Impairment of pre-mRNA splicing in liver disease: Mechanisms and consequences

Pre-mRNA splicing is an essential step in the process of gene expression in eukaryotes and consists of the removal ofintrons and the linking of exons to generate mature mRNAs. This is a highly regulated mechanism that allows the alternative usage of exons, the retention ofintronic sequences and the generation of exonic sequences of variable length. Most human genes undergo splicing events, and disruptions of this process have been associated with a variety of diseases, including cancer. Hepatocellular carcinoma (HCC) is a molecularly heterogeneous type of tumor that usually develops in a cirrhotic liver. Alterations in pre-mRNA splicing of some genes have been observed in liver cancer, and although still scarce, the available data suggest that splicing defects may have a role in hepatocarcinogenesis. Here we briefly review the general mechanisms that regulatepre-mRNA splicing, and discuss some examples that illustrate how this process is impaired in liver tumorigenesis, and may contribute to HCC development. We believe that a more thorough examination of pre-mRNA splicing is still needed to accurately draw the molecular portrait of liver cancer. This will surely contribute to a better understanding of the disease and to the development of new effective therapies.

Efficiency in hydrocarbon degradation and biosurfactant production by Joostella sp.A8 when grown in pure culture and consortia

Joostella strains are emerging candidates for biosurfactant production. Here such ability was analyzed for Joostella strain A8 in comparison with Alcanivorax strain A53 and Pseudomonas strain A6, all previously isolated from hydrocarbon enrichment cultures made of polychaete homogenates. In pure cultures Joostella sp. A8 showed the highest stable emulsion percentage（78.33%）, hydrophobicity rate（62.67%）, and an optimal surface tension reduction during growth in mineral medium supplemented with diesel oil（reduction of about 12 mN/m）, thus proving to be highly competitive with Alcanivorax and Pseudomonas strains. During growth in pure culture different level of biodegradation were detected for Alcanivorax strain A53（52.7%）,Pseudomonas strain A6（38.2%） and Joostella strain A8（26.8%）. When growing in consortia,isolates achieved similar abundance values, with the best efficiency that was observed for the Joostella-Pseudomonas co-culture. Gas-chromatographic analysis revealed an increase in the biodegradation efficiency in co-cultures（about 90%）, suggesting that the contemporary action of different bacterial species could improve the process. Results were useful to compare the efficiencies of well-known biosurfactant producers（i.e. Pseudomonas and Alcanivorax representatives） with a still unknown biosurfactant producer, i.e. Joostella, and to confirm them as optimal biosurfactant-producing candidates.

We report carbon stock in biomass, litter and soil estimated for six locations in natural Quercus ilex L. stands of the Middle and High Moroccan Atlas. Twenty trees at each location were selected according to their diameter classes and felled to measure the biomass of trunk, branches, twigs and leaves and determine allometric relationships. Soil was sampled in five depths (0 - 15, 15 - 30, 30 - 50, 50 - 70 and 70 - 100 cm) and litterfall production measured in all tree stands. The total carbon stock in above-ground biomass ranged between 17 Mg·haǃ in Aït Aamar stand (High Atlas) and 91 Mg·haǃ in Ksiba stand (Middle Atlas). Perennial organs (trunk, branches and twigs) stored over 95% of the tree carbon stock. Soil organic carbon concentrations ranged from 0.01% (in 70 - 100 cm in all stands) to 8.1% (in 0 - 15 cm in the Ajdir stand in Middle Atlas). The total organic carbon stock in the soil ranged between 141.4 t·haǃ in Ajdir and 24.6 t·haǃ in Asloul. The litter contained 0.2 Mg C haǃ in the clearing (C2) stand of High Atlas and 14.3 Mg C haǃ in (Ajdir) of carbon. The best fitted model for predicting carbon stock in tree biomass was obtained by applying the allometric equation Y = aXb for each biomass fraction and stand, where Y is the aboveground biomass (dry weight) and X is the DBH (Mean diameter at breast height, 1.30 m). These previous data obtained in the present study confirm the important function of these natural forests as longterm C sinks, in forest biomass, litter and soil. The potential long term C storage of these systems is moderately high, especially in less-intensively managed forests that include large trees. The established relationship between DBH and carbon stock in different tree organs can be used for forest carbon accounting, and also synthesize available information on oak forest as a sink for atmospheric CO2, and identify the management options that may enhance the capacity for C capture/ storage

Most studies of predator-induced plasticity have focused on documenting how prey species re- spond to predators by modifying phenotypic traits and how traits correlate with fitness. We have previously shown that Pleurodema thaul tadpoles exposed to the dragonfly Rhionaeschna varie- gata responded strongly by showing morphological changes, less activity, and better survival than non-exposed tadpoles. Here, we tested whether there is a functional link between morphological plasticity and increased survival in the presence of predators. Tadpoles that experienced predation risk were smaller, less developed, and much less active than tadpoles without this experience. Burst speed did not correlate significantly with morphological changes and predator-induced deeper tails did not act as a lure to divert predator strikes away from the head. Although we have previously found that tadpoles with predator-induced morphology survive better under a direct predator threat, our results on the functional link between morphology and fitness are not conclu- sive. Our results suggest that in P. thaultadpoles （1） burst speed is not important to evade preda- tors, （2） those exposed to predators reduce their activity, and （3） morphological changes do not divert predator attacks away from areas that compromise tadpole survivalEE. Our results show that morphological changes in P. thaul tadpoles do not explain burst speed or lure attraction, al- though there was a clear reduction of activity, which itself reduces predation. We propose that changes in tadpole activity could be further analyzed from another perspective, with morphological change as an indirect product of behavior mediated by physiological mechanisms.

