Metabolic syndrome and colorectal neoplasms:An ominous association

Background: Leprosy is a chronic infectious disease classified into two subgroups for therapeutic purposes:paucibacillary(PB)and multibacillary(MB),closely related to the host immune *** this context it is noteworthy looking for immunological biomarkers applicable as complementary diagnostic tools as well as a laboratorial strategy to follow-up leprosy household ***:: the cross-sectional study enrolled 49 participants,including 19 patients and 30 healthy *** blood mononuclear cells(PBMC)were isolated and incubated in the presence of Mycobacterium leprae *** cells were prepared for surface(CD4+and CD8+)and intracytoplasmic cytokine staining(IFN-γ,IL-4 and IL-10).Multiple comparisons amongst groups were carried out by ANOVA,Kruskal–Wallis,Student t or Mann–Whitney *** analysis of categorical variables was performed by *** biomarker signature analysis was conducted using the global median values for each biomarker index as the cut-off edge to identify the proportion of subjects with high biomarker ***: the cytokine signature analysis demonstrated that leprosy patients presented a polyfunctional profile of t-cells subsets,with increased frequency of IFN-γ+t-cell subsets along with IL-10+and IL-4+from CD4+t-cells,as compared to health Controls(Venn diagram report).Moreover,statistical analysis was carried out using parametric or non-parametric variance analysis followed by pairwise multiple comparisons,according to the data normality distribution.L(PB)displayed a polyfunctional profile characterized by enhanced percentage of IFN-γ+,IL-10+and IL-4+produced by most t-cell subsets,as compared to L(MB)that presented a more restricted cytokine functional profile mediated by IL-10+and IL-4+t-cells with minor contribution of IFN-γproduced by CD4+*** was that HHC(MB)exhibited enhanced frequency of IFN-γ+t-cells,contrasting with HHC(PB)that presented a cytokine profile limited t

Short stature is a clinical challenge in the daily practice of pediatric endocrinology, regarding the several technical, cultural and economic factors associated with its approach. this article intends to review the physiology of growth hormone secretion, the endocrine regulation of human growth and the clinical aspects of the diagnosis and treatment of short stature. It specifically analyses the treatment of short stature with growth hormone, along with its side effects, cost/benefit analysis and possible risks. A clinical case from a medical school is also described, intending a better understanding of this frequent ambulatory situation in endocrinology and pediatrics.

脑室下区(subventricular zone,SVZ)及海马齿状回颗粒下区(subgranular zone,SGZ)是哺乳动物脑内终生存在神经发生的两个主要区域。SVZ产生的多巴胺能神经元经吻侧迁移流(rostral migratory stream,RMS)迁移至嗅球(olfactory bulb,OB)并分化为颗粒细胞层(granular cell layer,GCl)、僧帽细胞层(mitral cell layer,MCl)、外从状层(external plexiform layer,EPl)、突触球层(glomerular layer,Gl)等内的中间神经元;SGZ产生的神经元则迁移至海马齿状回颗粒层成为颗粒细胞。这些细胞能整合入神经网络并发挥功能。已知a-synuclein(SNCA)编码突触核蛋白SNCA,能影响SVZ神经发生中的多巴胺能性神经传递及嗅球的分化。研究表明,SNCA在人类帕金森病等多巴胺能神经退行性疾病的发生发展中扮演着关键角色。现已在家族性帕金森病病例中发现了SNCA的3个点突变:A53t(丙氨酸53-苏氨酸)、A30P(丙氨酸30-脯氨酸)、E46K(谷氨酸46-赖氨酸)。但有关SNCA及其突变对成体SVZ神经发生及嗅球分化的确切作用尚不清楚。本研究采用了携带人SNCA顺式调控元件的细菌人工染色体转基因小鼠(遗传背景均为C57BL),设正常对照(C57BL)、SNCA敲除组(SNCA-/-)及SNCA敲除并表达人SNCAA30P突变(A30PSNCA-/-)3个组,用免疫荧光组织化学方法分析了3组间SVZ神经细胞增殖及嗅球细胞分化的差异。结果显示,①在SVZ内,磷酸化组蛋白3(PH3)阳性的增殖细胞在SNCA-/-鼠明显减少,在A30P SNCA-/-鼠显著减少;②Mash1+细胞在A30PSNCA-/-鼠SVZ内显著减少;③SNCA-/-及SNCAA30P突变致嗅球中心及突触球层(Gl)内的双皮质素Dcx+细胞显著减少;④SNCA-/-及SNCA A30P突变致嗅球Gl内的tH+、Calbindin(CalB)+、Calretinin(CalR)+细胞显著减少;⑤tunel染色显示,SNCA-/-及A30P SNCA-/-组小鼠嗅球中的细胞凋亡数减少。上述结果提示,SNCA A30P突变减弱了成体SVZ内的神经发生,迟滞了SVZ增殖神经细胞向嗅球的迁移并迟缓了这些迁移细胞向嗅球中间神元的分化。

