Host Active Defense Responses Occur within 24 Hours after Pathogen Inoculation in the Rice Blast System

Phenotypical, cytological and molecular responses of rice to the fungus magnaporthe grisea were studied using rice cultivars and lesion mimic plants. The cultivar Katy was susceptible to several virulent m. grisea isolates, and a Sekiguchi like-lesion mimic mutant of Katy （LmmKaty） showed enhanced resistance to these isolates. Lesion mimic phenotype of LmmKaty was rapidly induced by virulent m. grisea isolates or by avirulent ones only at high levels of inoculum. Autofluorescence （a sign of an active defense response） was visible under ultraviolet light 24 h after localized inoculation in the incompatible interaction, whereas, not evident in the compatible interaction. Autofluorescence was also observed in LmmKaty 20 h after pathogen inoculation, indicating that rapid cell death is a mechanism of LmmKaty to restrict pathogen invasion. Rapid accumulations of defense related （DR） gene transcripts, phenylalanine ammonia lyase and β-glucanase, were observed beginning at 6 h and were obvious at 16 h and 24 h after inoculation in an incompatible interaction. Rapid transcript accumulations of PR-1 and chitinase had occurred by 24 h after inoculation in an incompatible interaction. Accumulations of these transcripts were delayed in the compatible interaction. These results indicate that host active defense responses occur 24 h after pathogen inoculation and that LmmKaty exhibits enhanced resistance to m. grisea. It is suggested that the autofluorescence and expression of the DR genes after heavy inoculation are important cytological and molecular markers respectively for early determination of the host response to m. grisea in the rice blast system.

AIm To investigate the impact of medication beliefs, illness perceptions and quality of life on medication adherence in people with decompensated *** One hundred adults with decompensated cirrhosis completed a structured questionnaire when they attended for routine outpatient hepatology review. measures of self-reported medication adherence(morisky medication Adherence Scale), beliefs surrounding medications(Beliefs about medicines Questionnaire), perceptions of illness and medicines(Brief Illness Perception Questionnaire), and quality of life(Chronic Liver Disease Questionnaire) were examined. Clinical data were obtained via patient history and review of medical records. Least absolute shrinkage and selection operator and stepwise backwards regression techniques were used to construct the multivariable logistic regression model. Statistical significance was set at alpha = *** medication adherence was " High " in 42 % o f participants, "medium" in 37%, and "Low" in 21%. Compared to patients with "High" adherence, those with "medium" or "Low" adherence were more likely to report difficulty affording their medications(P ment helpfulness(P = 0.003) and stronger medication concerns relative to medication necessity beliefs(P = 0.003). Use of epoxy as an adhesive is a common practice. The most common applications are permanent sealants. Epoxies have a wide range of operating temperatures, and are very resistance to adhesive failure. When a need to remove this adhesive arises, it is not always easily accomplished especially if the part has excessive adhesive. To maintain fidelity of the parts attached by epoxy, a project evaluating several methods of epoxy removal was conducted. methods evaluated included low wavelength, near-ultraviolet radiation, solvent dissolution, and thermal cycling. The UV method failed to demonstrate a repeatable dissociation. The solvent study did result in dissociation of bonds, but introduced chemicals that could make subsequent chemical analysis of parts suspect. Thermal cycling showed a high repeatability for dissociation of bonds and may prove to be relatively inexpensive to implement.

Reconstituted type I collagen fibres have received considerable interest as tendon implant materials due to their chemical and structural similarity to the native *** produced through a semi-continuous extrusion process were cross-linked with different concentrations of the zerolength cross-linker 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide(EDC)in combination with N-hydroxysuccinimide(NHS).Tensile properties of the fibres were considered,along with imaging of both surface structure and fibrillar *** of the fibres to bacterial collagenase was investigated and fibre sections seeded with human tendon cells for biological characterization,including cell adhesion and *** work clearly demonstrated that whilst the concentration of EDC and NHS had no significant effect on the mechanics,a higher concentration was associated with higher collagenase resistance,but also provided a less attractive surface for cell adhesion and proliferation.A lower cross-linking concentration offered a more biocompatible material without reduction in mechanics and with a potentially more optimal degradability.

