Bottom-Up Approach towards a Human Wellbeing Assessment for the Design of a Management Plan:A Study Case with Contributions to Improve Sustainable Management of Resources in a Northern Area of Mozambique

The loss of biodiversity and ecosystem services disturbance will result in serious ecological and socioeconomic consequences. How can people be prepared to be more efficient in preserving ecosystems, its services and resources? The Messalo Wilderness Area (MWA) is an area lacking a bottom-up approach for the design of a management plan. This research intended to develop a strategy to gather people’s views to begin a human wellbeing assessment for the area, for which a Digital Questionnaire (DQ) was developed and tested. The specific objectives were to 1) verify the dispersion/distribution of people and elephants;2) study the natural resources used by the inhabitants;3) locate the areas from where the most important natural resources are extracted and where conflict between humans and animals occur;4) propose a Management Plan (MP) capable of mitigating the human-animals conflict, promoting the conservation of elephants and, consequently, of other types of biodiversity, thus working towards a better socio-economical development of the area. The elements gathered in the consulted references were used to design several maps and cartographic figures shown in this article, using the Map Window software. These maps describe: fresh water distribution, habitat distribution, population dispersal and main road connections, crop fields distribution and main exploitation spots of the different resources, orography and topography. The results from the DQ analysis helped to gather indicators to the human wellbeing assessment. The information gathered in the literature and by inquiry was effective in confirming the high dependency of local people on land and sea resources, as well as the conflicts between people and wild animals, such as elephants and African wild dogs, and the reasons for those conflicts. In this study,we investigated the transcriptional spatio-temporal dynamics of the taste 1 receptor(T1R)gene family repertoire in seabream(Sparus aurata[sa]),during larval ontogeny and in adult *** early larval development,sa T1R expression arises heterochronously,*** extraoral taste-related perception in the gastrointestinal tract(GIT)anticipates first exogenous feeding(at 9 days post hatching[dph]),followed by the buccal/intraoral perception from 14 dph onwards,supporting the hypothesis that the early onset of the molecular machinery underlying sa T1R expression in the GIT is not induced by food but rather genetically *** adulthood,we characterized the expression patterns of sa T1R within specific tissues(n=4)distributed in oropharingeal,GIT and brain regions substantiating their functional versatility as chemosensory signaling players to a variety of biological functions beyond oral taste ***,we provided for the first time direct evidences in fish for m RNA coexpression of a subset of sa T1R genes(mostly sa T1R3,*** common subunit of the heterodimeric T1R complexes for the detection of“sweet”and“umami”substances),with the selected gut peptides ghrelin(ghr),cholecystokinin(cck),hormone peptide yy(pyy)and proglucagon(pg).Each peptide defines the enteroendocrine cells(ECCs)identity,and establishes on morphological basis,a direct link for T1R chemosensing in the regulation of fish digestive ***,we analyzed the spatial gene expression patterns of 2 taste signaling components functionally homologous to the mammalian G(i)a subunit gustducin,namely sa G(i)a1 and sa G(i)a2,and demonstrated their co-localization with the sa T1R3in EECs,thus validating their direct involvement in taste-like transduction mechanisms of the fish *** conclusion,data provide new insights in the evolutionary conservation of gut sensing in fish suggesting a conserved role for nutrient sensors modulating entero-endocrine secretion.

Parkinson's disease is the second most prevalent neurodegenerative disorder ***,it is characterized by severe motor complications caused by progressive degeneration of dopaminergic *** treatment is focused on mitigating the symptoms through the administration of levodopa,rather than on preventing dopaminergic neuronal ***,the use and development of neuroprotective/disease-modifying strategies is an absolute need that can lead to promising gains on translational research of Parkinson's *** instance,N-acetylcysteine,a natural compound with strong antioxidant effects,has been shown to modulate oxidative stress,preventing dopamine-induced cell *** the evidence of neuroprotective and modulatory effects of this drug,as far as we know,it does not induce per se any regenerative ***,it would be of interest to combine the latter with innovative therapies that induce dopaminergic neurons repair or even differentiation,as stem cell-based *** cells secretome has been proposed as a promising therapeutic approach for Parkinson's disease,given its ability to modulate cell viability/preservation of dopaminergic *** approach represents a shift in the paradigm,showing that cell-transplantation free therapies based on the use of stem cells secretome may represent a potential alternative for regenerative medicine of Parkinson's ***,in this review,we address the current understanding of the potential combination of stem cell free-based strategies and neuroprotective/disease-modifying strategies as a new paradigm for the treatment of central nervous system neurodegenerative diseases,like Parkinson's disease.

