Metabolic alterations and hepatitis c:From bench tobedside

In addition to causing cirrhosis and hepatocellular carcinoma, hepatitis c virus(HcV) is thought to cause hypolipidemia, hepatic steatosis, insulin resistance, metabolic syndrome, and diabetes. the viral life cycle of HcV depends on cholesterol metabolism in host cells. HcV core protein and nonstructural protein 5A perturb crucial lipid and glucose pathways, such as the sterol regulatory element-binding protein pathway and t he prot e in kinase b /mammal ian t arget of rapamycin/s6 kinase 1 pathway. Although several lines of transgenic mice expressing core or full HcV proteins exhibit hepatic steatosis and/or dyslipidemia, whether they completely reflect the metabolic alterations in humans with HcV infection remains unknown. Many cross-sectional studies have demonstrated increased prevalences of metabolic alterations and cardiovascular events in patients with chronic hepatitis c(cHc); however, conflicting results exist, primarily due to unavoidable individual variations. Utilizing anti-HcV therapy, most longitudinal cohort studies of cHc patients have demonstrated the favorable effects of viral clearance in attenuating metabolic alterations and cardiovascular risks. to determine the risks of HcV-associated metabolic alterations and associated complications in patients with cHc, it is necessary to adjust for crucial confounders, such as HcV genotype and host baseline glucose metabolism, for a long follow-up period after anti-HcV treatment. Adipose tissue is an important endocrine organ due to its release of adipocytokines, which regulate lipid and glucose metabolism. However, most data on HcV infection and adipocytokine alteration are inconclusive. A comprehensive overview of HcV-associated metabolic and adipocytokine alterations, from bench to bedside, is presented in this topic highlight.

Vertically standing Ptse_2 film: a saturable absorber for a passively mode-locked Nd:LuVO_4 laser

the novel vertically standing Pt se2 film on transparent quartz was prepared by selenization of platinum film deposited by the magnetron sputtering method, and an Nd:Lu VO4 passively mode-locked solid-state laser was realized by using the fabricated Pt se2 film as a saturable absorber. the X-ray diffraction pattern and Raman spectrum of the film indicate its good crystallinity with a layered structure. the thickness of Pt se2 film is measured to be 24 nm according to the cross-section height profile of the atomic force microscope image. Highresolution transmission electron microscopy images clearly demonstrate its vertically standing structure with an interlayer distance of 0.54 nm along the c-axis direction. the modulation depth(Δt) and saturation fluence(Φs)of Pt se2 film are measured to be 12.6% and 17.1 μJ∕cm2, respectively. the obtained mode-locked laser spectrum has a central wavelength of 1066.573 nm, with a 3 d b bandwidth of 0.106 nm. the transform limited pulse width of the mode-locked laser was calculated to be 15.8 ps. A maximum average output power of 180 m W with a working repetition rate of 61.3 MHz is obtained. to the best of our knowledge, this is the first report of the generation of ultrafast mode-locked laser pulses by using layered Pt se2 as a saturable absorber.

there has recently been a dramatic renewal of interest in hadron spectroscopy and charm physics. this renaissance has been driven in part by the discovery of a plethora of charmonium-like XYZ states at bEsⅢ and b factories, and the observation of an intriguing proton-antiproton threshold enhancement and the possibly related X(1835) meson state at bEsⅢ, as well as the threshold measurements of charm mesons and charm baryons. We present a detailed survey of the important topics in tau-charm physics and hadron physics that can be further explored at bEsⅢ during the remaining operation period of bEPcⅡ. this survey will help in the optimization of the data-taking plan over the coming years, and provides physics motivation for the possible upgrade of bEPcⅡ to higher luminosity.

赣江上游桃江流域水沙变化及驱动因素

为揭示近年来赣江上游输沙量急剧减少的影响因素及其贡献率,以赣江上游桃江流域为例,采用流域内13个雨量站1958—2018年降水数据计算得出面雨量和流域控制站居龙滩站同期径流、输沙数据,结合趋势分析、突变检验等方法,对桃江流域61 a水沙变化特征进行分析,并采用累积量斜率变化率比较法计算气候因子和人类活动在各个时段对流域水沙变化的相对贡献率。结果表明:桃江流域多年平均降水量、径流量和输沙量分别为1565.5 mm、60.36亿m^(3)和112.5万t;流域61 a降雨和径流无显著变化趋势和突变点,但降雨、径流在1972和1985年有明显波动;年输沙量存在显著减少趋势(α<0.01),在2002年发生突变(P<0.001);若以人类活动相对较弱的1958—1972年为基准期,计算得到1973—1985、1986—2002和2003—2018年人类活动对流域输沙量减少的贡献率分别为79.14%、92.05%和100%;以水库建设为代表的水利工程和水土保持措施等人类活动是赣江上游桃江流域输沙量减少的主导因素。研究对于评价水利工程等人类活动对流域水沙的影响具有参考价值,可为流域实施水土保持措施提供科学依据。

