Identification of sumoylated targets in proliferating mouse spermatogonia and human testicular seminomas

Spermatogenesis is regulated by a complex network of posttranslation ***(a modification by small ubiquiti like modifiers,or SUMO proteins)was identified as an importa nt cellular event in d iff ere nt cell *** proteins are highly expressed in the testis,and their role during spermatogenesis has begun to be *** the important role of sumoylation in the regulation of mitosis and cancer progress!on in other tissues,the aim of the current study was to identify the targets of SUMO in proliferating mouse spermatog onia and huma n semi noma tissues and to in itially exami ne the level of sumoylation in relation to the proliferative activity of the *** freshly purified spermatogonia and Cl8-4 spermatogonia cell line,mass spectrometry analysis identified several SUMO targets implicated into the proliferation of spermatogonia(such as heat shock protein 60[HSP60]and prohibitin).Tissue array and western blot approaches showed that SUMO expression is a prominent feature of human seminomas and that the proliferative activity of the tumor tissues was positively correlated with the level of SUMO *** regulation of sumoylati on with si-RnA was not sufficie nt to sign ificantly affect the proliferation of C18-4 spermatogonia;however,SUMO overexpression in creased the proliferation rate of the *** data suggest that cells are more sensitive to an elevated level of SUMO,and that this situation may lead to an upregulated cellular proliferation and,possibly,*** spectrometry analysis identified around a hundred SUMO targets in seminoma ***,many of the identified proteins(such as proliferating cell nuclear antigen[PCnA],DnA topoisomerase 2-alpha[Top2A],prohibitin,14-3-3 protein,and others)were implicated in oncogenic transformation and cancer progression.

Genetic basis for body size variation between an anadromous and two derived lacustrine populations of threespine stickleback Gasterosteus aculeatus in southwest Alaska

Body size is a highly variable trait among geographically separated populations. Size-assortative reproductive isolation has been linked to recent adaptive radiations of threespine stickleback （Gasterosteus aculeatus） into freshwater, but the genetic basis of the commonly found size differ- ence between anadromous and derived lacustrine sticklebacks has not been tested. We studied the genetic basis of size differences between recently diverging stickleback lineages in southwest Alaska using a common environment experiment. We crossed stickleback within one anadromous （naknek River） and one lake （Pringle Lake） population and between the anadromous and two lake populations （Pringle and JoJo Lakes）, and raised them in a salinity of 4-6 ppt. The F1 anadromous and freshwater forms differed significantly in size, whereas hybrids were intermediate or exhibited dominance toward the anadromous form. Additionally, the size of freshwater Fls differed from their wild counterparts, with within-population Fls from Pringle Lake growing larger than their wild counterparts, while there was no size difference between lab-raised and wild anadromous fish. Sexual dimorphism was always present in anadromous fish, but not in freshwater, and not always in the hybrid crosses. These results, along with parallel changes among anadromous and fresh- water forms in other regions, suggest that this heritable trait is both plastic and may be under di- vergent and/or sexual selection.

In addition to landscape changes,urbanization also brings about changes in environmental factors that can affect *** the common referral in the published literature to multiple environmental factors such as light and noise pollution,there is a gap in knowledge about their combined *** developed a multidimensional environmental framework to assess the effect of urbanization and multiple environmental factors(light,noise,and temperature)on life-history traits and breeding success of Barn Swallows(Hirundo rustica)across rural to urban gradients in four locations spanning over 2500 km from north to South *** a single breeding season,we measured these environmental factors nearby nests and quantified landscape urbanization over a 1km~*** then analysed the relationships between these multiple environmental factors through a principal component analysis and conducted spatially explicit linear-mixed effects models to assess their effect on lifehistory traits and breeding *** were particularly interested in understanding whether and how Barn Swallows were able to adapt to such environmental conditions associated with *** results show that there is significant variation in the exposure to environmental conditions experienced by Barn Swallows breeding across urbanization gradients in *** changes and their effects are complex due to the behavioural responses ameliorating potential negative effects by selecting nesting sites that minimize exposure to environmental ***,significant relationships between landscape urbanization,exposure to environmental factors,and life-history traits such as laying date and clutch size were ***,the impact on breeding success was,at least in our sample,negligible,suggesting that Barn Swallows are extremely adaptable to a wide range of environmental features.
