Idiopathic pulmonary fibrosis (IPF) is a dreadful, chronic, and irreversibly progressive disease leading to death with a few effective treatments. Our previous study suggested that repetitive hyperbaric oxygen (HBO) treatment alleviates bleomycin-induced pulmonary fibrosis in mice. Here, we investigated the protective mechanism of HBO treatment against pulmonary fibrosis using an integrated approach. Analyzing publicly available expression data from the mouse model of bleomycin-induced pulmonary fibrosis as well as IPF patients, several potential mechanisms of relevance to IPF pathology were identified, including increased epithelial-to-mesenchymal transition (EMT) and glycolysis. High EMT or glycolysis scores in bronchoalveolar lavage were strong independent predictors of mortality in multivariate analysis. These processes were potentially driven by hypoxia and blocked by HBO treatment. Together, these data support HBO treatment as a viable strategy against pulmonary fibrosis.

Water surface wave turbulence is one of the factors afecting the performances of underwater optical wireless communication(UOWC)systems.In our research,a phase-conjugate beam was used to correct the beam distortion and enhance the communication performances when a system is subject to surface wave turbulence.The phase-conjugate beam was generated by a phase-conjugate mirror(PCM),and a turbulence generator was used to generate surface wave turbulence in the experiment.We calculated the beam centroid distribution and the results showed that the phase-conjugate beam had a better propagation performance than the distorted beam at the diferent water depths.The root mean square(RMS)of the beam centroid for the phase-conjugate beam was 11 times less than that for the distorted beam,which meant that the phase-conjugate beam could efectively correct the beam drift.We further investigated the scintillation index and the signal-to-noise ratio(SNR);the results showed that the phase-conjugate beam was able to reduce the scintillation and an obvious improvement in SNR could be obtained.This research has the potential to be applied in UWC.

It shows that detrimental exposures and conditions in mothers can lead to the development of obesity and type 2 diabetes in offspring.This can lead to a vicious cycle of metabolic dysfunction,where rising rates of obesity,pre-diabetes,and diabetes in individuals of reproductive age,propagating risks to subsequent generations.It is well established that regular exercise has important health benefits for people with obesity and type 2 diabetes.Recently,increasing studies aim to examine the effects of maternal exercise on metabolic health in offspring.This review aims to demonstrate the evidence linking maternal exercise during critical periods of development and its implications for glucose metabolism in offspring,including intervention timing,sexual dimorphism,different exercise type,and intensity.Then we further examine the potential role of epigenetic modifications in this process.

The interaction between cancer cells and M2 tumor-associated macrophages(M2-TAMs)facilitates tumor growth and metastasis.However,cancer cells and M2-TAMs have different spatial distribution patterns,which requires distinct drug delivery strategies.Herein,based on different tumor-penetrating ability of nanocarriers,we developed a combinatory strategy that consists of a TAMs-targeting liposome(alanine-alanine-asparagine(AAN)-lip-regorafenib(Rego))and a cancer cells-targeting copolymer(internalizing RGD modified with N-(2-hydroxypropyl)methacrylamide-doxorubicin(iRGD-HD)).Our study confirmed AAN-lip-Rego accumulated at perivascular sites where M2-TAM is located,while iRGD-HD penetrated into deep site of tumor to enter cancer cells.Thereafter,we found iRGD-HD induced cancer cells undergoing immunogenic cell death to enhance tumor infiltration of CD8^(+)T cells.Meanwhile,AAN-lip-Rego efficiently repolarized TAMs from M2 into M1 to alleviate tumor immunosuppression,thus reviving CD8^(+)T cells.Moreover,the repolarization of TAMs led to dramatic downregulation of prometastatic factors expressed on cancer cells.As a result,such combinatory approach elicited robust antitumor immune responses and generated considerable anti-tumor and anti-metastasis efficacy to markedly inhibit primary tumor and spontaneous lung metastasis.

