Dietary substitution of soybean oil with coconut oil in the absence of dietary antibiotics supports growth performance and immune function in nursery and grower pigs

Background:We hypothesized that supplementation of nursery and grower pig diets with coconut oil in the absence of antibiotics would yield maintenance of glucose homeostasis,growth performance,and immune function similar to what is achieved with nursery and grower pig diets containing *** received the same base treatment diets from d24(weaning)to d71 of age and had blood and fecal samples collected on d24,d31,d45 and d71 for measurement of whole blood glucose,serum insulin,cortisol and cytokines,and fecal *** had weekly weights and daily feed consumption measured throughout the *** were euthanized at d71 and subcutaneous fat and ileal contents were collected for assessment for fatty acids and microbiome,*** treatments consisted of 2%soybean oil plus antibiotics(ABX;n=22),2%soybean oil without antibiotics(NABX;n=22),and 2%coconut oil without antibiotics(cOc;n=22).Statistical analysis examined the effect of diet within each timepoint using a repeated measures ***:Pigs fed cOc diet had decreased serum insulin levels,maintained feed intake,feed conversion and weight gain,and,based on serum cytokines and fecal microbiome,were immunologically similar to ABX-fed ***,NABX-fed pigs performed similarly to the ABX-fed pigs in all parameters except for serum ***,there was no difference in the incidence of diarrhea between any of the diet ***:This study demonstrates that dietary antibiotics are not necessary to maintain growth performance in nursery and grower ***,dietary antibiotics appear to modulate circulating cytokine *** coconut oil is neither harmful nor helpful to growth performance or immune function in nursery and grower pigs but does modulate serum insulin ***,while coconut oil fed at 2%by weight is a suitable substitute for dietary antibiotics,this study suggests that no substitute for dietary antibiotics is needed at all.

Development of mucoadhesive sericin/alginate particles loaded with ibuprofen for sustained drug delivery

A sericin/alginate blend was successfully applied as a matrix for incorporation of *** provided a sustained release formulation,which could improve the therapeutic efficacy of ibuprofen and patient adherence to *** increased the proportion of drug incorporated into the particles (i.e., the drug incorporation efficiency),with incorporation rates between 73.01%±1.70%and 94.15%±4.21%. Environmental drivers of seasonal shifts in abundance of wild pigs(Sus scrofa)in a tropical island environment

Gender differences of lower urinary tract symptoms in older chinese Americans

Objective To describe whether or not there are gender differences in lower urinary tract symptoms(LUTS)prevalence and risk factors in community-dwelling older chinese *** We performed a secondary analysis of a prospective cross-sectional population-based survey of chinese Americans aged 60 years and older between January 2011 and December 2013 in English,Mandarin,cantonese,Taishanese,or Teochew.A clinical review of systems was used to assess LUTS,which included urinary frequency,urgency,burning and/or pain,blood in urine,and urinary *** Of the total 3157 people queried,42%were men and 58%were *** men reported LUTS compared to women(32.9%vs.28.6%,p=0.01).In a multivariable analysis,female gender(adjusted odds ratio[aOR]0.60,95%confidence interval[cI]0.49-0.73),being married(aOR 0.79,95%cI 0.65-0.97),and smoking(aOR 0.66,95%cI 0.49-0.88)were found to be protective,while traditional chinese medicine use(aOR 1.51,95%cI 1.28-1.78),heart disease(aOR 1.54,95%cI 1.24-1.91),and anxiety(aOR 1.69,95%cI 1.25-2.28)were most strongly associated with increased odds of *** examining genders separately,being married was found to be protective only in ***,unique factors found in men were hypertension,heart disease,and practice of Tai *** In this large population-based study,LUTS were more prevalent in older chinese American men than *** also found gender-specific factors that influenced the odds of reporting LUTS;however,traditional chinese medicine use was the only factor that was shared by both *** longitudinal investigations are needed to elucidate these underlying mechanisms to provide evidence-based and culture-specific guidelines for this rapidly growing population.

Omeprazole induces gastric transmucosal permeability to the peptide bradykinin

AIM:To investigate omeprazole-induced transepithelial gastric leak and its effects on the permeability of the peptides bradykinin and ***:Rat gastric corpus tissue was isolated and mounted in an Ussing chamber apparatus to evaluate the permeability of 3H-bradykinin,3H-oxytocin,and 14cEDTA in the presence or absence of *** chromatography was performed to identify any metabolic breakdown products of the peptides resulting from permeation through the gastric tissue,and thereby calculate the true flux of the ***:The flux rate ofintact 3H-bradykinin increased substantially after omeprazole addition (109.5%) compared to the DMSO vehicle control (14%).No corresponding change in flux ofintact 3H-oxytocin was observed under the same conditions (11.9% and 6.4% in the DMSO-and omeprazole-treated conditions,respectively).After exposure to omeprazole,the flux rate of 14c-EDTA also increased dramatically (122.3%) compared to the DMSO condition (36.3%).cONcLUSION:The omeprazole-induced gastric leak allows for transmucosal permeability to charged molecules as well as *** induced leak will allow certain peptides to permeate.

