Cell Survival Assays for Individualised Chemotherapy in Primary Glioma Cultures—Colourmetric or Luminescent?

In vitro chemosensitivity testing of short term primary glioma cultures derived from brain biopsies is still in the research phase and has not yet found a place in clinical use. The main reasons for this slow progression are the small amounts of tissue available and the lack of a suitably sensitive assay capable of use in the clinical setting. This study examines whether the MTS and ATP cell survival assays, which determine cytotoxicity via colorimetric and luminescence analysis respectively, could potentially fulfill this role. Primary glioma cultures were tested for chemosensitivity using the MTS and ATP assays and were found to be generally sensitive to cisplatin and paclitaxel but relatively resistant to carmustine and etoposide. For both assays, LD50 values lay in the range 2 - 130 μg/ml but in the vast majority of cases, those obtained by the ATP assay were markedly lower those obtained by the MTS assay. Moreover, at cell numbers less than 2000 in the cases of paclitaxel and carmustine and less than 4500 in the case of cisplatin, these drugs were generally indicated as ineffective against the glioma cultures tested by the MTS assay but effective against these cultures by the ATP assay. These data clearly demonstrate that the ATP assay is more sensitive when estimating small cell numbers generated by primary glioma cultures from brain biopsies and more reliably detects higher kill rates by anticancer drugs. This study also supports the feasibility of using the ATP assay for chemosensitivity testing in a clinical setting.

The aim of this study was to evaluate the yield of clinical symptoms, signs, and radiological studies in the diagnosis of foreign body aspiration (FBA) in children. Methods: During the 2- year study, we performed on all children admitted to the pediatric department for suspected FBAa protocol that included the following: thorough medical history, physical examination, radiological studies (chest x-ray and fluoroscopy), and rigid bronchoscopy by a senior otolaryngologist. The yield of these measures for the diagnosis of FBA was evaluated. Results: Foreign bodies were found in 56 (57% ) of the 98 children with suspected FBA. Thirty-three (59% ) children were boys and 23 (41% ) girls (P h a mean age of 24 months (range, 8- 84 months). Main symptoms in the children with FBA were the following: choking (76.8% ), prolonged cough (14.3% ), dyspnea (3.6% ), and nonresolving pneumonia (1.8% )- . Physical examination, chest x-ray, and fluoroscopy findings were abnormal in 80.4% , 67.9% , and 46.9% of the children with FBA, respectively. The diagnostic yield of physical examination and radiological studies increased 24 hours after the event of FBA. In 45.2% of the children with positive history but with normal findings from physical examination and radiological studies, foreign bodies were found. Of the children with “ doubtful” history, physical examination, chest x-ray, and fluoroscopy findings were abnormal in 58% , 38% , and 12.5% , respectively. Foreign bodies were found in 9.5% of these children. Conclusion: Medical history is the key for the diagnosis of FBA. Choking followed by an acute episode of coughing is the most common presentation of FBA. The yield of physical examination and radiological studies in the diagnosis of FBA is relatively low but is increased when the presentation is delayed and when history is doubtful. If FBA is suspected, bronchos- copy should be performed.

Transesterification is the most common production process for biodiesel. From this reaction, a glycerin phase is produced that is impure, thus lowering market value. however, because it is rich in carbon, it is an alternative for generating bioproducts with a higher added value through bioconversion by microorganisms. The aim of this study was to screen parameters, such as ph （4, 5, 6, 7 and 8） and the initial glycerol concentration at 30 ± ℃ with agitation at 150 rpm for bioemulsifier and lipid synthesis in a submerged medium by Yarrowia lipolytica IMUFRJ 50678 from crude glycerin. The best conditions for bioemulsifier production were 30 ± ℃ at ph： 6 with 50 g/L of initial substrate, which produced 2.7 g/L of lipids, from which the optimum 300.5 mg/L of triglycerides was produced over 48 h of microorganism growth.

Gastric colonization by helicobacter pylori increases the risk of gastric disorders, including atrophic gastritis which can be diagnosed based on levels of serum biomarkers like Gastrin and Pepsinogen. We therefore examined the efficacy of a serological-based method namely GastroPanel Blood kit, in diagnosing and scoring gastritis associated to helicobacter pylori infection. Patients with dyspeptic symptoms were prospectively recruited on voluntary basis at the Yaounde Central hospital and University Teaching hospital, from March to July 2011. The degree of atrophy was classified according to levels in patient serum of pepsinogens I and II (PGI and PGII) and Gastrin 17 (G17) and compared with histological profiles as reference method. A specific ELISA test was used for the detection of h. pylori IgG antibodies. In total, 86 volunteers from 21 to 83 years old (mean = 46.4 ± 3.3) were enrolled, including 74.4% of women and 25.6% of men. The prevalence of gastritis was statistically similar between Gastro Blood Panel test and histology used as reference method (89.5% versus 83.7%: p > 0.20). Diagnosis based on serum makers showed high sensitivity (93.1%) in comparison with the reference method. however, the serological based method has diagnosed more atrophic gastritis than the reference (17.4% versus 7.0%: p 0.05). Furthermore, the prevalence of h. pylori infection did not differ significantly between serological method (84.9%) and reference method (81.4%). These results suggest that diagnosis of atrophic gastritis and h. pylori infection obtained with an optional serological method (GastroPanel) is in a strong agreement with the biopsy findings, and thus can be a useful non endoscopic assessment of stomach mucosal atrophy in patients with dyspepsia.

