Background: Mesenchymal stromal cells (MSC) are being tested for the treatment of immune diseases. MSC are present in several adult tissues which milieu may influence MSC behavior particularly under inflammatory conditions. Additionally, culture conditions also can modify cell function or state of activation. Methods: To address the influence of the MSC source on its characteristics, we studied a xenofree, platelet lysate supplemented MSC from dental pulp, adipose tissue and bone marrow, co-cultured with isolated T cells and PBMC subset, and studied the effect of culture animal or human supplements immunomodulatory effect. Results: All three sources were efficient in inhibiting T cells. Among all MSC sources, as also described by others, adipose MSC was capable to significantly induce Treg phenotype and decrease T CD8+. Furthermore, comparing fetal bovine serum and platelet lysate, results demonstrate that platelet lysate alone is capable to induce immunomodulatory phenotype. Additional studies have to be made to elucidate the PL immunomodulatory effect.

Cassava flours are obtained from the roots of Manihot esculenta Crantz and daily fairly consumed by the Brazilian population considering it comes to hum food, and also inserted into food baskets, besides to present low cost and easy access, in addition to being plant easily cultivated by the population. However, the food obtained with cassava may contain hydrocyanic acid, in the form of cyanogenic glycoside when those meals are not cooked properly. The oral toxicity can range 30 - 210 mg/kg body, the average daily consumption per capita in larger rural areas than in urban areas for cassava flour 19.1 g against 4.7 g, respectively. Brazil’s North and Northeast have higher consumption of cassava flour, associated with family monthly monetary income, making this region more vulnerable to poisoning. The objective of this study was to quantify cyanogenic glycosides present in some types of Brazilian cassava foods by means of spectrophotometric technique (reading at a wavelength of 530 nm). For quantitative determination of cyanide content, linamarase hydrolysis was used. The amount of cyanide was determined, and results were: artisan toasted cassava flour: 15 mg/500g, sweet cassava starch: 32.5 mg/500g, artisan dried cassava flour: 37.5 mg/500g, “bijuzada” cassava flour: 60 mg/500g, industrialized, toasted cassava flour: 115 mg/500g, industrialized, raw cassava flour: 140 mg/500g, and wet cassava flour: 225 mg/500g. Considering the chronic ingestion of cyanide present in these foods, they can cause public health problems.

The essential oils from Mentha piperita, Cymbopogon citratus, Rosmarinus officinalis, Peumus boldus and Foeniculum vulgare were extracted by hydrodistillation and characterized and quantified by GC-MS and GC-DIC. The oils induced hemolysis with all the doses evaluated (0.6 to 1.8 μL), and the diameters of the halos varied between 9 and 15 mm. Pre-incubation of P. boldus oil with Bothrops jararacussu venom resulted in potentiation of venom-induced hemolysis (30%) (proteases and phospholipases A2). The essential oil from M. piperita (0.6 μL) inhibited venom-induced hemolysis by 45%, whereas 0.6 μL of R. officinalis oil increased the hemolysis by 20%. For the essential oil from F. vulgare, 100% inhibition of activity (0.6 and 1.2 μL) was observed. The application of C. citratus oil induced hemolysis with all the volumes evaluated. Phospholipase activity induced by the venom was only inhibited (10%) with the 0.6 μL volume of R. officinalis oil. The oils from M. piperita and F. vulgare (1.8 μL) and C. citratus oil (0.6 μL) potentiated the phospholipase activity. The results highlight the need for a broad characterization and regulation of the use of natural products, because they can have therapeutic or toxic actions.

The CBM （chromoblastomycosis） is a disease caused by dematiaceous fungi, which has the species Fonsecaea pedrosoi as main agent. This fungus is found in warm and moist climates, characteristically found in the amazonic lands of the region, where its environmental isolation was once described. This research aimed to identify the healthy population exposed to agent F. pedrosoi in four villages located in the Legal Amazon. In order to clarify the risk factors for allergic immune exposure, a survey was conducted by the technique of delayed skin reaction （IDR） with the metabolic antigen （chromomycin） in 449 healthy individuals. The results showed that 14.9% （67 subjects） were IDR positive, with induration 〉 5 mm. The possibility of commercialization of Moro blood oranges in tropical countries such as Brazil was evaluated to verify whether post-harvest management through storage at low temperatures for a period of 60 days can improve the bioactive properties and quality parameters. Moro blood oranges cultivated in Brazil did not contain significant amounts of anthocyanins at the time of harvesting, but these compounds were activated by post-harvest management through storage at low temperatures (4°C and 8°C) for a period of 60 days. The emergence of the anthocyanins in the juices occurred within a few weeks of storage, but the maximum levels were attained after 60 days and at the temperature of 8°C. Cold storage positively influenced other bioactive compounds such as total phenolic compounds, individual phenolic compounds, β-carotene and the antioxidant activity determined by the sequestration of DPPH free radicals. It did not influence the vitamin C content. In addition, storage significantly altered the color, total acidity and pH of the fruits, but it did not prevent its commercial use. The remaining quality parameters were not influenced. It is possible to commercialize these oranges in Brazil through post-harvest management.

