A Dominant Allele of Arabidopsis Pectin-Binding Wall-Associated Kinase Induces a Stress Response Suppressed by MPK6 but Not MPK3 Mutations

The plant cell wall is composed of a matrix of cellulose fibers, flexible pectin polymers, and an array of as- sorted carbohydrates and proteins. The receptor-like Wall-Associated Kinases （WAKs） of Arabidopsis bind pectin in the wall, and are necessary both for cell expansion during development and for a response to pathogens and wounding. Mitogen Activated Protein Kinases （MPKs） form a major signaling link between cell surface receptors and both transcrip- tional and enzyme regulation in eukaryotes, and Arabidopsis MPK6 and MPK3 indeed have important roles in develop- ment and the response to stress and pathogens. A dominant allele of WAK2 requires kinase activity and activates a stress response that includes an increased ROS accumulation and the up-regulation of numerous genes involved in pathogen resistance, wounding, and cell wall biogenesis. This dominant allele requires a functional pectin binding and kinase domain, indicating that it is engaged in a WAK signaling pathway. A null mutant of the major plasma membrane ROS-producing enzyme complex, rbohd/f does not suppress the WAK2cTAP-induced phenotype. A mpk6, but not a mpk3, null allele is able to suppress the effects of this dominant WAK2 mutation, thus distinguishing MPK3 and MPK6, whose activity previously was thought to be redundant. Pectin activation of gene expression is abated in a wak2-null, but is tempered by the WAK-dominant allele that induces elevated basal stress-related transcript levels. The results suggest a mechanism in which changes to the cell wall can lead to a large change in cellular responses and help to explain how pathogens and wounding can have general effects on growth.

Approaches to antiretroviral therapy in China

China has recognized the threat of HIV to its population and responded with a national antiretroviral treatment (ART) program. However, high ART failure rates and the spread of resistance within populations are important realities to consider when developing and managing ART programs in China and worldwide. Concepts which will define treatment success and local and national programmatic goals are 1) access to ART, 2) durability of ART at the patient level, 3) scalability of treatment modalities, and the 4) sustainability of the program at the community or national level. In the face of limited resources, China must also consider when to start ARV therapy, which agents to use, when to switch them, and how to treat highly experienced patients with drug resistance. The optimal ARV regimen to start with is changing frequently with the introduction of new agents and the presentation of new data. Currently, a regimen including tenofovir, emtricitabine or lamivudine and a nonnucleoside reverse transcriptase inhibitor appears to have optimal characteristics to treat HIV/AIDS in China. However, critical to all of these choices is the evaluation of programs implemented to insure wide scale success. China has wisely begun this process of evaluating the perfor- mance of local programs through systematic monitoring and evaluation of treatment outcomes. This will allow regi- mens and programs that work to be expanded, and programs with high failure rates to be eliminated. In the end, evidence based data supporting treatment strategies will allow China to successfully confront its AIDS epidemic early and prevent its tragic consequences

PROBING THE IMPACT OF GAMMA-IRRADIATION ON THE METABOlIC STATE OF NEURAl STEM AND PRECURSOR CEllS USING DUAl-WAVElENGTH INTRINSIC SIGNAl TWO-PHOTON EXCITED FlUORESCENCE

Two-photon excitedfluorescence(TPEF)spectroscopy and imaging were used to investigate the effects of gamma-irradiation on neural stem and precursor cells(NSPCs).While the observed signal from reduced nicotinamide adenine dinucleotide(NADH)was localized to the mitochondria,the signal typically associated with oxidizedflavoproteins(Fp)was distributed diffusely throughout the *** measured TPEF emission and excitation spectra were similar to the established spectra of NAD(P)H and *** intensity was markedly increased by addition of the electron transport chain(ETC)modulator menadione to the medium,along with a concomitant decrease in the NAD(P)H ***-dimensional(3D)neurospheres were imaged to obtain the cellular metabolic index(CMI),calculated as the ratio of Fp to NAD(P)Hfluorescence *** effects were found to differ between low-dose(50 cGy)and high-dose(50 cGy)***-dose irradiation caused a marked drop in CMI values accompanied by increased cellular *** higher doses,both NAD(P)H and Fp signals increased,leading to an overall elevation in CMI *** underscore the complex relationship between radiation dose,metabolic state,and proliferation status in NSPCs and highlight the ability of TPEF spectroscopy and imaging to characterize metabolism in 3D spheroids.

