Dear Editor,Recent studies from us and others have deciphered the clinical relevance of a variety of oncohistones1 in different *** H3K27m and H3K36m mutant histones exert dominant-negative effects on methylation levels of histone H3K27 and H3K36 on wildtype histone protein in pediatric brain cancers2–4 and chondroblastoma,5,6 *** contrast to H3K27m and H3K36m that act in trans to cause global reduction of methylation at the respective residues,the H3G34 mutations,including H3G34V/R and H3G34W/L found in pediatric high-grade glioma and giant cell tumor of the bone,affect the H3K36 methylation in cis and alter the H3K36 methylation on the nucleosome containing the H3G34 mutation(s).

Rationale Haematoma growth is common early after intracerebral haemorrhage(ICH),and is a key determinant of *** acid,a widely available antifibrinolytic agent with an excellent safety profile,may reduce haematoma *** and design Stopping intracerebral haemorrhage with tranexamic acid for hyperacute onset presentation including mobile stroke units(STOP-mSU)is a phase Ⅱ double-blind,randomised,placebo-controlled,multicentre,international investigator-led clinical trial,conducted within the estimand statistical *** In patients with spontaneous ICH,treatment with tranexamic acid within 2 hours of onset will reduce haematoma expansion compared with *** size estimates A sample size of 180 patients(90 in each arm)would be required to detect an absolute difference in the primary outcome of 20%(placebo 39%vs treatment 19%)under a two-tailed significance level of *** adaptive sample size re-estimation based on the outcomes of 144 patients will allow a possible increase to a prespecified maximum of 326 *** Participants will receive 1 g intravenous tranexamic acid over 10 min,followed by 1 g intravenous tranexamic acid over 8 hours;or matching *** efficacy measure The primary efficacy measure is the proportion of patients with haematoma growth by 24±6 hours,defined as either≥33%relative increase or≥6 mL absolute increase in haematoma volume between baseline and follow-up CT *** We describe the rationale and protocol of STOP-mSU,a phase Ⅱ trial of tranexamic acid in patients with ICH within 2 hours from onset,based in participating mobile stroke units and emergency departments.

Dear Editor,Fascin actin-bundling protein 1(Fascin,gene name:FSCN1)is the primary actin-bundling protein that clusters filaments into stable and bundled filopodia[1].In previous studies,elevated levels of Fascin promoted filopodia formation and enhanced the invasive ability of tumor cells,suggesting that Fascin is a target for therapeutic intervention[2,3].Therefore,it is necessary to elucidate the detailed molecular mechanism by which Fascin bundles F-actin filaments.

Our previous studies found that Zinc-finger protein 382(ZNF382)played as a tumor suppressor gene in esophageal and gastric cancers,and a positive correlation between the high expression of ZNF382 and better outcome in breast cancer ***,the biological roles and mechanisms of ZNF382 in breast cancer remains *** detected ZNF382 expression by reverse-transcription PCR(RT-PCR)and real-time quantitative PCR(qRT-PCR)in breast cancer cells and tissues,and explored the impacts and mechanisms of ectopic ZNF382 expression in breast cancer cells in vitro and in vivo,*** results revealed that ZNF382 was significantly down-regulated in breast cancer tissues compared with adjacent non-cancer *** of ZNF382 expression in silenced breast cancer cells not only inhibited tumor cell colony formation,viability,migration and invasion,and epithelial-mesenchymal-transition(EmT),but also induced apoptosis and G0/G1 *** conclusion,ZNF382 could induce G0/G1 cell cycle arrest through inhibiting CDC25A signaling,and,inhibit cell migration,invasion and EmT by antagonizing ZEB1 signaling in breast cancer cells.

APPL2 Negatively Regulates Olfactory Functions by Switching Fate Commitments of Neural Stem Cells in Adult Olfactory Bulb via Interaction with Notch1 Signaling

Adult olfactory neurogenesis plays critical roles in maintaining olfactory ***-generated neurons in the subventricular zone migrate to the olfactory bulb(OB) and determine olfactory discrimination,but the mechanisms underlying the regulation of olfactory neurogenesis remain *** previous study indicated the potential of APPL2(adaptor protein,phosphotyrosine interacting with PH domain and leucine zipper 2) as a modulating factor for neurogenesis in the adult olfactory *** the present study,we report how APPL2 affects neurogenesis in the OB and thereby mediates olfactory discrimination by using both in vitro neural stem cells(NSCs) and an in vivo animal model-APPL2 transgenic(Tg) *** the in vitro study,we found that APPL2 overexpression resulted in NSCs switching from neuronal differentiation to gliogenesis while APPL2 knockdown promoted *** the in vivo study,APPL2 Tg mice had a higher population of glial cells and dampened neuronal production in the olfactory system,including the corpus callosum,OB,and rostral migratory *** APPL2 Tg mice displayed impaired performance in olfactory discrimination tests compared with wild-type ***,we found that an interaction of APPL2 with Notch1 contributed to the roles of APPL2 in modulating the neurogenic lineage-switching and olfactory *** conclusion,APPL2 controls olfactory discrimination by switching the fate choice of NSCs via interaction with Notch1 signaling.