Obesity has become a global health problem. lifestyle modification and medical treatment only appear to yield short-term weight loss. Roux-en-Y gastric bypass (RYGB) is the most popular bariatric procedure, and it sustains weight reduction and results in the remission of obesity-associated comorbidities for obese individuals. However, patients who undergo this surgery may develop hypoglycemia. To date, the diagnosis is challenging and the prevalence of post-RYGB hypoglycemia (PRH) is unclear. RYGB alters the anatomy of the upper gastrointestinal tract and has a combined effect of caloric intake restriction and nutrient malabsorption. Nevertheless, the physiologic changes after RYGB are complex. Although hyperinsulinemia, incretin effects, dysfunction of β-cells and α-cells, and some other factors have been widely investigated and are reported to be possible mediators of PRH, the pathogenesis is still not completely understood. In light of the important role of the gut microbiome in metabolism, we hypothesized that the gut microbiome might also be a critical link between RYGB and hypoglycemia. In this review, we mainly highlight the current possible factors predisposing individuals to PRH, particularly related to the gut microbiota, which may yield significant insights into the intestinal regulation of glucose metabolic homeostasis and provide novel clues to improve the treatment of type 2 diabetes mellitus.

Tumor recurrence and metastasis is the leading cause of mortality for postoperative breast cancer patients. However, chemotherapy intervention after surgery is often unsatisfactory, because residual microtumors are difficult to target and require frequent administration. Here, an all-in-one and oncefor-all drug depot based on in situ-formed hydrogel was applied to fit the irregular surgical trauma, and enable direct contact with residual tumors and sustained drug release. Our immunological analysis after resection of orthotopic breast tumor revealed that postsurgical activation of CXCR4-CXCL12 signal exacerbated the immunosuppression and correlated with adaptive upregulation of PD-L1 in recurrent tumors. Thus, a multifunctional hydrogel toolkit was developed integrating strategies of CXCR4 inhibition,immunogenicity activation and PD-L1 blockade. Our results showed that the hydrogel toolkit not only exerted local effect on inhibiting residual tumor cell “seeds” but also resulted in abscopal effect on disturbing pre-metastatic “soil”. Furthermore, vaccine-like effect and durable antitumor memory were generated, which resisted a secondary tumor rechallenge in 100% cured mice. Strikingly, one single dose of such modality was able to eradicate recurrent tumors, completely prevent pulmonary metastasis and minimize off-target toxicity, thus providing an effective option for postoperative intervention.

A novel method for road network mining from floating car data

Vehicles have been increasingly equipped with GPS receivers to record their trajectories,which we call floating car data.Compared with other data sources,these data are characterized by low cost,wide coverage,and rapid updating.The data have become an important source for road network extraction.In this paper,we propose a novel approach for mining road networks from floating car data.First,a Gaussian model is used to transform the data into bitmap,and the Otsu algorithm is utilized to detect road intersections.Then,a clothoid-based method is used to resample the GPS points to improve the clustering accuracy,and the data are clustered based on a distance-direction algorithm.Last,road centerlines are extracted with a weighted least squares algorithm.We report on experiments that were conducted on floating car data from Wuhan,China.To conclude,existing methods are compared with our method to prove that the proposed method is practical and effective.

Type 2 diabetes mellitus and metabolic disorders have become an epidemic globally.However,the pathogenesis remains largely unclear and the prevention and treatment are still limited.In addition to environmental factors during adulthood,early life is the critical developmental window with high tissue plasticity,which might be modified by external environmental cues.Substantial evidence has demonstrated the vital role of early-life nutrition in programming the metabolic disorders in later life.In this review,we aim to overview the concepts of fetal programming and investigate the effects of early-life nutrition on energy metabolism in later life and the potential epigenetic mechanism.The related studies published on PubMed database up to March 2020 were included.The results showed that both maternal overnutrition and undernutrition increased the riskes of metabolic disorders in offspring and epigenetic modifications,including DNA methylation,miRNAs,and histone modification,might be the vital mediators.The beneficial effects of early-life lifestyle modifications as well as dietary and nutritional interventions on these deleterious metabolic remolding were initially observed.Overall,characterizing the early-life malnutrition that reshapes metabolic disease trajectories may yield novel targets for early prevention and intervention and provide a new point of view to the energy metabolism.

