Programmed death-ligand-1(PD-L1)plays a crucial role in the suppression of the antitumor immune response upon interaction with programmed cell death protein-1(PD-1)on cytotoxic T ***-L1 is constitutively expressed on antigen-presenting cells or tumor ***,PD-L1 has been detected by immunohistochemistry in a variety of tumors,including malignant pleural mesothelioma(MPM),in which it has emerged as a predictive biomarker for PD-1/PD-L1 immune checkpoint blockers(ICBs).1,2 PD-L1 expression on tumor cells can be upregulated by several factors including the interferon gamma(IFN-γ)cytokine produced by tumor-infiltrating lymphocytes(TILs).3 In MPM cells,it has been reported that IFN-γupregulates PD-L1 mRNA expression due to the activation of the interferon regulatory factor 1(IRF1)transcription factor,4 but no data have been shown regarding cell surface PD-L1 that is functionally relevant for its contact with PD-1-positive cells.

Head geometric morphometrics of two Chagas disease vectors from Venezuela

Triatominae species are considered the main vectors of Chagas disease or American Trypanosomiasis. In Venezuela, the principal vectors are Rhodnius prolixus （Stal, 1959） and Triatoma maculata （Erichson, 1848）, which are belonged to the tribe Rhodniini and Triatomini, respectively. The head conformation and size development of these species can reflect ontogenetic changes which contribute with the vectors biology studies, as well to support of instars determination. The goal of the paper is to the application of geometric morphometric techniques for describing head conformation and size of instars of these species. We photographed 140 heads in R. prolixus： First instar （I： 16）, second instar （II： 17）, third instar （III： 18）, fourth instar （IV： 21）, fifth instar （V： 21）, adult female （F： 26） and adult males （M： 21）; in T. maculata heads of 136 specimens were photographed,I： 20, II： 17, III： 26, IV： 15, V： 19, F： 20 and M： 19. landmark coordinate （x, y） configurations were registered and aligned by Generalized Procrustes Analysis. Covariance Analyses were implemented with proportions of re-classified groups and MANOVA. Statistical analyses of variance found not significant differences in head isometric size （Kruskal-Wallis） among IV and V instars in both species. The a posteriori re-classification was almost perfect in R. prolixus （82%） and T. maculata （86%）; the main head differences occurs in antenniferous tubercles, postocular and preocular. Our study using quantitative tools for describing the shape differences contributes to explain the morphology variability and development of Chagas disease vectors.

The design and construction of an experimental solar hybrid vehicle based on the combination of photovoltaic solar energy as the main source of electricity and electric power supplied by a generator activated by the driver＇s pedaling is introduced. The vehicle has a battery to store the energy provided by both systems. The development was motivated by a latin American solar car race through the Atacama Desert in Chile and the initiative to promote the use of clean energy for transport. A general description of the vehicle, its energetic aspects and experimental results are presented.

Association of Serum Antioxidant Enzymes and Nervous Tissue Markers in Hypertensive Patients

Background and Purpose: Hypertension has serious effects on cerebral blood vessels. Oxidative stress seems to be implicated in blood pressure elevation, through increased reactive oxygen species and/or decreased antioxidant capacity. Recently blood markers indicating damage to the central nervous system were reported to be increased in hypertensive patients. However, it is unknown whether antioxidant capacity is related to these changes. This study was designed to explore if the concentration of blood markers for nervous tissue damage was associated to antioxidant capacity in hypertensive patients. Methods: Twenty hypertensive patients and 23 healthy controls were studied. They were paired by age, sex, ethnicity, or risk factors. Serum neuron specific enolase (NSE) and S100 calcium binding protein B (S100B) were measured as nervous tissue damage markers, as well as the activity of antioxidant enzymes (catalase, glutathione peroxidase, glutathione reductase and gamma-glutamyltransferase). Results: Serum neuronal specific enolase (NSE) and S100 calcium binding protein B (S100B) concentrations determined by immunoassay were significantly increased in patients vs. controls. The activities of antioxidant enzymes measured by spectrophotometry showed that plasmatic catalase and erythrocytic glutathione peroxidase were significantly increased in patients, but erythocytic catalase was decreased. Gamma-glutamyltransferase activity was significantly correlated with S100B in hypertensive patients, while erythrocytic catalase activity was decreased in subjects with higher NSE levels. Conclusion: This preliminary investigation suggested that antioxidant status might be modulated through changes in antioxidant enzymatic activity in hypertensive patients. The association of some of these changes with peripheral markers of damage to the central nervous system could indicate that the increased levels of these proteins in hypertension are partly related to oxidative stress.

