Laser additive manufacturing of Zn porous scaffolds: Shielding gas flow, surface quality and densification

Zn based metals have exhibited promising prospects as a structural material for biodegradable applications. Pure Zn porous scaffolds were produced by laser powder bed fusion(LPBF) based on data files of designing and CT scanning. massive Zn evaporation during laser melting largely influenced the formation quality during LPBF of Zn metal. The metal vapor in processing chamber was blown off and suctioned out efficiently by an optimized gas circulation system. Numerical analysis was used to design and testify the performance of gas flow. The surface of scaffolds was covered with numerous particles in different sizes. Processing pores occurred near the outline contour of struts. The average grain size in width was8.5m, and the hardness was 43.8 HV. Chemical plus electrochemical polishing obtained uniform and smooth surface without processing pores, but the diameter of struts reduced to 250 αm from the design value 300 m. The poor surface quality and processing pores were resulted by the splashing particles included spatters and powders due to the recoil force of evaporation, and the horizontal movement of liquid metal due to overheating and wetting. The insufficient melting at the outline contour combined with good wetting of Zn liquid metal further increased the surface roughness and processing pores.

Tumor–stromal cross-talk modulating the therapeutic response in pancreatic cancer

Background: Pancreatic ductal adenocarcinoma(PDAC) is a highly malignant solid tumor with a dismal prognosis. The stroma component makes up to 90% of the tumor mass and is thought to be one of the main reasons for the tumor’s high chemoresistance. Cancer associated fibroblasts(CAFs) have previously been identified to be the key stromal players. This is the first time we provide detailed in vitro experiments investigating tumor–stromal interactions when exposed to three well-known chemotherapeutic agents. methods: monocultures, indirect and direct co-cultures of two PDAC cell lines(AsPC and Panc-1) and six primary patients derived CAFs were treated with gemcitabine, nab-paclitaxel and the γ-secretaseinhibitor(GSI) DAPT. The cell viability of each component was measured with XTT. Finally, IL-6 concentrations of the supernatants were analyzed. Results: On the contrary to PDAC cell lines, CAF monocultures hardly responded to any treatment which suggested that stroma(CAFs) itself is more resistant to standard chemo-treatments than the epithelial cancer cells. moreover, only a weak chemotherapeutic response was observed in direct co-cultures of cancer cells with CAFs. A change in the morphology of direct co-cultures was accompanied with the chemoresistance. CAFs were observed to build cage-like structures around agglomerates of tumor cells. High levels of IL-6 were also associated with a reduced response to therapy. Indirect co-cultures make the tumor–stromal interaction more complex. Conclusions: CAFs are highly chemoresistant. Direct cell–cell contact and high levels of IL-6 correlate with a high chemoresistance.

TLR3-dependent immune regulatory functions of human mesangial cells

In glomerulonephritis, the migration of inflammatory cells into the glomerulus is an important step in disease initiation and progression. The viral receptor Toll-like receptor 3 （TLR3） is known to play a role in virus-associated glomerulonephritis. Based on this knowledge, this study aimed to define the effects of the TLR3 ligand polyriboinosinic：polyribocytidylic acid （poly（hC）） on the expression of adhesion molecules and macrophage colony-stimulating factor （m-CSF） on resident glomerular cells. Experiments in mCs demonstrated that the activation of viral receptors by poly（I.C） leads to a time- and dose-dependent induction of intercellular adhesion molecule 1 （ICAm-1）, vascular cell adhesion molecule 1 （VCAm-1） and m-CSF at both the mRNA and protein levels; these results were confirmed by incubating mCs with HCV RNA. As shown in knockdown experiments, this effect is specifically mediated by TLR3. The prestimulation of mCs with proinfiammatory cytokines increases the effects of poly（hC）, except for its induction of VCAm-I. Tumor-necrosis factor （TNF）-a, likewise, induces ICAm-1, VCAm-1 and m-CSF, and amplifies the mesangial response to poly（hC）. These results were confirmed by incubating mCs with HCV RNA. We thus provide evidence that human mCs represent a potential target of the leukocytes and monocytes that infiltrate the glomerulus in viral disease-associated GN, highlighting the possibility that mCs may act as resident antigen-presenting cells.

Summary: The purpose of this study was to evaluate relative regional blood volume (rCBV) in meningioma and compare the utility of reconstruction using both gradient-echo sequence and echo-planar-imaging (EPI) *** patients with meningiomas were studied on a Siemens 1.5-T scanner. During the gradient-echo sequence (n=12) and EPI sequence (n=6), a bolus ^(0.2 mmol/kg) of Gd-DTPA was injected mechanically with a flow rate of 5 ml/second. Image processing of dynamic data was performed on a pixel-by-pixel basis. The ratio of tumor rCBV/gray matter rCBV was 3.01±1.18 (3.07 ± 1.39 in gradient sequence and 2.84 ± 0.94 in EPI sequence). The ratio of gray matter/ white matter as the reference tissue had a mean of 2.79 ± 0.76 using the FLASH sequence, and a mean of 3.04 ± 1.31 using EPI. These differences were not statistically signifcant (P>0.5, t-test). According to the ratio of tumor rCBV/gray matter rCBV, a mean value, 14.5 ml/100 g, of rCBV in meningiomas was calculated. Compared with gray matter, increased inhomogeneous rCBV was observed in meningioma. Based on the two different sequences, no bias can be observed in our rCBV reconstruction.

