RNA viromes of Dermacentor nuttalli ticks reveal a novel uukuvirus in Qinghai Province,China

Ticks are a major parasite on the Qınghai-Tibet Plateau,western China,and represent an economic burden to agriculture and animal *** research on tick-borne pathogens that threaten humans and animals,the viromes of dominant tick species in this area remain *** this study,we collected Dermacentor nuttalli ticks near Qınghai Lake and identified 13 viruses belonging to at least six families through metagenomic *** viruses were of high abundance in pools,including Xınjiang tick-associated virus 1(XJTAV1),and three novel viruses:Qınghai Lake virus 1,Qınghai Lake virus 2(QHLV1,and QHLV2,unclassified),and Qınghai Lake virus 3(QHLV3,genus Uukuvirus of family Phenuiviridae in order Bunyavirales),which lacks the m *** minimum infection rates of the four viruses in the tick groups were 8.2%,49.5%,6.2%,and 24.7%,respectively,suggesting the prevalence of these viruses in *** ticks.A putative m segment of QHLV3 was identified from the next-generation sequencing data and further characterized for its signal peptide cleavage site,N-glycosylation,and transmembrane ***,we probed the L,m,and S segments of other viruses from sequencing data of other tick pools by using the putative m segment sequence of *** revealing the viromes of *** ticks,this study enhances our understanding of tick-borne viral communities in highland *** putative m segment identified in a novel uukuvirus suggests that previously identified uukuviruses without m segments should have had the same genome organization as typical *** findings will facilitate virus discovery and our understanding of the phylogeny of tick-borne uukuviruses.

Surface ozone in global cities:A synthesis of basic features,exposure risk,and leading meteorological driving factors

Long-term exposure to high surface ozone(O_(3))concentrations,a complex oxidative atmospheric pollutant,can adversely impact human *** on O_(3)monitoring data from 261 cities worldwide in 2020,generalized additive model(GAm)and spatial data analysis(SDA)methods were applied in this study to quantitatively evaluate the spatiotemporal distribution of O_(3)concentration,exposure risk,and dominant meteorological *** indicated that over 40%of the cities worldwide were exposed to harmful O_(3)concentration ranges(40-60μg/m^(3)),with most cities distributed in China and ***,significant seasonal variations in global O_(3)concentrations were observed,presenting as summer(45.6μg/m^(3))>spring(47.3μg/m^(3))>autumn(38.0μg/m^(3))>winter(33.6μg/m^(3)).Exposure analysis revealed that approximately 12.2%of the population in 261 cities were exposed to an environment with high O_(3)concentrations(80-160μg/m^(3)),with about 36.32 million people in major ***,the persistent increase in high O_(3)levels worldwide is a critical factor contributing to threats to human ***,GAm results indicated temperature,relative humidity,and wind speed as primary determinants of O_(3)*** synergy of meteorological factors is critical for understanding O_(3)*** findings are important for enforcing robust air quality policies and mitigating public risk.

Type I interferon(IFN)inhibits a wide spectrum of viruses through stimulating the expression of antiviral *** an IFN-induced protein,myxovirus resistance B(mXB)protein was reported to inhibit multiple highly pathogenic human *** remains to be determined whether mXB employs a common mechanism to restrict different ***,we find that IFN alters the subcellular localization of hundreds of host proteins,and this IFN effect is partially lost upon mXB *** results of our mechanistic study reveal that mXB recognizes vimentin(VIm)and recruits protein kinase B(AKT)to phosphorylate VIm at amino acid S38,which leads to reorganization of the VIm network and impairment of intracellular trafficking of virus protein complexes,hence causing a restriction of virus *** results highlight a new function of mXB in modulating VIm-mediated trafficking,which may lead towards a novel broad-spectrum antiviral strategy to control a large group of viruses that depend on VIm for successful replication.

