As a late endosomal/lysosomal transport protein of the P5-type, ATP13A2 is capable of removing the abnormal accumulation of α-synuclein, which maintains the homeostasis of metal ions and polyamines in the central nervous system. Furthermore, ATP13A2 regulates the normal function of several organelles such as lysosomes, endoplasmic reticulum (ER) and mitochondria, and maintains the normal physiological activity of neural cells. Especially, ATP13A2 protects dopaminergic (DA) neurons against environmental or genetically induced Parkinson's disease (PD). As we all know, PD is a neurodegenerative disease characterized by the loss of DA neurons in the substantia nigra pars compacta. An increasing number of studies have reported that the loss-of- function of ATP13A2 affects normal physiological processes of various organelles, leading to abnormalities and the death of DA neurons. Previous studies in our laboratory have also shown that ATP13A2 deletion intensifies the neuroinflammatory response induced by astrocytes, thus inducing DA neuronal injury. In addition to elucidating the normal structure and function of ATP13A2, this review summarized the pathological mechanisms of ATP13A2 mutations leading to PD in existing literature studies, deepening the understanding of ATP13A2 in the pathological process of PD and other related neurodegenerative diseases. This review provides inspiration for investigators to explore the essential regulatory role of ATP13A2 in PD in the future.

Lithium metal anode is considered the alternative to graphite anode due to its ultra-high theoretical capacity of 3860 mAh·g^(-1).However,serious Li dendrite growth and drastic electrolyte side reactions restrain the commercial application of Li metal anode.In this work,a Li_(3)Bi/LiF interfacial layer is constructed on the surface of the Li metal anode by a spontaneous substitution reaction.The composite interfacial layer possesses excellent ionic conductivity,high mechanical strength,and great electrolyte wettability,which ensures fast Li-ion transfer and uniform Li deposition of the Li_(3)Bi/LiF@Li anode.Impressively,the Li_3Bi/LiF@Li symmetric cell provides a cycle life of more than 400 h with only 73 mV voltage polarization at 10 mA·cm^(-2).By pairing with commercial NCM622 cathode,the Li_(3)Bi/LiF@Li full cell exhibits a long cycle at a rate of 2 C.

目的探索信迪利单抗联合安罗替尼治疗晚期非小细胞肺癌(NSCLC)的疗效和安全性。方法回顾性分析2019年5月至2021年8月于本院就诊的信迪利单抗(200 mg ivgtt qd,第1日,21 d为一个周期)联合安罗替尼(10 mg po qd,第1~14日,停7 d,21 d为一个周期)治疗的60例晚期NSCLC患者的临床资料。记录患者的临床特征、客观缓解率(ORR)、疾病控制率(DCR)、无进展生存期(PFS)和不良反应发生情况,分析不同临床特征与近期疗效的关系,多因素回归分析预后危险因素。结果所有患者的ORR为33%,DCR为72%;其中29例一线治疗患者的ORR为52%、DCR为83%,31例二线及以上治疗患者的ORR为16%、DCR为61%。Ⅲ期患者有效率高于Ⅳ期患者(80%vs.24%,P<0.05),鳞癌患者有效率高于腺癌和其他病理类型患者(56%vs.13%vs.50%,P<0.05),一线治疗患者有效率高于二线及以上治疗患者(52%vs.16%,P<0.05)。所有患者的中位PFS为5.1个月,其中一线治疗患者为23.3个月,二线及以上的为3.0个月。一线治疗中位PFS远长于二线及以上(23.3个月vs.3.0个月,P<0.05)。年龄≥65岁患者较年龄<65岁患者疾病进展风险增加(HR=2.215,95%CI:1.043~4.705,P<0.05),ECOG 2分患者较ECOG 1分患者疾病进展风险增加(HR=8.905,95%CI:3.671~21.603,P<0.05),一线治疗患者较二线及以上治疗患者疾病进展风险降低(HR=0.233,95%CI:0.107~0.506,P<0.05)。不良反应总发生率为60%,≥3级不良反应占8%,分别为乏力、肝功能损伤和糖尿病,均在对症处理后改善。结论信迪利单抗联合安罗替尼治疗晚期NSCLC具有一定疗效,且患者可耐受。年龄、ECOG评分和治疗线数是影响患者预后的独立危险因素。

加工和维护时长的波动使生产实际运行与计划之间产生较大偏离,使所预留的维护契机变得不可行。为此,引入两个不确定参数描述加工和维护时长的波动程度和约束条件的允许违背程度,将考虑预防维护的多工艺路线柔性作业车间鲁棒调度问题转换为确定条件下的鲁棒对等问题;基于编码方式和邻域搜索算法设计了一种模拟退火算法求解该问题,将鲁棒调度方案与企业实际生产方案进行对比,通过实验证明了鲁棒优化方案的有效性。

A Bio-Inspired Integration Model of Basal Ganglia and Cerebellum for Motion Learning of a Musculoskeletal Robot