Clinical and radiographic Evaluation of Cattle Tail before and after the <i>Vaquejada</i>race

The vaquejada race is a sport with animals which has the greatest socio-economic impact in Brazil and is responsible for generating an intense cultural and economic movement. However, this activity has been the subject of questions about well-being involving the environmental and mental comfort conditions offered to equine and cattle interactions. It is necessary to perform technical evaluations of the cattle involved on the basis of scientific study to better understand the processes of sport and, with this, promote their evolution on good welfare practices in the 21st century. Therefore, to test the hypothesis that the use of the official rules of the vaquejada where cattle use a protective harness on the tail protects the cattle’s tail from injuries in this region that compromise health. Before, during and after races, a study was developed that aimed to perform a general and radiological clinical evaluation in the proximal coccygeal region in cattle that ran in a modern cattle stall. Forty male bovines were evaluated (average: 2 year-old and 420 kg). Oxen were subjected to clinical and radiological examination before and after racing with official rules. Results of the clinical and radiological evaluations performed by veterinarians, did not observe clinical occurrences before and after the races in the 40 cattle evaluated. The increase in heart and respiratory rates was a reflection of temporary management stressors, as the animals returned to the feeding/rumination/interaction pattern after running in the rest pen. The radiographic findings in the tail, represented by the reduction of the intervertebral space and areas of subcutaneous radiolucency, suggest technical artifacts derived from

Proteomic analysis for developing new biomarkers of hepatocellular carcinoma

AIM: To identify new markers of hepatocellular carcinoma (HCC) using a proteomic analysis. METHODS: Patients with liver cirrhosis of the three most frequent etiologies: hepatitis C virus, hepatitis B virus and alcoholic liver disease, were included in the study. The samples were analysed by 2D-electrophoresis in order to determine the differential protein expression. The proteins were separated according to the charge in immobilized pH 3-10 gradient strips and then by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Proteins of interest were excised, digested with trypsin and the resulting peptides were separated and identified. RESULTS: Three differentially expressed apolipoproteins (Apo) were identified based on the protein profile using proteomic techniques: Apo-A1, Apo-A4 and Apo-E. Apo-A4 levels were significantly lower in HCC than in non-HCC patients regardless of etiology (P lated to HCC diagnosis (P rating characteristic (ROC) curve including both Apo-A4 and Apo-A1 showed an area under the ROC of 0.944 (P < 0.001), a sensitivity of 0.89 and a specificity of 0.81 for diagnosis of HCC. CONCLUSION: Apo-A4 and Apo-A1 may be used clinically as biomarkers of HCC with a high sensibility and specificity. These findings may provide additional insights into the mechanism of HCC development and progression.

Esterase Activities and Biochemical Properties of lactic Acid Bacteria Isolated from Goat＇s Milk Cheese in Argentina

Twenty-two lactic acid bacteria （laB） strains isolated from Argentinean goat dairy products were evaluated for its biochemical properties and esterase activities relevant to flavor development. Streptococcus thermophilus （UNSE314）, lactobacillus （L.） delbrueckii subsp, bulgaricus （UNSE309）, L. rhamnosus （UNSE308）, L. plantarum （UNSE287, UNSE316, UNSE317） and Pediococcus pentosaceus （UNSE315） strains presented high acidifying activity. All strains tested metabolized citrate and produced diacetyl-acetoin in goat milk. Based on these results, 10 strains with the best performance in diverse technological properties were selected to determine esterolytic activity. In all evaluated strains, esterase specific activity （ESA） was detected on ct-naphthyl （ct-NA） acetate and 13-naphthyl （[3-NA） acetate, propionate, eaprylate and ct-NA butyrate. No activity was detected on [3-NA laurate. The highest values were detected when using a-NA instead of fI-NA derivatives as substrate. In Pediocoecus strains, wide variability in ESA were observed, which were species- and strain-specific. These results allow us to select strains with biochemical properties and esterase activities to design starter and adjunct cultures that contribute to flavor development during cheese ripening, thus preserving the typical organoleptic characteristics of Argentinean goat cheeses.
