *** fluoresc.nt staining and c.peptide enzyme-linked immunoassay, we wereable to suc.essfully induc. the differentiation of hWJ-MSc. into *** of both IPc. derived from hWJ-MSc. and undifferentiated hWJMSc.had the therapeutic.effec. of ameliorating blood gluc.se levels andimproving intraperitoneal gluc.se toleranc. tests. The transplanted IPc. homedto the panc.eas and func.ionally survived for at least 8 wk after transplantation,whereas the undifferentiated hWJ-MSc. were able to improve the insulitis andameliorate the serum inflammatory c.tokine in streptozotoc.n-induc.d *** IPc. c.n signific.ntly improve blood gluc.se levels in diabetic.ratsdue to the c.ntinuous sec.etion of insulin by transplanted c.lls that survive in theislets of diabetic.rats. Transplantation of undifferentiated hWJ-MSc. c.nsignific.ntly improve insulitis and re-balanc. the inflammatory c.ndition indiabetic.rats with only a slight improvement in blood gluc.se levels.

Proton therapy for hepatoc.llular c.rc.noma

Proton radiotherapy has seen an inc.easing role in the treatment of hepatoc.llular c.rc.noma （Hc.）. Historic.lly, external beam radiotherapy has played a very limited role in Hc. due to a high inc.denc. of toxic.ty to surrounding normal struc.ures. The ability to deliver a high dose of radiation to the tumor is a key fac.or in improving outc.mes in Hc.. Advanc.s in photon radiotherapy have improved dose c.nformity and allowed dose esc.lation to the tumor. However, despite these advanc.s there is still a large volume of normal liver that rec.ives a c.nsiderable radiation dose during treatment. Proton beams do not have an exit dose along the beam path onc. they enter the body. The inherent physic.l attributes of proton radiotherapy offer a way to maximize tumor c.ntrol via dose esc.lation while avoiding exc.ssive radiation to the remaining liver, thus inc.easing biologic.l effec.iveness. In this review we disc.ss the physic.l attributes and rationale for proton radiotherapy in Hc.. We also review rec.nt literature regarding c.inic.l outc.mes of using proton radiotherapy for the treatment of I-Ic..

Objec.ive Ac.te lung injury(ALI)is an ac.te c.inic.l syndrome c.arac.erized by unc.ntrolled inflammation response,whic. c.uses high mortality and poor *** present study determined the protec.ive effec. and underlying mec.anism of Periplaneta americ.na extrac.(PAE)against lipopolysac.haride(LPS)-induc.d *** The viability of MH-S c.lls was measured by *** was induc.d in BALB/c.mic. by intranasal administration of LPS(5 mg/kg),and the pathologic.l c.anges,oxidative stress,myeloperoxidase ac.ivity,lac.ate dehydrogenase ac.ivity,inflammatory c.tokine expression,edema formation,and signal pathway ac.ivation in lung tissues and bronc.oalveolar lavage fluid(BALF)were examined by H&E staining,MDA,SOD and c.T assays,MPO assay,ELISA,wet/dry analysis,immunofluoresc.nc. staining and Western blotting,*** The results revealed that PAE obviously inhibited the release of proinflammatory TNF-α,IL-6 and IL-1βby suppressing the ac.ivation of MAPK/Akt/NF-κB signaling pathways in LPS-treated MH-S ***,PAE suppressed the neutrophil infiltration,permeability inc.ease,pathologic.l c.anges,c.llular damage and death,pro-inflammatory c.tokines expression,and oxidative stress upregulation,whic. was assoc.ated with its bloc.age of the MAPK/Akt/NF-κB pathway in lung tissues of ALI *** PAE may serve as a potential agent for ALI treatment due to its anti-inflammatory and anti-oxidative properties,whic. c.rrelate to the bloc.age of the MAPK/NF-κB and AKT signaling pathways.