Role of stem cell therapies in treating chronic wounds:A systematic review

BAcKGROUND The impairment of cutaneous wound healing results in chronic,non-healing wounds that are caused by altered wound environment oxygenation,tissue injury,and permissive microbial *** modalities for the treatment of these wounds inadequately address the complex changes involved in chronic wound ***,stem cell therapies have emerged as a potential therapeutic modality to promote cutaneous regeneration through trophic and paracrine *** To investigate current literature regarding use of stem cell therapies for the clinical treatment of chronic,non-healing *** PubMed,EMBASE,cochrane Library,Web of Science,and Scopus were queried with combinations of the search terms“mesenchymal stem cells,”“adult stem cells,”“embryonic stem cells,”“erythroid precursor cells,”“stem cell therapies,”and“chronic wounds”in order to find relevant articles published between the years of 2000 and 2019 to review a 20-year *** lists from the articles were reviewed to identify additional pertinent *** manuscripts(reviews,case reports/series,retrospective/prospective studies,and clinical trials)were evaluated by the authors for their depiction of clinical stem cell therapy *** were extracted from the articles using a standardized collection *** A total of 43 articles describing the use of stem cell therapies for the treatment of chronic wounds were included in this *** stem cell therapies have been explored in in vitro and in vivo applications in the past,recent efforts are geared towards assessing their clinical role.A review of the literature revealed that adipose-derived stem cells,bone marrow-derived stem cells,bone marrowderived mononuclear cells,epidermally-derived mesenchymal stem cells,fibroblast stem cells,keratinocyte stem cells,placental mesenchymal stem cells,and umbilical cord mesenchymal stem cells have all been employed in the treatment of chronic wounds of various etiolo

The antibody repertoire of Bos taurus is characterized by a subset of variable heavy(VH)chain regions with ultralong third complementarity determining regions(cDR3)which,compared to other species,can provide a potent response to challenging antigens like HIV *** unusual cDR3 can range to over seventy highly diverse amino acids in length and form uniqueβ-ribbon‘stalk’and disulfide bonded‘knob’structures,far from the typical antigen binding *** genetic components and processes for forming these unusual cattle antibody VH cDR3 are not well *** we analyze sequences of Bos taurus antibody VH domains and find that the subset with ultralong cDR3 exclusively uses a single variable gene,IGHV1-7(VHBUL)rearranged to the longest diversity gene,*** eight nucleotide duplication at the 3′end of IGHV1-7 encodes a longer V-region producing an extended Fβ-strand that contributes to the stalk in a rearranged cDR3.A low amino acid variability was observed in cDR1 and cDR2,suggesting that antigen binding for this subset most likely only depends on the *** a novel,potentially AID mediated,deletional diversification mechanism of the *** VH ultralong cDR3 knob was discovered,in which interior codons of the IGHD8-2 region are removed while maintaining integral structural components of the knob and descending strand of the stalk in *** deletions serve to further diversify cysteine positions,and thus disulfide bonded ***,both germline and somatic genetic factors and processes appear to be involved in diversification of this structurally unusual cattle VH ultralong cDR3 repertoire.

The effects of glycyrrhizic acid and glabridin in the regulation of cXcL5 inflammation gene on acceleration of wound healing

Objective: To evaluate the anti-inflammatory property of both glycyrrhizic acid(GA)and glabridin in reducing inflammation to accelerate wound regeneration on 3T3-L1 and NIH-3T3 fibroblast cell ***: cell proliferation and viability assay(MTT assay), scratch wound healing assays,and quantitative real-time PcR were conducted to investigate the effects on cell proliferation,cell migration, and expression of cXc chemokine ligand 5 inflammation gene ***: Results showed that at a low concentration of 1 × 10^(-8)mol/L, glabridin down regulated cell proliferation in NIH-3T3 significantly, suggesting its involvement in ERK1/2 signaling pathway. GA and glabridin significantly accelerated cell migration through wound healing in both 3T3-L1 and NIH-3T3 and significantly down regulated the expression of cXc chemokine ligand 5 in 3T3-L1 at concentration 1 × 10^(-8)mol/L,indicating the possible involvement of nuclear factor-k B and cyclooxygenase 2 transcriptions ***: Both GA and glabridin can serve as potential treatment for chronic inflammatory disease, and glabridin as an oncogenic inhibitor due to its anti-proliferative property.

The Utility of 1000 - 500 mb Thickness and Weather Type as a Rain-Snow Divide: A 30-Year Study at Albany, NY

Winter synoptic conditions that produce snowfall with bitterly cold temperatures create both social and economic hazards in the capital city of Albany, NY. Sometimes these systems are forecasted in error to produce rain or mixed precipitation. It is beneficial for meteorologists to better understand the commonly used 5400 and 1300 GPM line to better forecast rain versus snow events. Other studies have looked into the use of the 5400 GPM (540 dm) line but none have assessed the validity of this boundary with respect to weather type characterization at Albany. This study aims to determine the reliability of the widely referenced guides for depicting the rain-snow line, and improve forecast aids for the vertical atmosphere during winter precipitation events. The mean daily 500, 850, 925 and 1000 mb heights and weather type frequency of the Spatial Synoptic classification between November and March of 1980 - 2012 are analyzed. Results indicate that the standard vertical boundaries are inaccurate indicators of a rain versus snow event in Albany. More reasonable rain-snow cut offs for the 1000 - 500 and 1000 - 850 mb thicknesses are 5222 and 1262 GPM. For the 1000 - 925 mb level, 606 GPM is a helpful aid of identifying the rain-snow boundary. Further scrutinizing by weather type indicates that the rain-snow boundary also varies depending on what air mass/weather type is present on a given day. For instance, when the most prominent weather type is observed over Albany (Dry Polar), at the 1000 - 850 mb and 1000 - 500 mb layers, a boundary of 1242 GPM and 5152 GPM is found to be most representative. Results indicate only for the rarest of winter weather types observed over Albany, Moist Tropical, are the standard cut offs useful. Determining the reliability of this precipitation indicator at a specific station, like Albany, could enable meteorologists in other regions of the country to draw parallels between weather type, precipitation, and thickness in their forecast zones.