It is a significant research direction for highly complex musculoskeletal robots that how to develop the ability of motion learning and generalization.The cooperations of multiple brain regions are crucial to improving motion performance.Inspired by the neural mechanisms of structures,functions,and interconnections of basal ganglia and cerebellum,a biologically inspired integration model for motor learning of musculoskeletal robots is proposed.Based on the neural characteristics of the basal ganglia,the basal ganglia actor network,which mainly simulates the dorsal striatum,outputs motion commands,and the basal ganglia critic network,which simulates the ventral striatum,estimates actionstate values.Their network parameters are updated using the soft actor-critic method.Based on the sensorimotor prediction mechanism of the cerebellum,the cerebellum network evaluates the state feature extraction quality of the basal ganglia actor network and then updates the weights of its feature layer.This learning method is proven to converge to the optimal policy.Furthermore,drawing on the mechanism of dopaminergic dynamic regulation in the basal ganglia,the adaptive adjustment of target entropy and the dopaminergic experience replay are proposed to further improve the integration model,which contributes to the exploration-exploitation trade-off of motor learning.The bio-inspired integration model is validated on a musculoskeletal system.Experimental results indicate that this model can effectively control the musculoskeletal robot to accomplish the motion task from random starting locations to random target positions with high precision and robustness.

The roles of dietary cholesterol in fish physiology are currently contradictory.The issue reflects the limited studies on the metabolic consequences of cholesterol intake in fish.The present study investigated the metabolic responses to high cholesterol intake in Nile tilapia(Oreochromis niloticus),which were fed with four cholesterol-contained diets(0.8,1.6,2.4 and 3.2%)and a control diet for eight weeks.All fish-fed cholesterol diets showed increased body weight,but accumulated cholesterol(the peak level was in the 1.6%cholesterol group).Then,we selected 1.6%cholesterol and control diets for further analysis.The high cholesterol diet impaired liver function and reduced mitochondria number in fish.Furthermore,high cholesterol intake triggered protective adaptation via(1)inhibiting endogenous cholesterol synthesis,(2)elevating the expression of genes related to cholesterol esterification and efflux,and(3)promoting chenodeoxycholic acid synthesis and efflux.Accordingly,high cholesterol intake reshaped the fish gut microbiome by increasing the abundance of Lactobacillus spp.and Mycobacterium spp.,both of which are involved in cholesterol and/or bile acids catabolism.Moreover,high cholesterol intake inhibited lipid catabolic activities through mitochondrialβ-oxidation,and lysosome-mediated lipophagy,and depressed insulin signaling sensitivity.Protein catabolism was elevated as a compulsory response to maintain energy homeostasis.Therefore,although high cholesterol intake promoted growth,it led to metabolic disorders in fish.For the first time,this study provides evidence for the systemic metabolic response to high cholesterol intake in fish.This knowledge contributes to an understanding of the metabolic syndromes caused by high cholesterol intake or deposition in fish.

Calculation of Surrounding Rock Pressure of Undercut Subway Station Based on Multi-Factor Affecting Pressure Arch Theory

The formation mechanism of surrounding rock pressure in large-section tunnels of underground excavation is still unclear. The traditional methods for calculating surrounding rock pressure have certain limitations. Based on the theory of pressure arch and parameter sensitivity analysis, the internal and external boundaries of the pressure arch are solved by numerical simulation. The loose pressure is obtained according to the distance from the inner boundary to the inner wall of the tunnel. The pressure arch deformation is analyzed based on the Winkler model. The surrounding rock pressure is calculated, and the surrounding rock pressure based on the pressure arch theory is calculated. The calculation method of surrounding rock pressure based on the pressure arch theory large section tunnel is proposed. The orthogonal experimental design method was used to select four representative factors: tunnel depth H, span B, internal friction angle φ and rock weight γ. The calculation results of each group of experiments were compiled and multivariate linear regression was used. The method is statistically analyzed, and finally, the calculation formula of the surrounding rock pressure of the large-section tunnel affected by multiple factors is obtained and applied to the calculation of the surrounding rock pressure of the metro station.

环境干扰噪声是泥石流次声现场监测的主要挑战,极大限制了泥石流次声信号识别的准确率。鉴于深度学习在声学信号识别中的优异表现,本文提出一种基于改进的AlexNet网络的泥石流次声信号识别方法,有效提升泥石流次声信号识别准确率和收敛速度。首先对原始次声数据集进行数据扩充、滤波降噪等预处理,并利用小波变换生成时频谱图像,然后将得到的时频谱图像作为输入,通过减小卷积核、引入批量归一化层和选择Adam优化算法搭建改进的AlexNet网络模型。实验结果表明,改进的AlexNet网络模型识别准确率为91.48%,实现了泥石流次声信号的智能识别,可为泥石流次声监测预警提供高效、可靠的技术支撑。