Objective:To study the inhibitory effects on colorectal cancer(CRC)and the underlying mechanism of the petroleum ether extract of Aegiceras corniculatum leaves(PACL).materials and methods:The effect of PACL on the proliferation of CRC cell lines DLD-1,HT-29,and SW480was measured by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium assay and colony-forming *** then,a wound-healing assay was used to measure the migration ability of three CRC *** cell cycle and apoptosis of three CRC cells were measured by PI/RNase staining and annexin V-FITC/double staining,respectively,and the intrinsic apoptosis pathway was studied by the Western *** anti-CRC effect of PACL in vivo was evaluated by HT-29 xenograft zebrafish ***:PACL inhibited cell viability and proliferation in DLD-1,HT-29,and SW480 cells in a dose-and time-dependent *** can inhibit cell migration in DLD-1and SW480 cells but not in the less mobile phenotype cell *** treatment resulted in cell cycle arrest of DLD-1 and HT-29 cells in the G2/m ***,PACL can induce apoptosis in all three CRC cells,which may be achieved by regulating the intrinsic apoptosis pathway mediated by mitochondria and the endoplasmic ***,the tumor sizes were decreased after treatment with PACL and PACL combined with fluorouracil in HT-29 xenograft zebrafish ***:These findings suggested that PACL may exert its anti-CRC effect by inducing apoptosis through the intrinsic apoptosis pathway and show a significant anti-CRC effect in vitro and in vivo,so it might be potentially developed as an anti-CRC agent.

针对英特尔光峰技术提出了一种非球面微透镜模块的光学优化设计及评价方法。该非球面微透镜模块可经由GGP多模光纤应用于高速数据的传输,非球面微透镜的直径为800μm,数值孔径为0.275。借助光学设计软件Code V及LightTools进行仿真,评估了微透镜模块制造及组装的公差对系统耦合效率的影响,同时考虑了一些影响光功率损耗因素。结果表明,经光学优化设计得到的非球面微透镜模块的耦合损耗为-0.75dB。最后,对于球面微透镜模块和非球面微透镜的性能进行了评价。

目的:比较不同种类计算机辅助设计/辅助制作(computer aided design/manufacturing,CAD/CAm)修复材料抛光后表面粗糙度值(Ra值)与光泽度值的差异,测定适宜的抛光时间,为操作者抛光椅旁可切削修复体提供参考。方法:选择5种不同种类CAD/CAm修复材料长石瓷(vita markⅡ,Vm)、弹性瓷(vita enamic,VE)、优韧瓷(lava ulimate,LU)、复合树脂A(shofu block HC,SB)和复合树脂B(brilliant crios,BC),每种材料制备6个试样,共30个试样。将试样固定于自制抛光装置,使用Sof-Lex抛光碟系统中的中碟(medium disk,m碟,磨粒粒径10~40μm)、细碟(fine disk,F碟,磨粒粒径3~9μm)和超细碟(superfine disk,SF碟,磨粒粒径1~7μm)依次对试样进行序列抛光。试样每抛光10 s测量一次Ra值及光泽度值,数值不再变化时更换下一级抛光碟,每个抛光碟仅使用一次。更换下一级抛光碟的同时记录试样的Ra值、光泽度值以及抛光时间,实验完成后用SPSS 24.0软件进行统计学分析。结果:序列抛光后所有材料Ra值较抛光前显著降低(P0.05),而LU的光泽度值[(68.1±4.5)GU]与SB的光泽度值[(68.2±5.8)GU]显著高于VE[(48.1±8.1)GU]与BC[(53.2±5.8)GU],Pm[40(30,55)s]所需总抛光时间最短,VE[140(135,145)s]、LU[130(120,140)s]、SB[140(130,150)s]与BC[130(120,140)s]的抛光时间差异无统计学意义。结论:所有CAD/CAm修复材料经Sof-Lex抛光碟系统序列抛光后均能显著降低表面粗糙度值和提高光泽度值;不同材料达到最佳表面粗糙度和光泽度所需的抛光时间不同;推荐使用Sof-Lex系统抛光时,对于长石瓷,仅用m碟抛光40 s即可。对于弹性瓷、优韧瓷、复合树脂A与复合树脂B,要序列使用m碟、F碟和SF碟抛光,总体抛光时间约130~140 s。
