Modulation of signal transduction pathways by natural compounds in cancer

Cancer is generally regarded as the result of abnormal growth of cells. According to World Health Organization, cancer is the leading cause of mortality worldwide. Mother nature provides a large source of bioactive compounds with excellent therapeutic efficacy. Numerous phytochemicals from nature have been investigated for anticancer properties. In this review article, we discuss several natural compounds, which have shown anti-cancer activity. Natural compounds induce cell cycle arrest, activate intrinsic and extrinsic apoptosis pathways, generate Reactive Oxygen Species(ROS), and down-regulate activated signaling pathways, resulting in inhibition of cell proliferation, progression and metastasis of cancer. Several preclinical studies have suggested that natural compounds can also increase the sensitivity of resistant cancers to available chemotherapy agents. Furthermore, combining FDA approved anti-cancer drugs with natural compounds results in improved efficacy. On the basis of these exciting outcomes of natural compounds against several cancer types, several agents have already advanced to clinical trials. In conclusion, preclinical results and clinical outcomes against cancer suggest promising anticancer efficacy of agents from natural sources.

AIM: To assess the clinical significance of incidental focal colorectal 18 F-fluorodeoxyglucose ( 18 F-FDG) uptake on 18 F-FDG-positron emission tomography/computed tomography (PET/CT). METHODS: The records of all the cases which had undergone colonoscopy after PET/CT within a two weeks interval were reviewed. Adenomas were considered advanced when they were villous, ≥ 10 mm in size, or had high-grade dysplasia. Colorectal cancers and advanced adenomas are collectively referred to as advanced colorectal neoplasms. Receiver-operating characteristic curve analysis was used to determine thesignificant predictive maximum standardized uptake value (SUVmax) cutoff value for advanced colorectal neoplasms and cancer. RESULTS: Ninety-five colorectal lesions matched the site of incidental focal colorectal 18 F-FDG uptake on PET/CT and 146 did not. Colonoscopy showed advanced colorectal neoplasms corresponding to the site of 18 F-FDG uptake in 49 of the 95 (51.5%) lesions with incidental uptake. Of the lesions without incidental uptake, only 6 of 146 (4.1%) had advanced colorectal neoplasms on colonoscopy, indicating a statistically significant difference between the two groups (P < 0.001). The sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy of incidental focal 18 F-FDG uptake in identifying advanced colorectal neoplasms were 89.1%, 75.3%, 51.6%, 95.9%, and 78.4%, respectively. In detecting only CRC, these values were 89.2%, 69.6%, 34.7%, 97.3%, and 72.6%, respectively. The significant SUVmax cutoff value for advanced colorectal neoplasms (area under the curve 0.755, P < 0.001) was 4.35, with a sensitivity, specificity, PPV, NPV, and accuracy of 75.5%, 65.2%, 69.8%, 71.4% and 70.5%, respectively. For CRC, 5.05 was the significant SUVmax cutoff value (area under the curve 0.817, P < 0.001), with a sensitivity, specificity, PPV, NPV, and accuracy of 84.8%, 71.0%, 80.9%, 89.8%, and 75.8%, respectively. Objectives:We investigated whether the selective cerebral perfusion(SCP)technique causes differences in changes in cerebral perfusion between both hemispheres in young infants,using cerebral oxygen saturation(ScO2)as an ***,we determined the association between the discrepancy in ScO2 and cerebral perfu-sion pressure during ***:The difference in ScO2 between the left and right cerebral hemispheres(ΔScO2 Rt-Lt)was calculated during clamping of the innominate artery(IA)and during ***:In 25 infants(aged 2 to 78 days),the left and right ScO2 were well maintained(median 63.2%and 60.9%during IA clamping,respectively;64.0%and 65.6%during SCP,respectively).During IA clamping,right and left ScO2 decreased(med-ian-1.4%and-1.0%,respectively).During SCP,rightΔScO2 was higher compared to leftΔScO2(median 1.5%vs.0.6%;p<0.001).Eight patients had a higher rightΔScO2 than leftΔScO2 throughout *** had lowerΔScO2 Rt-Lt during IA clamping(median-3.2%vs.0.0%;p<0.001)and higherΔScO2 Rt-Lt during SCP than others(median 5.0%vs.-0.8%;p<0.001).During and after SCP,the correlation coefﬁcient between rightΔScO2 and change in the mean arterial pressure was higher in patients with a discrepancy than in others(r=0.731 vs.r=0.519;p<0.001).Conclusions:This study suggests that SCP permits adequate bilateral cerebral ***,the unilateral cerebral perfusion technique may cause a difference in cerebral perfusion between both hemispheres in young infants;this may depend on the perfusion pressure.