the bipartite begomoviruses (Geminiviridae family), which are DNA viruses that replicate in the nucleus of infected cells, encode the nuclear shuttle protein (NSP) to facilitate the translocation of viral DNA from the nucleus to the cytoplasm via nuclear pores. this intracellular trafficking of NSP-DNA complexes is accessorized by the NSP-interacting guanosine triphosphatase (NIG) at the cytosolic side. Here, we report the nuclear redistribution of NIG by AtWWPI, a WW domain-containing protein that forms immune nuclear bodies (NBs) against begomoviruses. We demonstrated that AtWWPI relocates NIG from the cytoplasm to the nucleus where it is confined to AtWWP1-NBs, suggesting that the NIG-AtWWP1 interaction may interfere with the NIG pro-viral function associated with its cytosolic localization. Consistent with this assumpti on joss AtWWPI functi on cuased plants more susceptible to begomovirus infection, whereas overexpression of AWWP1 enhaneed plant resistance to begomovirus. Furthermore, we found that a mutant versi on of AtWWPI defective for NB formatio n was noIonger capable of interacti ng with and relocating NIG to the nucleus and lost its immune function against begomovirus. the antiviral function of AtWWP1-NBs, however, could be antagonized by viral infection that induced either the disruption or a decrease in the number of AtWWP1-NBs. Collectively, these results led us to propose that AtWWPI organizes nuclear structures into nuclear foci, which provide intrinsic immunity against begomovirus infection.

Despite the precise controllability of droplet samples in digital microfluidic(DMF)systems,their capability in isolating single cells for long-time culture is still limited:typically,only a few cells can be captured on an *** fabricating small-sized hydrophilic micropatches on an electrode aids single-cell capture,the actuation voltage for droplet transportation has to be significantly raised,resulting in a shorter lifetime for the DMF chip and a larger risk of damaging the *** this work,a DMF system with 3D microstructures engineered on-chip is proposed to form semi-closed micro-wells for efficient single-cell isolation and long-time *** optimum results showed that approximately 20%of the micro-wells over a 30×30 array were occupied by isolated single *** addition,low-evaporation-temperature oil and surfactant aided the system in achieving a low droplet actuation voltage of 36V,which was 4 times lower than the typical 150 V,minimizing the potential damage to the cells in the droplets and to the DMF *** exemplify the technological advances,drug sensitivity tests were run in our DMF system to investigate the cell response of breast cancer cells(MDA-MB-231)and breast normal cells(MCF-10A)to a widely used chemotherapeutic drug,Cisplatin(Cis).the results on-chip were consistent with those screened in conventional 96-well *** novel,simple and robust single-cell trapping method has great potential in biological research at the single cell level.

Freshwater biota are more comprehensive and direct indicators of biological impacts,and more meaningful to the public than water quality or physical habitat *** biotic data and the multiple biological indicators developed from them offer a much richer array of data for assessing the impacts of pollution controls than a limited set of physical or chemical *** recent decades,assemblage-based assessments by ecologists,environmental scientists,and water quality agencies have been employed globally for determining the condition of,and threats to,freshwater ecosystems.A key step in this advance has been the development of multimetric indices(MMIs)or indices of biotic integrity(IBIs)based on quantitative assessments of algae,macrophyte,macroinvertebrate,fish or riparian bird *** Europe,where biological assemblages are mandated for assessing freshwater ecosystem health,many indices are ***,the proliferation of MMIs globally has not always occurred through the application of rigorous study designs and monitoring protocols,nor have they always effectively incorporated functional metrics,stressor assessments,and statistical ***,in this review,we discuss eleven major concerns with the development and application(including logistical limitations)of multimetric indicators based on freshwater biota to encourage more rigorous and widely applicable(transferable)MMI use and ***,our concerns focus on reference conditions;sampling effort,methods,and season;trophic guild definition;metric comprehensiveness,options,screening and scoring;and MMI *** could also benefit from increased attention to ecological mechanisms and metric development,to further improve our understanding of anthropogenic impacts as well as rehabilitation effects on freshwater ecosystems *** closer attention to study designs,ecological mechanisms and metric development should further improve our understa