Role of circular RNA expression in the pathological progression after spinal cord injury

Aquaporins（AQPs） are specific membrane channels for water and other small nonionic *** order to overcome the difficulties to generate the effictive antibody of membrane protein,we selected the cytoplasmic c-terminus of Aquaporin 1（AQP1） as an unique *** long c-terminus of mouse AQP1 was overexpressed in the Glutathione s-tansferase Gene Fusion *** the basis of the resonable amounts of soluable membrane protein peptides,we prepared the specific *** pursure this object,we constructed pGEX-4t-1/mAQP1（DNA sequence from 700 to 801 bp） recombinant plasmid and transformed it into Escherichia coli bL21 *** Gst-AQP1 c-terminal hydrophilic peptide fusion protein was induced by IPtG and further purified by Glutathione sepharose 4b to obtain the right size fusion *** we immunized the New Zealand rabbits to prepare the *** purified AQP1 antibody showed high sensitivity by ELIsA assay and high specificity by Western blot with AQP1 null mice served as negative ***,we also checked the AQP1 localization in the mouse renal tissues in wild type of mice and AQP1 null mice served as negative *** demonstrated that AQP1 was highly expressed at the descending limb of Henle tube using our purified AQP1 antibody,which was consistent with previous *** successful design and preparation of AQP1 antibody through Gst technique is an example as making antibodies against a specific membrane protein.

Novel HbV mutations and their value in predicting efficacy of conventional interferon

bAcKGROUND： Accumulating studies assessing the impacts of hot spot mutations on conventional interferon（IFN） efficacy come to discrepant conclusions; studies regarding the mutations in s and Rt regions are also unclear. the present study aimed to evaluate the impacts of HbV mutations on the efficacy of conventional IFN. MEtHODs： A total of 126 patients who received conventional IFN treatment for 48 weeks were enrolled. biochemical and serological parameters were routinely tested. the sequences of HbV from 78 serum samples were amplified by nested-PcR; mutations were identified with sequence scanner V1.0 after AbI 3730 xl direct sequencing, HbV genotypes were determined according to Rt gene sequences utilizing NcbI Genotyping tool which was based on phylogenetic ***： the baseline DNA levels of virological response（VR） group were significantly lower than those of no VR group [7.13±0.76 vs 7.69±0.56 lg（copies/m L）, P=0.001]. the baseline ALt levels were significantly higher in the Hbe Ag clearance group（204.72±88.65 vs 162.80±85.81 IU/L, P〈0.05） andHbe Ag seroconversion group（204.89 ±95.68 vs 166.75±84.43 IU/L, P〈0.05）. Females and lower bMI levels（20.01±2.33 vs 21.65±3.66 kg/m^2, P〈0.05） were prone to acquired biochemical response（bR）. Pc-W28stOP（ntG 1896A） was significantly higher in the combined response（cR） group than that in the no cR group（91.7% vs 39.7%, P=0.001）. Multivariate logistic regression analysis showed that baseline DNA, Pc-P159t（ntc2288A）, bcP-N118t（ntA 1726c） and bcP-L134L（ntA 1775c/G/t） influenced VR independently. Pc-G182c（nt G2357t）, Pc-s64A/t（nt t2003G/A） and bMI were independent influence factors for Hbe Ag clearance, Hbe Ag seroconversion and bR, respectively. the new predicting model concluded that baseline DNA and new mutations for VR were established successfully, and ROc analysis showed that AUc was 0.842（P〈0.001） with a sensitivity of 0.652 and a specificity of ***： Pc-P159t（ntc 2288A）, bcP-N118t（ntA 1726c）, bcP-

During the 2016-17 and 2018-19 running periods, the bEsⅢ experiment collected 7.5 fb-1of e~+e~- collision data at center-of-mass energies ranging from 4.13 to 4.44 GeV. these data samples are primarily used for the study of excited charmonium and charmoniumlike states. by analyzing the di-muon process e~+e~-→(γIsR/FsR)μ~+μ~-, we measure the center-of-mass energies of the data samples with a precision of 0.6 MeV. through a run-by-run study, we find that the center-of-mass energies were stable throughout most of the data-collection period.