Vitamin D Is Associated with Severity and Mortality of Non-alcoholic Fatty Liver Disease: A US Population-based Study

Background and Aims:There has been increasing evidence that vitamin D deficiency may increase the risk of metabolic *** metabolic syndrome is a major risk factor for non-alcoholic fatty liver disease (NAFLD),we aimed to investigate the association between vitamin D and the severity and mortality of ***:Data was obtained from the United States Third National Health and Nutrition Examination Survey conducted in 1988-1994,with followup mortality data through *** was defined by ultrasonographic detection of hepatic steatosis in the absence of other liver diseases and categorized as normal,mild,moderate or *** severity of hepatic fibrosis was determined by NAFLD fibrosis score (NFS).ANOVA (F-test) was used to evaluate the association between vitamin D level and degree of NAFLD,and Cox proportional hazards regression analysis was used for survival ***:Vitamin D levels for normal,mild,moderate and severe steatosis were 25.1 ± 0.29 ng/mL,24.7 ± 0.42 ng/mL,23.7 ± 0.37 ng/mL and 23.6 ± 0.60 ng/mL,respectively (trend p < 0.001).Likewise,vitamin D levels for low,intermediate and high NFS categories were 24.7 ± 0.38 ng/mL,23.4 ± 0.42 ng/mL and 21.5 ± 0.57 ng/mL,respectively (trend p < 0.001).After median-follow up over 19 years,vitamin D deficiency was significantly associated with diabetes-and Alzheimer's diseaserelated mortality (hazard ratio (HR):3.64,95%CI:1.51-8.82 and HR:4.80,95%CI:1.53-15.1,respectively),with a borderline significance in overall mortality (HR:1.16,95%CI:0.99-1.36,p =0.06).Conclusion:Vitamin D level was inversely related to the degree of liver steatosis and ***,vitamin D deficiency was associated with diabetes-and Alzheimer's disease-related mortality in NAFLD patients.

Though Hongbaekjeong(HBJ),a hebal mixture of three medicinal plants,has been traditionally used for arthritis and muscular pain,its scientific evidence still remains ***,in the present study,analgesic and anti-inflammatory mechanism of HBJ was evaluated in vitro and in *** significantly reduced NO production and prostaglandin E2(PGE2)release and also attenuated the expression of cyclooxygenase 2(COX-2)in lipopolysaccharides(LPS)and interferon(IFN)-c treated RAW 264.7 ***,HBJ abrogated the production of proinflammatory cytokines such as interleukin(IL)1b,IL-6,IL-8 and monocyte chemoattractant protein-1(MCP-1)in LPS and IFN-c treated RAW 264.7 *** addition,HBJ significantly decreased the number of writhing syndrome induced by acetic acid,and also increased latency in hot-plate method and tail flick test in ***,HBJ significantly reduced the edema volume in the hind paw of the rats with arthritis induced by Freund’s complete adjuvant(FCA)compared to untreated ***,our findings demonstrate the antiinflammatory and analgesic potential of HBJ via inhibition of proinflammatory cytokines and PGE2 release for treatment of arthritis and muscular pain.

AIM:To assess the prognostic value of preoperative 18 fluorodeoxyglucose positron emission tomography(FDG-PET)/computed tomography(CT) in patients with resectable colorectal ***:One hundred sixty-three patients with resectable colorectal cancer who underwent FDG-PET/CT before surgery were *** data including pathologic stage at presentation,histology,treatment,disease-free survival and the maximum standardized uptake value(SUVmax) of the primary tumor on FDG-PET/CT were retrospectively *** follow up duration was 756(range,419-1355).The primary end point was disease-free ***:Twenty-five of 163 patients(15.3%) had *** median SUVmax values of the recurrence and no-recurrence groups were 8.9(range,5-24) and 8.2(range,0-23,P = 0.998).Receiver operating characteristic(ROC) curve analysis showed no significant association between SUVmax and recurrence(area under the curve = 0.5,P = 0.998,95% CI:0.389-0.611).Because a statistically significant value was not found,SUVmax was dichotomized at its median of *** disease-free survival curve was analyzed using the median SUVmax(8.6) as the cut *** and multivariate analysis did not provide evidence that disease-free survival rates for the subgroups defined by the median SUVmax were significantly different(P = 0.52,P = 0.25).CONCLUSION:Our study suggests that the high FDG uptake of primary mass in resectable colorectal cancer doesn't have a significant relationship with tumor recurrence and disease-free survival.

Magnesium oxysulfate(MOS)whisker is considered as a promising inorganic material recently attracting a great attention for being used as a reinforcing filler for polymer composites due to high aspect ratio and extremely-low bulk *** this study,the MOS was treated with 3-methacryloyloxypropyl-trimethoxy silane(MPS)via sol-gel condensation reactions,which successfully allowed melt mixing with polypropylene(PP)up to 30 wt%of *** tensile strength at yield and modulus of the MOS/PP composites were substantially increased by 50.8%and 362%,respectively,when compared with the pristine *** a novel finding,the flame retardancy of MOS was proved by identifying water evolution at elevated temperatures giving out 9 wt%of water in 250-320℃and 14 wt%in 350-420℃in two *** work demonstrated that the MOS could be an excellent filler for PP not only increasing the mechanical properties in a great extent but also imposing flame retarding capability.

Two novel Jr-conjugated cyanostilbene derivatives, FLU-CNPH and TPE-CNPH, were designed aria synthesized by introducing the strong electron donor 1, 4-dihydropyrro[3,2-b]indole and AIE electron donor tetraphenylethylene （TPE） to the （3＇,5＇-bis（trifluoromethyl）-biphenyl-4-yl）-acetonitrile, respec- tively, Both of them were fully characterized and their AIE behaviors were investigated using fluorescence spectroscopy and FE-SEM images. Their optimized structures and frontier molecular orbitals were calculated with the DFT by using Materials Studio 7,0 software to study the relationship between the structure and properties.