Characterization and spatial analysis of coseismic landslides triggered by the Luding ms 6.8 earthquake in the Xianshuihe fault zone, Southwest China

On September 5, 2022, a magnitude ms 6.8 earthquake occurred along the moxi fault in the southern part of the Xianshuihe fault zone located in the southeastern margin of the Tibetan Plateau,resulting in severe damage and substantial economic loss. In this study, we established a coseismic landslide database triggered by Luding ms 6.8 earthquake, which includes 4794 landslides with a total area of 46.79 km^(2). The coseismic landslides primarily consisted of medium and small-sized landslides, characterized by shallow surface sliding. Some exhibited characteristics of high-position initiation resulted in the obstruction or partial obstruction of rivers, leading to the formation of dammed lakes. Our research found that the coseismic landslides were predominantly observed on slopes ranging from 30° to 50°, occurring at between 1000 m and 2500 m, with slope aspects varying from 90° to 180°. Landslides were also highly developed in granitic bodies that had experienced structural fracturing and strong-tomoderate weathering. Coseismic landslides concentrated within a 6 km range on both sides of the Xianshuihe and Daduhe fault zones. The area and number of coseismic landslides exhibited a negative correlation with the distance to fault lines, road networks, and river systems, as they were influenced by fault activity, road excavation, and river erosion. The coseismic landslides were mainly distributed in the southeastern region of the epicenter, exhibiting relatively concentrated patterns within the IX-degree zones such as moxi Town, Wandong River basin, Detuo Town to Wanggangping Township. Our research findings provide important data on the coseismic landslides triggered by the Luding ms 6.8 earthquake and reveal the spatial distribution patterns of these landslides. These findings can serve as important references for risk mitigation, reconstruction planning, and regional earthquake disaster research in the earthquake-affected area.

Wheat germplasm is a fundamental resource for basic research,applied studies,and wheat breeding,which can be enriched normally by several paths,such as collecting natural lines,accumulating breeding lines,and introducing mutagenesis *** methane sulfonate(EmS)is an alkylating agent that can effectively introduce genetic variations in a wide variety of plant *** this study,we created a million-scale EmS population(mEP)that started with the Chinese wheat cultivars‘Luyan 128’,‘Jimai 38’,‘Jimai 44’,and‘Shannong 30’.In the m1 generation,the mEP had numerous phenotypical variations,such as>3,000 chlorophyll-deficient mutants,2,519 compact spikes,and 1,692 male sterile *** were also rare mutations,including 30 independent tillers each with double *** m1 variations of chlorophyll-deficiency and compact spikes were inheritable,appearing in the m2 or m3 *** advance the entire mEP to higher generations,we adopted a single-seed descendent(SSD)*** other seed composites of m2 were used to screen other agronomically important traits,such as the tolerance to herbicide *** mEP is available for collaborative projects,and provides a valuable toolbox for wheat genetics and breeding for sustainable agriculture.

This study presents a rapid and versatile low-cost sample-to-answer system for SARS-CoV-2 *** system integrates the extraction and purification of nucleic acids,followed by amplification via either reverse transcriptionquantitative polymerase chain reaction(RT–qPCR)or reverse transcription loop-mediated isothermal amplification(RT–LAmP).By meeting diverse diagnostic and reagent needs,the platform yields testing results that closely align with those of commercial RT-LAmP and RT‒qPCR *** advantages of our system include its speed and *** assay is completed within 28 min,including sample loading(5 min),ribonucleic acid(RNA)extraction(3 min),and RT-LAmP(20 min).The cost of each assay is≈$9.5,and this pricing is competitive against that of Food and Drug administration(FDA)-approved commercial *** some RNA loss during on-chip extraction is observed,the platform maintains a potential limit of detection lower than 297 *** makes the system particularly useful in environments where centralized laboratories are either unavailable or inconveniently *** key feature is the platform’s versatility,allowing users to choose between RT‒qPCR or RT‒LAmP tests based on specific requirements.

Rethinking the architecture design of data center networks

In the rising tide of the Internet of things, more and more things in the world are connected to the Internet. Recently, data have kept growing at a rate more than four times of that expected in moore＇s law. This explosion of data comes from various sources such as mobile phones, video cameras and sensor networks, which often present multidi- mensional characteristics. The huge amount of data brings many challenges on the management, transportation, and pro- cessing IT infrastructures. To address these challenges, the state-of-art large scale data center networks have begun to provide cloud services that are increasingly prevalent. How- ever, how to build a good data center remains an open chal- lenge. Concurrently, the architecture design, which signifi- cantly affects the total performance, is of great research inter- est. This paper surveys advances in data center network de- sign. In this paper we first introduce the upcoming trends in the data center industry. Then we review some popular design principles for today＇s data center network architectures. In the third part, we present some up-to-date data center frame- works and make a comprehensive comparison of them. Dur- ing the comparison, we observe that there is no so-called op- timal data center and the design should be different referring to the data placement, replication, processing, and query pro- cessing. After that, several existing challenges and limitations are discussed. According to these observations, we point out some possible future research directions.