目的:从肠道菌群角度探讨中药复方田黄方改善老年高脂血症小鼠脂代谢紊乱的作用机制。方法:3 w~4 w龄雄性c57bL/6J小鼠正常饮食喂养至12月成自然衰老小鼠,喂饲高脂饮食14 w构建老年高脂血症模型,给予田黄方提取物100 mg/kg干预10 w,正常对照组予普通饲料喂养至试验结束。试剂盒检测小鼠血清和肝脏总胆固醇(tc)、三酰甘油(tG)含量、胆盐水解酶(bsH)活力,靶向代谢组学联合16s rDNA测序分析胆汁酸代谢谱以及肠道菌群结构,Western blot、PcR法检测法尼醇X受体(FXR)、蛋白酪氨酸磷酸酶1(sHP1)、胆固醇7α-羟化酶(cYP7A1)、成纤维生长因子15(FGF15)的mRNA和蛋白表达情况。结果:与模型对照组相比,田黄方提取物100 mg/kg组小鼠肝脏tc、tG含量显著下降(Pscherichia)、阿克曼菌属(Akkermansia)、拟杆菌属(bacteroides)丰度明显减少(PbsH酶活力下降(PcA)、鹅脱氧胆酸(cDcA)和石胆酸(LcA)含量降低(PtβMcA)20 mg/kg组小鼠血清及肝脏tc、tG含量显著下降(Pcyp7a1 mRNA和蛋白表达上调(PbsH活性,使tβMcA水平增加进而抑制肠道FXR、FGF15表达,同时下调肝脏FXR和sHP表达,上调cYP7A1表达,导致胆汁酸合成增加,tc降低。田黄方通过肠道菌群-tβMcA-FXR轴发挥降脂作用。

Relationship between cytokines gene polymorphism and susceptibility to hepatitis b virus intrauterine infection

background the influences of genomic background are confirmed in more diseases. Immunologic tolerance after intrauterine infection of hepatitis b virus is considered to occur in t cells. cytokines work effectively in eliminating virus by immune system after hepatitis b virus infection. to explore the relationship between cytokines （tumor necrosis factor-α, interferon-γ, interleukin-4 and interleukin-10）, which expressed abnormal quantity in the peripheral blood to intrauterine hepatitis b virus infectious children, gene single nucleotide polymorphism （sNP） and susceptibility to hepatitis b virus intrauterine infection. Methods this is a cross sectional study of molecular clinical epidemiology. the subjects in this study were selected from outpatients of hepatitis b vaccine follow-up special clinics of our hospital in the period. According to intrant criteria, the high risk children of hepatitis b virus （HbV） intrauterine infection were divided into immune failure group （group Ⅰ） ; and immune effective group （group Ⅱ） and non high risk children belonged to the control group. Four gene sNP sites of tNF-α -238, IFN-γ ＋874, IL-4 -590 and IL-10 -1082 were determined by real-time quantitative fluorescent polymerase chain reaction （PcR）. Results the significant differences of tNF-α- 238 A allele frequency were found between group Ⅰand group Ⅱ（X^2 =6. 797, P 〈 0.05 ） and between group Ⅰ and the control group （ X^2 = 9. 513, P 〈 0.05 ）. No evident differences of tNF-α- 238 A were found between group Ⅱand control group （ X^2 = 0. 047, P 〉 0. 05 ） ; the significant differences of IFN-γ ＋ 874 A allele frequency were found between group Ⅰand group Ⅱ（ X^2 = 7. 238, P〈0. 05）, and between group Ⅰand the control group （X^2 =5. 199,P 〈0. 05）. No evident differences were found between groupⅡ and the control group （ X^2 =0. 602 ,P 〉0. 05 ） ; the significant differences of IL-4 -590 c/t allele frequency were not found between groupⅠand group Ⅱ （ X^2 = 0. 632, P 〉 0. 05 ） , also group Ⅰ