A cellular automaton （CA） model is established to simulate dynamic recrystallization （DRX） in the β single-phase field of Ti6Al4 V alloy, and the kinetics during DRX processing has been analyzed. The model employed considers the influences of dynamic recovery, nucleation rate, strain rate and dislocation density on DRX, and practical deformation parameters, such as temperature, strain and strain rate on DRX have been considered in the simulation. The simulated DRX grain size and DRX grain shape agree well with the experimental results, which shows the availability and lea- sibility of the cellular automaton method for the simulation of DRX. The result of kinetics analysis of DRX reveals that the Avrami exponent is variable ranging from 2.4 to 2.9, which increases with the increase of strain rate.

Background: Gastric ulcer is one of the most common gastrointestinal diseases with a worldwide prevalence of about 40% in the developed countries and 80% in Africa. Acid inhibitors, anticholinergics, histamine h2-antagonists and antibiotics are commonly used to treat gastric ulcer. however, the accumulating evidence for resistance to antibiotics and the side effects of antibiotics and acid inhibitors, anticholinergics and histamine h2-antagonists. Therefore, there is an urgent need for approaches to treat and prevent gastric ulcer. One alternative strategy is the use of probiotic lactic acid bacteria. Aim of the study: This study aimed to isolate and characterised probiotic lactic acid bacteria from palm wine and fermented milk, and to evaluate their antiulcerogenic potentials on ethanol-induced gastric ulcer in mice. Methods: Probiotic lactic acid bacteria were isolated from the fermented milk and palm wine using pour plate technique on MRS agar and identified using the 16S r RNA gene sequencing. For functional properties and selection, acid and bile salt tolerance were evaluated based on viable colony count on MRS agar. Two probiotic lactic acid bacteria were selected for in vivo studies. Fifty-four healthy young adult Balb/c mice were randomly divided into 9 groups of 6 mice each. Gastric ulcer was induced in mice using one oral dose of absolute ethanol (10 mL/kg body weight). The probiotic lactic acid bacteria (F1 and F2) at different doses (MF3 = 9 × 108 CFU/mL, MF6 = 1.8 × 109 CFU/mL and MF9 = 2.7 × 109 CFU/mL) and omeprazole (20 mg/kg) (a reference drug) were orally administrated daily for 14 days before ulcer induction. These mice were sacrificed 1hour after induction and the stomach contents were collected for volume and ph determination. AIM： To determine age norms in the first three years of life for grating visual acuity and contrast sensitivity obtained with lea grating test and hiding heidi low contrast face test. METhODS： lea grating test was used to estimate binocular grating acuity and hiding heidi low contrast face test was used to estimate contrast sensitivity in 600 healthy infants and children. Age ranged from 3 to 36 mo subdivided into 12 groups subjected for full ophthalmologic and pediatric ***： The grating acuity developed along the first three years of life. It ranged from 1.88±0.32 c/d at 3mo to 30.95±0.77 c/d at 36 mo. The most rapid development was during the first 12 mo and the slowest development was from 30 to 36 mo. The contrast sensitivity showed rapid development in the first two years of life. Its mean value ranged from 4.23±1.17 at 3mo to 78.26±8.21 at 24 mo. It was constant at the highest score（80） ***： Age norms for grating acuity along with contrast sensitivity offer a more comprehensive measure of spatial vision and should be incorporated in clinical practice for better visual assessment in preverbal and nonverbal children.

BACKGROUNDDiabetes mellitus causes a large majority of non-traumatic major and minoramputations globally. Patients with diabetes are clinically complex with amultifactorial association between diabetic foot ulcers (DFU) and subsequentlower extremity amputations (lea). Few studies show the long-term outcomeswithin the cohort of DFU-associated *** highlight the long-term outcomes of lea as a result of ***/MEDLINE and Google Scholar were searched for key terms, “diabetes”,“foot ulcers”, “amputations” and “outcomes”. Outcomes such as mortality, reamputation,re-ulceration and functional impact were recorded. Peer-reviewedstudies with adult patients who had DFU, subsequent amputation and follow upof at least 1 year were included. Non-English language articles or studiesinvolving children were *** total of 22 publications with a total of 2334 patients were selected against theinclusion criteria for review. The weighted mean of re-amputation was 20.14%,29.63% and 45.72% at 1, 3 and 5 years respectively. The weighted mean of mortality at 1, 3 and 5 years were 13.62%, 30.25% and 50.55% respectively withsignificantly higher rates associated with major amputation, re-amputation andischemic *** lea, level of the lea and patient comorbidities were significant riskfactors contributing to re-ulceration, re-amputation, mortality and depreciatedfunctional status.
