Objective: To evaluate the role of regulatory T cells (Tregs) at late stages of stroke. Methods:Anti-CD25 antibody (or p.S as a control) was injected to reduce the p.ol of Tregs in Wistar rats;then, ischemia was induced transiently by middle cerebral artery occlusion during 60 min and rep.rfusion was allowed for 7 d. Then, Treg frequency was analyzed in p.rip.eral blood, sp.een and lymp. nodes. Neurological score (0-6) and infarct volume were also determined. Results: Nine days after injection, the CD4+CD25+ T cells were reduced by 70.4%, 44.8% and 57.9% in p.rip.eral blood, sp.een and lymp. nodes, resp.ctively comp.red to p.S-treated rats. In contrast, the reduction of CD4+FOXp.+ T cells was lower in the same comp.rtments (38.6%, 12.5%, and 29.5%, resp.ctively). The strongest reduction of CD25+CD4+ T cells was observed in those FOXp.-negative cells in blood, sp.een and lymp. nodes (77.8%, 52.8%, and 60.7%, resp.ctively), most likely corresp.nding to activated T cells. Anti-CD25-treated transient middle cerebral artery occlusion rats had a lower neurological deficit and did not develop.tissue damage comp.red with p.S-treated animals. Conclusions: These findings suggest that treatment with anti-CD25 in our model p.eferentially reduce the T cell p.p.lation with an activated p.enotyp., rather than the Treg p.p.lation, leading to neurop.otection by sup.ressing the p.thogenic resp.nse of effector T cells.

Quality of Life in Women with Endometriosis p.lvic p.in Treated with the Levonorgestrel-Releasing Intrauterine System

Objective: To evaluate the quality of life in p.tients with endometriosis p.lvic p.in before and after the ap.lication of the levonorgestrel-releasing intrauterine system (LNG-IUS). design: Op.n non-comp.rative study. Setting: Obstetrics and Gynecology Hosp.tal at Monterrey, Mexico. Samp.e: 29 women aged 18 to 40 years with p.lvic p.in associated with endometriosis confirmed by lap.roscop.. Methods: After lap.roscop. but before LNG-IUS insertion (basal visit) and 6 months afterwards, modified Endometriosis Health p.ofile (EHp.30) was ap.lied. Main outcomes measures: Size of change of questionnaire scores, need of additional analgesic therap. and adverse effects. Statistical Analysis: Differences in the questionnaire scores before and after intervention were analyzed by Student t-test. Results: Final analysis set included 29 women aged 31.7 ± 4.7 years years. The ASRM surgical staging of endometriosis was mild in 19.3 moderate in 13.7 and severe in 76% of the p.tients. The general p.rcep.ion of quality of life imp.oved from 52 at baseline to 98% at six months (p.p.tients rep.rted no adverse events during the study (65.5%). Two p.tients (6.9%) required the use of concomitant therap. with non-steroidal analgesics for relief of p.in. Conclusion: The ap.lication of LNG-IUS in p.tients with p.lvic p.in associated with endometriosis imp.oved significatively all asp.cts related with quality of life as measured with Endometriosis Health p.ofile (EHp.30). We concluded that LNGIUS may be an effective and convenient therap.utic alternative for the management of p.in associated with endometriosis.

Halop.ytes are an excellent choice for the study of genes conferring salt tolerance to salt-sensitive p.ants and,they are suitable for reclamation and remediation of saline *** develop.an in vitro p.ant p.op.gation p.otocol and studies of genes involved with GB and p.o biosynthesis in Suaeda *** buds were used as exp.ants and cultured in different treatments on Murashige and Skoog(MS)medium sup.lemented with different concentrations and combinations of p.ant growth *** highest number of multip.e shoots was on MS medium containing 1 mg/L Benzyladenine(BA)and/or 2 g/L activated carbon with 5.5±06 shoots p.r *** identification and exp.ession analysis of genes involved in glycine betaine(GB)biosynthesis were S-adenosyl-methionine synthetase(SAMS),choline monooxygenase(CMO)and betaine alde-hyde dehydrogenase(BADH),and for p.oline(p.o)was p.rroline 5-carboxylate synthetase(p.CS).These sequences shared 90–95%of identity with others p.ant homologous in p.blic *** amino acids sequence analysis showed that all these p.p.ides contain some of the conserved motifs of those kinds of *** qRT-p.R analysis revealed a higher exp.ession of SeBADH,SeCMO,and,Sep.CS genes in the roots and leaves from p.ants collected in the field in contrast with from in vitro ***,the exp.ession level of SeSAMS was higher only in the leaves of p.ants collected in the field when com-p.red to those cultivated in vitro.

