Tea polyphenols increase X-ray repair cross-complementing protein 1 and apurinic/apyrimidinic endonuclease/redox factor-1 expression in the hippocampus of rats during cerebral ischemia/reperfusion injury

Recent studies have shown that tea polyphenols can cross the blood-brain barrier, inhibit apoptosis and play a neuroprotective role against cerebral ischemia. Furthermore, tea polyphenols can decrease DNA damage caused by free radicals. We hypothesized that tea polyphenols repair DNA damage and inhibit neuronal apoptosis during global cerebral ischemia/reperfusion. To test this hypothesis, we employed a rat model of global cerebral ischemia/reperfusion. We demonstrated that intraperitoneal injection of tea polyphenols immediately after reperfusion significantly reduced apoptosis in the hippocampal CA1 region; this effect started 6 hours following reperfusion. Immunohistochemical staining showed that tea polyphenols could reverse the ischemia/reperfusion-induced reduction in the expression of DNA repair proteins, X-ray repair cross-complementing protein 1 and apudnic/apyrimidinic endonuclease/redox factor-1 starting at 2 hours. Both effects lasted at least 72 hours. These experimental findings suggest that tea polyphenols promote DNA damage repair and protect against apoptosis in the brain.

This study aimed to investigate the dose-effect of iron on growth performance,antioxidant *** morphology,and mRNA expression of jejunal tight junction protein in 1-to21-d-old yellow-feathered broilers.A total of 7201-d-old yellow-feathered maleb roilers were allocated to 9 treatments with 8 replicate cages of 10 birds per *** dietary treatments were consisted of a basal diet(contained 79.6 mg Fe kg^(-1))supplemented with 0,20,40,60,80,160,320,640,and 1,280 mg Fe kg^(-1)in the form of FeSO_(4)·7H_(2)*** with the birds in the control group,birds supplemented with 20mg Fe kg^(-1)had higher average daily gain(ADG)(Pxidative stress,disrupted intestinal morphology,and reduced mRNA expression of jejunal tight junction protein.

Epidural scarring occurs inevitably in the defect after spinal laminectomy, and thus how to prevent or reduce it becomes a challenging topic. In the present study, an injectable hydrogel and its dexamethasone （DEX）-loaded hydrogel systems were adopted to prevent epidural scarring in a postlaminectomy rat model. The hydrogel system composed of poly（D,L-lactic acid-co-glycolic acid）-poly（ethylene glycol）-poly（D,L-lactic acid-co-glycolic acid）（PLGA-PEG-PLGA） triblock copolymers was a free-flowing sol at room temperature, and spontaneously turned into a semi-solid gel at body temperature. A lumbar 3 total laminectomy was performed on Sprague Dawley （SD） rats, and the efficacy of the injectable hydrogel with or without drugs in preventing epidural scar formation was evaluated via the gross anatomical observation and histological examination at one month post-surgery. The results demonstrate that the use of hydrogel alone reduced epidural scarring significantly, whereas the efficacy of the DEX-loaded hydrogels presented an irregular dose-dependency of drug and even the inappropriate drug doses resulted in the negative results. Therefore, the present study confirms that the PLGA-PEG- PLGA hydrogel holds potential as a barrier device to decrease peridural scarring, and reveals that the sustained delivery of the steroid hormone DEX to prevent surgery-related adhesions in the laminectomy defect is complex. Moreover, our in vivo studies also remind the researchers to pay attention to the irregular dose-dependency of the hormone drugs.

目的:探讨加速康复外科(enhanced recovery after surgery,ERAS)在冠心病体外循环下冠状动脉搭桥术中的应用价值。方法:采用回顾性队列研究方法。收集泰达国际心血管病医院外六科2020年1月至2021年12月行单纯冠状动脉搭桥术的患者资料。以2020年1月至2020年12月的238例为对照组,以2021年1月至2021年12月的226例为ERAS组。对照组采用传统的冠状动脉搭桥术的方法。ERAS组在传统方法的基础上改进和增加了ERAS措施。两组患者接受相同的手术组手术,均采用体外循环下行单纯冠状动脉搭桥术,术后采用相同的气管插管拔除标准、胸腔引流管拔除标准、转出重症监护病房(intensive care unit,ICU)标准及出院标准。主要观察指标为术后住院时间。次要观察指标为相关时间、术后相关并发症、二次入院发生率和总治疗费用。相关时间包括术前停药时间、带气管插管时间、下床时间、带引流管时间和ICU停留时间和总住院时间。术后并发症包括术后二次开胸、二次插管、肺感染、气管切开、需要外科处理的胸腔积液和心包积液、胸部切口愈合困难或感染以及二次入院。随访情况,采用门诊和电话方式进行随访。随访时间截至2022年3月。采用多元线性回归分析冠状动脉搭桥术患者术后住院治疗时间的影响因素。结果:两组患者一般情况和搭桥支数对比,差异均无统计学意义(均P>0.05)。两组在术前停药时间、带引流管时间和ICU停留时间上差异均无统计学意义(均P>0.05)。ERAS组在带气管插管时间、下床时间、术后住院时间和总住院时间均短于对照组,差异均有统计学意义(均P0.05)。总治疗费用ERAS组低于对照组,差异有统计学意义(P<0.001)。患者年龄、性别、体重指数(body mass index,BMI)、术前左室舒张末容积(left ventricular end-diastolic volume,LVEDV)和ERAS的应用与术后住院时间相关,男性和ERAS的应用是术后住院时间的保护因素,年龄、BMI和术后LVEDV则会导致住院时间延长(均P<0.001)。结论:ERAS理念应用在冠心病单纯冠状动脉搭桥术患者围手术期是安全可行的,可明显加快患者术后的恢复,缩短住院时间,减少住院治疗费用。