Graphene growth by low-pressure chemical vapor deposition on low cost copper foils shows great promise for large scale applications. It is known that the local crystallography of the foil influences the graphene growth rate. Here we find an epitaxial relationship between graphene and copper foil. Interfacial restructuring between graphene and copper drives the formation of （nl0） facets on what is otherwise a mostly Cu（100） surface, and the facets in turn influence the graphene orientations from the onset of growth. Angle resolved photoemission shows that the electronic structure of the graphene is decoupled from the copper indicating a weak interaction between them. Despite this, two preferred orientations of graphene are found, ±8° from the Cu[010] direction, creating a non-uniform distribution of graphene grain boundary misorientation angles. Comparison with the model system of graphene growth on single crystal Cu（110） indicates that this orientational alignment is due to mismatch epitaxy. Despite the differences in symmetry the orientation of the graphene is defined by that of the copper. We expect these observations to not only have importance for controlling and understanding the growth process for graphene on copper, but also to have wider implications for the growth of two-dimensional materials on low cost metal substrates.

Alkaptonuria (AKU) is an inherited disorder of tyrosine metabolism caused by lack of active enzyme homogentisate 1,2-dioxygenase (HGD). The primary consequence of HGD deficiency is increased circulating homogentisic acid (HGA), the main agent in the pathology of AKU disease. Here we report the first metabolomic analysis of AKU homozygous Hgd knockout (Hgd−/−) mice to model the wider metabolic effects of Hgd deletion and the implication for AKU in humans. Untargeted metabolic profiling was performed on urine from Hgd−/− AKU (n = 15) and Hgd+/− non-AKU control (n = 14) mice by liquid chromatography high-resolution time-of-flight mass spectrometry (Experiment 1). The metabolites showing alteration in Hgd−/− were further investigated in AKU mice (n = 18) and patients from the UK National AKU Centre (n = 25) at baseline and after treatment with the HGA-lowering agent nitisinone (Experiment 2). A metabolic flux experiment was carried out after administration of 13C-labelled HGA to Hgd−/−(n = 4) and Hgd+/−(n = 4) mice (Experiment 3) to confirm direct association with HGA. Hgd−/− mice showed the expected increase in HGA, together with unexpected alterations in tyrosine, purine and TCA-cycle pathways. metabolites with the greatest abundance increases in Hgd−/− were HGA and previously unreported sulfate and glucuronide HGA conjugates, these were decreased in mice and patients on nitisinone and shown to be products from HGA by the 13C-labelled HGA tracer. Our findings reveal that increased HGA in AKU undergoes further metabolism by mainly phase II biotransformations. The data advance our understanding of overall tyrosine metabolism, demonstrating how specific metabolic conditions can elucidate hitherto undiscovered pathways in biochemistry and metabolism.

Purpose: The purpose of this study was to evaluate the change in corneal wavefront aberrations in young adults who have been fit with multifocal soft contact lenses for myopia progression control. Findings have been analyzed for statistical significance and clinical relevance and compared to reportedly successful Orthokeratology outcomes. methods: The dominant eye of 40 participants (27 women, 13 men;mean age 27.3 ± 3.2 years;range 23 to 39 years) was fit with Proclear multifocal center distance lenses (Coopervision, Pleasanton, USA) having a variety of distance powers and reading additions. Refractive errors were limited to a range of –6.00 D up to +1.00 D of sphere, and no greater than –1.00 D of cylinder. Corneal wavefront measurements were performed over 6 mm diameters with a Zeiss Atlas 9000 corneal topographer (Zeiss meditec, Dublin, USA) prior to, and following lens fitting. Data were converted into rectangular Fourier optics terms m, J0, J45 and RmS values for each reading addition were statistically analyzed. Following evaluation of statistical significance and clinical relevance, results were compared to published data from successful Orthokeratology treatments. Results: Statistically significant changes in higher order aberrations were detected for lenses of all reading additions. Lens groups with higher Add-powers demonstrated stronger changes with increased significance. Final RmS values relating to 2nd, 3rd and 4th Zernike Orders reached clinical significance with a wavefront error of 0.10 μm, the equivalent of 0.25D. moreover, as Add-powers increased, 3rd and 4th order aberrations likewise showed an increase. Pre-fitting astigmatism values accounted for the highest recorded aberrations and remained predominantly unchanged. Conclusion: Proclear multifocal center-distance contact lenses were found to increase higher order wavefront aberrations in a manner dependent on their Add-power.