In the pursuit of advancing imidazolium-based energetic ionic liquids (EILs),the current study is devoted to the synthesis and characterization of 1,3-dibutyl-imidazolium azide ([BBIm][N_(3)]),as a novel member in this ionic liquids *** chemical structure of this EIL was rigorously characterized and confirmed using FTIR spectroscopy,1D,and 2D-NmR *** thermal behavior assessment was conducted through DSC and TGA *** analysis revealed an endothermic glass transition at T_(g)=-61℃,followed by an exothermic degradation event at T_(onset)=311℃.Similarly,TGA thermograms exhibited a one-stage decomposition process resulting in 100% mass loss of the ***,the short-term thermal stability of the azide EIL was investigated by combining the non-isothermal TGA data with the TAS,it-KAS,and VYA/CE isoconversional kinetic ***,the Arrhenius parameters(E_(a)=154 kJ·mol^(-1),Log(A/s^(-1))=11.8) and the most probable reaction model g(a) were *** observed high decomposition temperatures and the significantly elevated activation energy affirm the enhanced thermal stability of the modified *** findings revealed that[BBIm][N_(3)]EIL can be a promising candidate for advanced energetic material application.

The application of polymers to replace oleylamine(OLA)and oleic acid(OA)as ligands for perovskite nanocrystals is an effective strategy to improve their stability and durability especially for the solution-based ***,we report a mechanosynthesis of lead bromide perovskite nanoparticles(NPs)stabilized by partially hydrolyzed poly(methyl methacrylate)(h-PmmA)and highmolecular-weight highly-branched poly(ethylenimine)(PEI-25K).The as-synthesized NP solutions exhibited green emission centered at 516 nm,possessing a narrow full-width at half-maximum of 17 nm and as high photoluminescence quantum yield(PL QY)as 85%,while showing excellent durability and resistance to polar solvents,e.g.,*** colloids of polymer-stabilized NPs were directly processable toform stable and strongly-emitting thin films and solids,making them attractive as gain ***,the roles of h-PmmA and PEI-25K in the grinding process were studied in *** h-PmmA can form micelles in the grinding solvent of dichloromethane to act as size-regulating templates for the growth of *** PEI-25K with large amounts of amino groups induced significant enrichment of PbBr_(2)in the reaction mixture,which in turn caused the formation of CsPb_(2)Br_(5)-mPbBr_(2)and CsPbBr_(3)-Cs_(4)PbBr_(6)-nCsBr *** presence of CsPbBr_(3)-Cs_(4)PbBr_(6)-nCsBr NPs was responsible for the high PL QY,as the Cs_(4)PbBr_(6)phase with a wide energy bandgap can passivate the surface defects of the CsPbBr_(3)*** work describes a direct and facile mechanosynthesis of polymer-coordinated perovskite NPs and promotes in-depth understanding of the formation and phase conversion for perovskite NPs in the grinding process.

ER stress-induced aggresome trafficking of HtrA1 protects against proteotoxicity

High temperature requirement A1 （HtrA1） belongs to an ancient protein family that is linked to various human disorders. The pre- cise role of exon 1-encoded N-terminal domains and how these influence the biological functions of human HtrAZ remain elusive. In this study, we traced the evolutionary origins of these N-terminal domains to a single gene fusion event in the most recent common ancestor of vertebrates. We hypothesized that human HtrA1 is impticated in unfotded protein response. ｜n highly secre- tory cells of the retinal pigmented epithelia, endoplasmic reticulum （ER） stress upregulated HtrA1. HtrA1 co-localized with vimen- tin intermediate filaments in highly arborized fashion. Upon ER stress, HtrA1 tracked along intermediate filaments, which collapsed and bundled in an aggresome at the microtubule organizing center. Gene silencing of HtrA1 altered the schedule and amplitude of adaptive signaling and concomitantly resulted in apoptosis. Restoration of wild-type HtrA1, but not its protease inactive mutant, was necessary and sufficient to protect from apoptosis. A variant of HtrA1 that harbored exon 1 substitutions dis- played reduced efficacy in rescuing cells from proteotoxicity. Our results illuminate the integration of HtrA1 in the toolkit of mam- malian cells against protein misfolding and the implications of defects in HtrA1 in proteostasis.

BACKGROUND Pancreatic panniculitis is an extremely rare condition associated with different underlying pancreatic disorders and characterized by subcutaneous fat necrosis induced by elevated serum lipase *** lesions usually affect the lower extremities and may precede abdominal symptoms of pancreatic *** cell carcinoma(ACC)of the pancreas is a rare pancreatic neoplasm,accounting for only 1%-2%of pancreatic tumors in *** present the case of a 72-year-old man with ACC of the pancreatic head and synchronous liver *** the primary tumor and liver metastases were *** lipase was elevated before surgery and decreased to normal *** serum lipase levels at follow-up led to the diagnosis of hepatic *** disease progression was then accompanied by pancreatic panniculitis,with subcutaneous fat necrosis and acute *** the best of our knowledge,only 4 cases have been reported in the literature and each showed a similar association of serum lipase levels with pancreatic panniculitis and progression of *** Clinical symptoms and progression of ACC may correlate with serum lipase levels,suggesting potential usefulness as a follow-up biomarker.