Background:In stage Ⅳ breast cancer,surgical resection of the primary tumor was traditionally performed solely to palliate symptoms such as bleeding,infection,or *** ongoing discussion has shown that there are many research gaps in the current literature and differences in clinical ***,this systematic review and meta-analysis was designed to evaluate how primary tumor resection(PTR)affects the overall survival(OS)of patients with stage Ⅳ breast ***:A thorough literature search was completed using different databases(PubMed,Google Scholar,Scopus,ScienceDirect,and Cochrane Library)to find papers contrasting PTR with no *** quality of research articles was evaluated using the Cochrane Risk of Bias 2.0 Tool and the Newcastle-Ottawa Scale(NOS).Review Manager 5.4 was used to determine how much demographic and clinical factors contribute to heterogeneity through subgroup and meta-regression ***:Data derived from 44 observational studies(OS)and four randomized controlled trials(RCTs)including 227,889 patients were *** all cases,150,239 patients were included in the non-PTR group,and 70,795 patients in the PTR group(37 observational studies and 4 randomized control trials).The pooled outcomes of four RCT studies(Hazard Ratio(HR)=1.03,95%CI:0.67-1.58;I2=88%;P<0.0001;chi-square 24.57)favor *** pooled outcomes of 43 observational studies showed PTR significantly improved OS(HR=0.66,95%CI:0.61-0.71;I2=87%;P<0.00001;chi-square 359.12).Additionally,subgroup analysis that compared PTR with non-PTR in patients with stage IV breast cancer for progression free-survival(HR=0.89,95%CI:0.62-1.28;P=0.03;I2=71%)and locoregional progression-free survival(LPFS)(HR=0.33,95%CI:0.14-0.74;P=0.0004;I^(2)=87%)was found to be significant favoring the PTR *** progression-free survival(DPFS)had a non-significant relationship(HR=0.42,95%CI:0.29-0.60;P=0.12;I^(2)=53%),while overall,there was a significant relationship(HR=0.49,95%CI:0

The Arabidopsis XBAT35 is one of five structurally related ankyrin repeat-containing Really Interesting New Gene （RING） E3 ligases involved in ubiquitin-mediated protein degradation, which plays key roles in a wide range of cel- lular processes. Here, we show that the XBAT35 gene undergoes alternative splicing, generating two transcripts that are constitutively expressed in all plant tissues. The two splice variants derive from an exon skipping event that excludes an in-frame segment from the XBAT35 precursor mRNA, giving rise to two protein isoforms that differ solely in the presence of a nuclear localization signal （NLS）. Transient expression assays indicate that the isoform lacking the NLS localizes in the cytoplasm of plant cells, whereas the other is targeted to the nucleus, accumulating in nuclear speckles. Both isoforms are functional E3 ligases, as assessed by in vitro ubiquitination assays. Two insertion mutant alleles and RNA-interference （RNAi） silencing lines for XBAT35 display no evident phenotypes under normal growth conditions, but exhibit hyper- sensitivity to the ethylene precursor 1-aminocyclopropane-l-carboxylate （ACC） during apical hook exaggeration in the dark, which is rescued by an inhibitor of ethylene perception. Independent expression of each XBAT35 splice variant in the mutant background indicates that the two isoforms may differentially contribute to apical hook formation but are both functional in this ethylene-mediated response. Thus, XBAT35 defines a novel player in ethylene signaling involved in negatively regulating apical hook curvature, with alternative splicing controlling dual targeting of this E3 ubiquitin ligase to the nuclear and cytoplasmic compartments.

This research aimed to quantify concentrations of ammonia(NH3),carbon dioxide(CO_(2))and methane(CH_(4)),estimate emissions,and analyze the factors influencing them during warm periods in an open dairy barn equipped with two cooling systems in a Mediterranean climate *** distribution within the barn was observed to vary both vertically and horizontally,with the highest gas concentrations observed in the central area of the ***_(3),CH_(4)and CO_(2)ranged in 1.7–7.4,7–18,560–724μg·g^(–1),*** ventilation through openings and the operation of cooling systems induced changes in indoor microclimate conditions,influencing cow behavior and,consequently,gas *** concentrations were the highest at air velocities below 0.5 m·s^(–1).The highest concentration of NH_(3)was observed when the temperature-humidity index(THI)was>72 and≤78;and CO_(2)and CH_(4)concentrations were the highest with THI≥72 and decreased with THI≤***_(3)concentrations when barn management included three daily milkings were higher than those measured when barn management was based on two daily milkings,and lower for CH_(4)and CO_(2).NH_(3)and CH_(4)emissions were the highest during barn cleaning,while the lowest NH_(3)emissions occurred during activity of the cows(i.e.,feeding,walking).

A 74-year-old man with terminal chronic kidney disease, under hemodialysis and with residual diuresis, was admitted due to myalgia, arthralgia, fever and pyuria in the previous 10 days. The patient had a recent diagnosis of high-grade non-invasive bladder cancer and was doing weekly BCG intravesical administrations. The symptoms started three days before the fifth administration. He had done cefixime as an outpatient and started piperacillin-tazobactam on hospital admission, but the fever persisted, and there was no bacterial isolation in urine or blood culture. On the tenth and seventeenth day after the last BCG intravesical administration Mycobacterium bovis was still isolated in the urine culture. The diagnosis of BCGitis was made and treatment was started, with a good response. Forty days after the last administration and under treatment, the culture remained positive for Mycobacterium bovis in the urine. We raise the question about the safety of BCG administration in patients with residual diuresis.