Activation of mitochondrial ATP-sensitive potassium channels delays ischemia-induced cellular uncoupling in rat heart

AIM: To test the hypothesis that cellular uncoupling induced by myocardial ischemia is mediated by activation of cmitochondrial ATP-sensitive potassium channels (mitoKATP). METHODS: Rat hearts were perfused on a langendorff apparatus and subjected to 40-min ischemia followed by 30-min reperfusion (I/R). Changes in cellular coupling were monitored by measuring whole-tissue resistance. RESUlTS: (1) In hearts subjected to I/R, the onset of uncoupling started at (13.3±1.0) mm of ischemia; (2) Ischemic preconditioning (IPC) delayed the onset of uncoupling until (22.7±1.3) min. Assessment of Risk of Establishment of Wheat Dwarf Bunt (Tilletia controversa) in China

Wheat dwarf bunt, caused by Tilletia controversa Kühn （TCK）, is an important quarantine wheat disease throughout the world. Based on published research results of the biology and the epidemiology of the disease, the main factors including temperature, humidity, snow cover, and their parameters relating to teliospore germination, infection and epidemics of TCK were determined. The geophytopathological models for the risk analysis of wheat dwarf bunt establishment were modified. Fifty-year meteorologic data from about 500 weather stations in China were used to calculate the probabilities of TCK establishment in different geographic sites with the models. The map that displays the establishment risk of TCK in winter wheat growing regions in China was generated by using geographical information system （GIS）. The zones showing high, moderate, low, and very low, including no risk, of TCK establishment accounted for 27.33, 27.69, 38.12, and 6.86% of total winter wheat growing areas in China, respectively. These results will provide useful information to formulate quarantine regulations and wheat importation policy in China.

Effect of diets on the feeding behavior and physiological properties of suspension-feeding sea cucumber Cucumaria frondosa

The suspension-feeding sea cucumber Cucumaria frondosa has become commercially important in recent *** proper diets is the first important step for intensive aquaculture of this sea *** this study,adult *** were exposed to one of the following diet treatments:control(no diet provided),two powdered seaweeds(Ascophyllum nodosum and Saccharina latissima),a commercially available microalgal diet(shellfish diet)and natural *** effects of diets on the feeding behavior and physiological properties of sea cucumbers were investigated after a 5-week rearing *** show that sea cucumbers fed with shellfish diet exhibited a significantly higher tentacle insertion rate(1.80±0.20 insertion/min)than these fed with seaweed powders,and there was no significant different between the two groups fed by seaweed *** significant difference was found on the fecal production rate among the feeding *** minimum oxygen consumption rate was observed in the control group(5.76±0.99μg O2/(g·h)),which is significantly lower than individuals fed with ***,shellfish diet,and natural seston;however,no significant difference was shown between those of control and *** *** maximum ammonium excretion rate was found in the *** group(0.03±0.01μmol/(g·h)),which is significantly higher than other *** minimum O/N ratio was observed in the *** group(14.57±1.04),which is significantly lower than the ***,shellfish diet,and natural seston *** fed with seaweed powders had similar physiological properties with these fed with microalgae diet and natural seston,indicating that *** and *** can be explored as promising diets for intensive aquaculture of ***.

Bovine immunoglobulin protein isolates for the nutritional management of enteropathy

The gastrointestinal tract is responsible for a multitude of digestive and immune functions which depend upon the balanced interaction of the intestinal microbiota, diet, gut barrier function, and mucosal immune response. Disruptions in one or more of these factors can lead to intestinal disorders or enteropathies which are characterized by intestinal inflammation, increased gut permeability, and reduced capacity to absorb nutrients. Enteropathy is frequently associated with human immunodeficiency virus(HIV) infection, inflammatory bowel disease, autoimmune enteropathy, radiation enteritis, and irritable bowel syndrome(IBS), where pathologic changes in the intestinal tract lead to abdominal discomfort, bloating, abnormal bowel function(e.g., diarrhea, urgency, constipation and malabsorption). Unfortunately, effective therapies for the management ofenteropathy and restoring intestinal health are still not available. An accumulating body of preclinical studies has demonstrated that oral administration of plasmaor serum-derived protein concentrates containing high levels of immunoglobulins can improve weight, normalize gut barrier function, and reduce the severity of enteropathy in animal models. Recent studies in humans, using serum-derived bovine immunoglobulin/protein isolate, demonstrate that such protein preparations are safe and improve symptoms, nutritional status, and various biomarkers associated with enteropathy. Benefits have been shown in patients with HIV infection or diarrhea-predominant IBS. This review summarizes preclinical and clinical studies with plasma/serum protein concentrates and describes the effects on host nutrition, intestinal function, and markers of intestinal inflammation. It supports the concept that immunoglobulin-containing protein preparations may offer a new strategy for restoring functional homeostasis in the intestinal tract of patients with enteropathy.