Objective:Pulmonary infectious diseases(PID)include viral pneumonia(VP)and pulmonary tuberculosis(PT).Mongolian medicine(MM)is an effective treatment option in China,however,the core group medicines(CGMs)in the treatment of PID and their underlying therapeutic mechanisms remain unclear.In this study,through the method of data mining,the CGMs of MM for the treatment of PID were excavated,and the possible mechanism of action of the CGMs in the treatment of PID was explored by using network pharmacology.Methods:First,89 MM formulae for the treatment of pulmonary infectious diseases collected from Gan Lu Si Bu,Meng Yi Jin Kui,People’s Republic of China Ministry of Health Drug Standards(Mongolian Medicine Volume),Standard of Mongolian Medicine Preparations in Inner Mongolia(2007 Edition),and Standard of Mongolian Medicine Preparations in Inner Mongolia(2014 Edition).The CGMs of MM for PID were excavated through association rule analysis and cluster analysis.Then,the active ingredients and potential targets of the CGMs were obtained from TCMSP,TCMIP,BATMAN-TCM databases.PID targets information was collected from OMIM,GeneCards,and DrugBank databases.The possible targets of CGMs treatment for PID were obtained by intersection.The PPI network was constructed through the STRING database,and the topology analysis of the network was performed.Through the enrichment analysis of the intersection targets by R language,the main action pathways and related target proteins of CGMs in the treatment of PID were screened out.The results were verified by molecular docking.Results:A total of 89 formulae were included,involving 164 MM herbs.The efficacy of the drugs was mainly cough-suppressing and panting-calming herbs,and heat-clearing herbs.The nature and favor were mainly bitter and cold.The CGMs of MM to treatment of PID was excavated as the classic famous formula Sanzi Decoction(Toosendan Fructus-Chebulae Fructus-Gardeniae Fructus).A total of 28 candidate components and 237 predicted targets of CGMs were collected,and 61 common t

Removal efficacy and mechanism of nitrogen and phosphorus by biological aluminum-based P-inactivation agent (BA-PIA)

In this study,aluminum-based P-inactivation agent(Al-PIA)was used as a high-efficiency microbial carrier,and the biological Al-PIA(BA-PIA)was prepared by artificial aeration.Laboratory static experiments were conducted to study the effect of BA-PIA on reducing nitrogen and phosphorus contents in water.Physicochemical characterization and isotope tracing method were applied to analyze the removal mechanism of nitrogen and phosphorus.High-throughput techniques were used to analyze the characteristic bacterial genus in the BA-PIA system.The nitrogen and phosphorus removal experiment was conducted for 30 days,and the removal rates of NH_(4)^(+)-N,TN and TP by BA-PIA were 81.87%,66.08%and 87.97%,respectively.The nitrogen removal pathways of BA-PIA were as follows:the nitrification reaction accounted for 59.0%(of which denitrification reaction accounted for 56.4%),microbial assimilation accounted for 18.1%,and the unreacted part accounted for 22.9%.The characteristic bacteria in the BA-PIA system were Streptomyces,Nocardioides,Saccharopolyspora,Nitrosomonas,and Marinobacter.The loading of microorganisms only changed the surface physical properties of Al-PIA(such as specific surface area,pore volume and pore size),without changing its surface chemical properties.The removal mechanism of nitrogen by BA-PIA is the conversion of NH_(4)^(+)-N into NO_(2)^(−)-N and NO_(3)^(−)-N by nitrifying bacteria,which are then reduced to nitrogen-containing gas by aerobic denitrifying bacteria.The phosphorus removal mechanism is that metal compounds(such as Al)on the surface of BA-PIA fix phosphorus through chemisorption processes,such as ligand exchange.Therefore,BA-PIA overcomes the deficiency of Al-PIA with only phosphorus removal ability,and has better application prospects.