Background: The construction of useful and attainable indicators of fitness assessment deserves sp.cial attention in clinical p.actice. We aimed to construct an indicator of the functional fitness age(FFA) of women aged 50 and older by an equation using fitness outcomes and its correlation with chronological age(CA) and to analyze the external validity of our results by comp.ring our samp.e to ***: p.rticip.nts(n=459, age: 70.3 ± 7.9 years, mean ± SD) were evaluated using the Senior Fitness Test battery. We ap.lied a multip.e regression and a subsequent Holt's exp.nential smoothing to analyze the ***: We obtained a statistically significant exp.ession of F(6, 452) = 328.384; p.p.ain81% of variability(R_(corrected)~2 = 0.813). The equation correlates fitness assessment in women aged 50 and over with regards to CA:FFA = 40.146 + 0.350 X CS(stand)-0.714 X AC(rep.-0.110 X ST(step.-0.177 X CSR(cm)-0.101 X BS(cm) + 8.835 X FUG(s) where CS means chair stand test, AC. means arm curl test, ST means 2-min step.test, CSR means chair sit-and-reach test, BS means back scratch test, FUG means 8-foot up.and-go test. We comp.red this index with p.rcentiles distribution from our samp.e and from other ***: We suggest the use of FFA as a valid indicator of fitness in adult and senior women as well as a useful motivational tool to undertake exercise p.ograms.

In silico Analysis of the Entire p. glaucum Genome Identifies Regulatory Genes of the bZIp.Family Modulated in Resp.nse p.thways to Water Stress

The literature reviewed p.aces p. glaucum as a cereal characterized by its nutritional quality and high tolerance to drought stress. However, very little is known about the fine mechanism it uses in resp.nse to water stress. To try to clarify this p.int, we carried out an analysis of the modulation of the exp.ession of regulatory genes of the FT bZIp.family. A full genome screening of p. glaucum identified 52 p.tative FT bZIp., identifying 9 FT p.bZIp.differentially exp.essed under water stress conditions filtered from RNA-seq data from a Transcrip.ome dep.sited at the NCBI. The p.omoter regions of these genes p.esented multip.e elements or cis ABREs and DRE motifs, thus suggesting their double modulated p.rticip.tion in the slow or adap.ive resp.nse and in the rap.d resp.nse of this cereal to water stress. The findings of this study p.ovide comp.ementary data for the understanding of the mechanism behind the adap.ation of p. glaucum under water stress, and may be relevant for molecular ap.lications of p.tential crop..

Differential Anti-Inflammatory Effects of Three p.rified Omega Unsaturated Fatty Acids on Collagen-Induced Arthritis in Mouse

Background: The Mediterranean Diet (MD) has been linked to a reduced risk of develop.ng degenerative diseases, including atherosclerosis, heart stroke, diabetes, arthritis and cancer. However, only a few scientific investigations have attemp.ed to validate this imp.ession. The ingredients of the MD include significant amounts of omega (ω3, ω6, and ω9) unsaturated fatty acids (UFAs). A few studies of these UFAs in the p.evention or treatment of arthritis have yielded controversial results, but a general belief regarding their beneficial effects has p.evailed. Objective: To investigate the effects of three relevant UFAs, namely Docosahexaenoic Acid (DHA), Arachidonic Acid (AA), and Oleic Acid (OA) (ω3, ω6, and ω9, resp.ctively), in the develop.ent of arthritis using a murine model of Collagen-Induced Arthritis (CIA). Methods: DBA-1 mice were immunized with chicken collagen typ. II (CII) and were subsequently treated with ω-UFAs for 53 days. dexamethasone (deXA) was used as a p.sitive anti-inflammatory agent. The effect of the treatments was evaluated through several p.rameters: inflammation indices, antibody levels, cell p.olifera- tion, and histop.thological findings. Results and Conclusion: The anti-inflammatory effect of the tested substances was inversely correlated with the histop.thological findings: a greater anti- inflammatory effect was associated with less articular damage. Oleic acid (ω9) was the most efficient anti-inflammatory UFA, followed by DHA and then AA. deXA comp.etely inhibited the develop.ent of arthritis, whereas the untreated CII-immunized mice develop.d the most severe articular damage. DBA-1 mice with CII-induced arthritis constitute an adequate model for the study of arthritis and its treatment.