Cervical cancer is a priority health problem in Mexico. The objective of this work was to know the degree of satisfaction of the service provider with the organization of the program of Timely Detection of Cervical Cancer, specifically in taking of the cervical cytology. The study design was observational, transversal and prospective conducted from January to may 2015 in Jurisdiction 1 of the Health Services of San Luis Potosí, Mexico, in 20 health centers. Participants were 87 service providers whose function is to take cervical cytology (Pap smears). A 32-question questionnaire was applied, with Likert scale with 5 levels of response: 1 (dissatisfied), 2 (little satisfied), 3 (indifferent), 4 (satisfied) and 5 (very satisfied). The score range was 32 points (minimum job satisfaction) up to 160 points (maximum job satisfaction). We analyzed 2 dimensions: 1) intrinsic factors that have to do with motivation and professional recognition, and 2) extrinsic factors related to infrastructure, materials, remuneration and labor policies. Each factor consisted of four categories. Descriptive statistics and linear correlation of Pearson were applied for the analysis of data. Participants signed an informed consent letter. Of 4 categories, in the intrinsic dimension, the one with the highest percentage of satisfaction was “delegation of activities” with 28.8%. In the extrinsic dimension, the category with the highest percentage of satisfaction was “infrastructure and materials” with 49.4%;while satisfaction increase in intrinsic dimensions also does in extrinsic dimensions, such correlations are significant (p delegation of activities and remuneration (r = 0.074 and p = 0.497). The higher hierarchical level, the men, the bachelor academic level and older people showed the highest level of satisfaction.

The gastroesophageal reflux and/or peptic ulcer diseases are clinical conditions that occur usually accompanied of symptomatic pain. Lansoprazole, a proton pump inhibitor class drug is widely used in clinical practice for treatment of these diseases. However, its efficacy can be improved by combining with spasmolytic and/or visceral analgesic such as hyoscine butylbromide. Since hyoscine butylbromide is barely absorbed and exerts some local effects at gastrointestinal tract which may modify the absorption of lansoprazole, it is important to establish if there is a pharmacokinetic interaction after the oral concomitant administration of both drugs. For this objective, twenty-five subjects received under a crossover design an oral administration of lansoprazole (15 mg) plus placebo or a fixed-dose combination with hyoscine butiylbromide (15 mg + 10 mg, respectively). Plasma samples were obtained at different times during 10 hours. Lansoprazole plasma concentrations were determined by a high performance liquid chromatography method coupled to tandem mass spectrometry. Fixed-dose combination was well tolerated. Lansoprazole pharmacokinetic parameters were: Cmax 621.81 ± 212.79 and 450.38 ± 192.14 ng/mL;AUC0-t 1941.36 ± 845.57 and 1454.66 ± 757.28 ng·h/mL;tmax 2.83 ± 0.99 and 3.40 ± 1.82h;t1/2 1.35 ± 0.39 and 1.45 ± 0.51 h, for alone and combined fixed-dose formulation, respectively. Pharmacokinetic parameters were compared by analysis of variance and ratios of AUC0-t, Cmax and 90% confidence intervals obtained. Since confidence intervals exceed the 80% - 125% limits for these parameters, we conclude that there is a significantly pharmacokinetic interaction of lansoprazole when it is administered concomitantly with hyoscine butylbromide.

The airborne pollutants monitoring is an overriding task for humanity given that poor quality of air is a matter of public health, causing issues mainly in the respiratory and cardiovascular systems, specifically the PM10 particle. In this contribution is generated a base model with an Adaptive Neuro Fuzzy Inference System (ANFIS) which is later optimized, using a swarm intelligence technique, named Bacteria Foraging Optimization Algorithm (BFOA). Several experiments were carried with BFOA parameters, tuning them to achieve the best configuration of said parameters that produce an optimized model, demonstrating that way, how the optimization process is influenced by choice of the parameters.

Renin-Angiotensin System (RAS) is involved with hypertension and other cardiovascular diseases. However, the association of RAS components to cancer still causes suspicion. To try to clarify this, here we aimed to show this association for three important components: Angiotensin Converting Enzyme 1 (ACE1), Angiotensin Type 1 Receptor (AGTR1) and Angiotensin Type 2 Receptor (AGTR2). The first articles show that association of RAS components with cancer dates back to the 70’s. ECA1 and AGTR1 have close association with cancer and ACE1 inhibitors or AGTR1 blockers are candidates to treatment of some tumors. Moreover, the action of AGTR2 is still controversial, but most studies show that the increased expression of AGTR2 can attack the cancer cells. In breast cancer, these components have also been widely studied and many works have shown that the correlation exists. Therefore specific target using these RAS components could be a beneficial, novel therapy to various tumors.

The lamina cribrosa thickness（LCT） could be affected by dynamic changes in its *** spectral-domainoptical coherence tomography（SD-OCT）,we have studied the behaviour of the laminar region in 14 young subjects over 24 *** changes in LCT were observed,depending on the time at which the measurement was taken,with the maximum thickness being observed at 7.30 p.m.,and the minimum at 7.30 *** finding could suggests a circadian pattern in the LCT thickness in healthy subjects,which could have implications for the classification,diagnosis and prognosis of both normal and glaucomatous subjects.