Parathyroid ultrasonography and bone metabolic profile of patients on dialysis with hyperparathyroidism

AIM to evaluate the parathyroid ultrasonography and define parameters that can predict poor response to treatment in patients with secondary hyperparathyroidism due to renal *** this cohort study evaluated 85 patients with chronic kidney disease stage V with parathyroid hormone levels above 800 pg/mL. All patients underwent ultrasonography of the parathyroids and the following parameters were analyzed: Demographic characteristics(etiology of chronic kidney disease, gender, age, dialysis vintage, vascular access, use of vitamin D), laboratory(calcium, phosphorus, parathyroid hormone, alkaline phosphatase, bone alkaline phosphatase), and the occurrence of bone changes, cardiovascular events and death. the χ~2 test were used to compare proportions or the Fisher exact test for small sample frequencies. Student t-test was used to detect differences between the two groups regarding continuous *** Fifty-three patients(66.4%) had parathyroid nodules with higher levels of parathyroid hormone, calcium and phosphorus. Sixteen patients underwent parathyroidectomy and had higher levels of phosphorus and calcium × phosphorus product(P = 0.03 and P = 0.006, respectively). they also had lower mortality(32% vs 68%, P = 0.01) and lower incidence of cardiovascular or cerebrovascular events(27% vs 73%, P = 0.02). Calcium × phosphorus product above 55 mg^2/dL^2 [RR 1.48(1.06, 2.08), P = 0.03], presence of vascular calcification [1.33(1.01, 1.76), P = 0.015] and previous occurrence of vascular events [RR 2.25(1.27, 3.98), P tors for mortality in this population. there was no association between the occurrence of nodules and *** the identification of nodules at ultrasonography strengthens the indication for parathyroidectomy in patients with secondary hyperparathyroidism due to renal failure.

this study investigates the biofilm formation, presence and distribution of virulence genes and the capacity to induce an inflammatory response in strains of Staphylococcus aureus isolated from milk samples in Bahia, Brazil. A total of 132 samples of raw milk were collected from four dairy farms (designated A to D) located in southwestern Bahia, in the municipality of Vitória da Conquista, from October/2009 to September/2010. After processing of the samples, 94 (71.2%) S. aureus isolates were obtained. these strains were subjected to the antibiogram method MIC (Minimum Inhibitory Concentration). As for the pathogenicity, tests were performedin vitrobiofilm formation induced by glucose. Moreover, we performed PCR for their virulence genes: sea (enterotoxin A), seb (B), sec (C), pvl (Panton-Valentine Leukocidin), clfA (Clumping Factor A) and spa (protein A) and analysis of cytokine induction in the inflammatory response of J774 macrophages by exocellular lipoteichoic acid. No isolates were resistant to oxacillin and vancomycin. In biofilm production, 5.31% (5/94) isolates did not produce biofilm, 5.31% (5/94) of the samples were poor producers, 15.96% (15/94) strains were moderate producers, 18.09% (17/94) were producers and 55.32% (55/94) of isolates were strong biofilm producers. One (1.06%) isolate expressed the seb gene, one (1.06%) sec, 18 (19.2%) cflA and 44 (46.8%) had spa. there was no expression of sea and pvl between isolates analyzed. the analysis of cytokine induction in the inflammatory response did not show statistical difference in the levels of IL-6, tNF-α and IL-10 induction. However, there was statistical difference in IL-1 induction between isolates from different farms. thus, it appears that the results obtained in this study show significant effects for the region studied, since it is an important dairy region, hence the need for further studies, with the intent of attracting funding that contributes to improving prevention and control in both dai