The chemical compositions of the essential oils from the leaves and flowers of Callistemon viminalis and their insecticide and fungitoxic activities were determined. The essential oil was extracted by the hydrodistillation method using a modified Clevenger apparatus. The chemical characterization was performed by GC-MS and GC-FID. The evaluation of the insecticidal activity was performed with the Myzus persicae aphid, and the antifungal potential was determined via the inhibition of the mycelial growth of Alternaria alternata, Fusarium oxysorum and Botrytis cinérea phytopathogenic fungi. The principal components of the essential oils from the leaves and flowers were eucalyptol (84.60% and 61.47%), α-pinene (10.28% and 21.48%) and α-terpineol (2.59% and 2.79%), respectively. The use of a 0.5% concentration of the essential oil from the flowers influenced the preference of aphids and their reproduction. The number of adult aphids decreased within a period of 48 hours in the presence of the essential oil from the leaves. In the test with no chance of choice, the mean number of adults decreased with both oils within 48 hours. No inhibition of the mycelial growth of the A. alternate microorganism was observed in any of the treatments with the two essential oils. However, the growth of F. oxysporum and B. cinerea fungi was inhibited. The mycelial growth of F. oxysporum was inhibited with the concentration of 100 μL·Lˉ1 of the oil from the flowers and 250 μL·Lˉ1 of the oil from the leaves. The inhibition of the growth of B. cinerea was observed at concentrations of 500 and 100 μL·Lˉ1 for the oils from the flowers and leaves, respectively. The essential oils presented insecticidal and antifungal potentials. However, further studies are needed for these oils to be used in integrated pest management.

Importance:Cleft lip and palate(CLP)is globally among the most common childhood *** disorder impacts childhood development,including speech and language,and affects children ***:To analyze child development skills(adaptive fine motor,gross motor,personal-social,and language)in preschool children with isolated CLP compared with children without this ***:The participants included an experimental group of 27 children with isolated CLP and a comparison group of 27 children without CLP aged between 48 and 59 *** groups were evaluated using two instruments:the Denver Developmental Screening Test II(DDST-II)and the Avaliapao do Desenvolvimento da Linguagem(ADL-Language Development Assessment).Data were analyzed by descriptive and inductive analyses,using the Student’s Mest and the Mann-Whitney test,at a significance level of P<***:All children in the comparison group performed within normal standards for their age range in the DDST-II and the *** worst performance in the experimental group was observed in language skills,followed,in declining order,by adaptive fine motor,personal-social,and gross motor as measured by the *** with isolated CLP also performed poorly in receptive,expressive,and global language in the *** statistically significant differences were observed in the experimental group’s scores for the ADL and the ***:Developmental skill levels were below expectations for children of this age with isolated CLP.

The objective of this study was to characterize and verify the in vitro antitumor activity of essential oils (EOs) extracted from the leaves and flowers of Callistemon viminalis. The EOs were extracted by hydrodistillation using a modified Clevenger apparatus. The identification and quantification of constituents were performed on a gas chromatograph coupled to a mass spectrometer and a gas chromatograph with a flame ionization detector. The antitumoral activity was evaluated by a colorimetric assay (MTS) using different cell lines derived from human tumors (breast, lung, glioblastoma, and melanoma). The major constituents of the EOs of leaves and flowers were similar, only quantitative differences being observed. The compounds 1,8-cineole, α-pinene and α-terpineol were found in concentrations of 50.4%, 25.8% and 8.7% in the EOs obtained from the leaves and 48.8%, 24.5% and 3.9% in the EOs obtained from the flowers, respectively. The cytotoxic activity of the EOs was observed only in melanoma cultures (HT144). Cultures treated for 48 h with EOs from leaves and flowers (200 μg·mL-1) reduced the viability by 40% and 25%, respectively. Thus, the antiproliferative activity of the EO from leaves was more pronounced than the EO from flowers in cells derived from melanoma.

Cathepsin D (CD), a lysosomal protease, and S100A4 protein, a calcium binding motif, are considered to be involved in metastasis in various human cancers. No data regarding such proteins are available for canine mammary carcinomas (CMCs). Accordingly, their expression in association with known factors of prognosis was investigated in this study. For that, 66 surgically resected CMCs were submitted to an immunohistochemical evaluation using anti CD, S100A4 protein, HER2, estrogen receptor α, cytokeratin 5, and p63 antibodies, further characterizing the tumors' molecular subtype. An increase in S100A4 immunoexpression by neoplastic luminal mammary cells was associated with an infiltrative tumor mode of growth, consequently leading us to conclude that S100A4 protein could be related to progression in CMCs. Additionally, the occurrence of the luminal A molecular subtype was associated with the complex histotype in CMCs. Although we have demonstrated that changes in S100A4 protein immunoexpression occurs in CMCs, further studies are needed to determine whether this represents important independent biomarkers for CMCs.

This work describes the chemical composition of the essential oils extracted from fresh leaves of Lippia origanoides Kunth. and Mentha spicata L. and their antioxidant and insecticidal activities. The essential oils were extracted by steam distillation using a modified Clevenger apparatus and subsequently analyzed by gas chromatography-flame ionization detector and gas chromatography-mass spectrometry. The antioxidant activities were determined by the β-carotene-linoleic acid method and by sequestration of the 2,2-diphenyl-1-picryl-hidrazila radical. The concentrations of the essential oils and the synthetic standard, butylated hydroxyltoluene, were 25, 50, 100, 150, 200, 250, 300 and 500 μg mL-1. Insecticidal activity was analyzed by non-preference with choice and no choice against the aphid Myzus persicae Sulzer. Gas chromatography analysis of the essential oil from Lippia origanoides Kunth. revealed carvacrol (41.51%), p-cymene (18.36%), γ-terpinene (17.03%) and thymol (4.86%) as major constituents, and the essential oil from Mentha spicata L. contained piperitona (81.18%), piperitenone (14.57%) and limonene (1.47%) as the principal components. The essential oils and the standard exhibited dose-dependent antioxidant activities at the concentrations tested. The essential oils were shown to be potential agents in the integrated management of the aphid Myzus persicae Sulzer.