Nicotine-induced upregulation of antioxidant protein Prx 1 in oral squamous cell carcinoma

Nicotine is a source of exogenous oxidative stress, which is associated with the pathogenesis of numerous diseases including oral squamous cell carcinoma (OSCC), whereas an antioxidant protein, peroxiredoxin 1 (Prx 1), plays an important role in the modulation of this condition. This study was to investigate the association between Prx 1 and tobacco-induced oxidative stress. The expression of Prx 1 and GST in OSCC Tca8113 cells, which were pre-treated with nicotine, was determined. In the present study, MTT assay, reactive oxygen species (ROS) assay, RT-PCR and Western blot analyses, respectively, were conducted to assess cell viability, ROS level, and expression level of Prx 1 and GST in nicotine-treated Tca8113 cells. Nuclear factor kappa B (NF- B) expression was detected by immuno-fluorescence. Our results showed the growth of Tca8113 cells was increased in a dose-dependent manner when cells were treated with nicotine at concentrations from 0.1 to 10 mol/l, but the proliferation of the cells decreased at 100 mol/l. ROS levels increased in all groups treated with nicotine at concentrations of 0.1, 1, 10, or 100 mol/l for 24h. Prx 1 and GST mRNA and protein expression were up-regulated in cells treated with nicotine for the same time at different concentrations or at the same concentration for different times (Plocated from cytoplasm to nucleus, the expression of NF- B was increased in nucleus. These results suggest that up-regulation of Prx1 expression appears to be associated with tobacco-induced oxidative stress, which may play an important role in the pathogenesis of OSCC.

Journalism Teacher Assessment of Education Inhibitors

Combining,, cultural-historical theory＇ of human development with research pinpointing education inhibitors, a survey was developed to investigate how journalism teachers evaluated the effects of five education inhibitors on their students＇ learning. The list of inhibitors included electronic gadgets, lack of nutrition, parental education levels, family support levels, and racial subcultures. Results showed that use of electronic gadgets and lack of parental edxlcation were perceived as having the most negative influence on learning, while students＂ subcultures were perceived as having the smallest effect. Respondents also knew the least about their students＇ nutritional intake. Further, school size and student socio-economic status seem to play some role in teacher awareness of education inhibitors, and this is an important finding. It shows that while increasing teacher awareness of various education inhibitors, demographic factors also must be considered when targeting specific inhibitors.

Assessment of VIIRS 375 m active fire using tropical peatland combustion algorithm applied to landsat-8 over Indonesia’s peatlands

VIIRS 375 m active fire data(VNP14IMG),the highest spatial resolution available cost-free fire product,were assessed for representing fire in typical degraded tropical peatlands in *** results of applying the Tropical Peatland Combustion Algorithm to landsat-8(ToPeCAl-l8)daytime imagery were utilised as the fire *** permit the comparison of non-simultaneous VNP14IMG and ToPeCAl-l8,peatland fire propagation speeds resulting from previous study using TET-1 data in Central Kalimantan’s peatlands were *** peatland fires were still within 750 m from their source over 15 h under uniform conditions,except for very large *** detection rates of nighttime VNP14IMG compared with ToPeCAl-l8 showed about 80%agreement for small fire areas(larger than 14 ha,a dissolved 375 m buffer(cluster buffer)of VNP14IMG active fires with an integration of nighttime and daytime acquisitions,produced a probability of detection up to 90%.These results generated a recommendation for implementing cluster buffer analysis and integration of nighttime and daytime analysis of VNP14IMG data for better accuracy in fire detection for ground fire *** also demonstrate the utility of the ToPeCAl-l8 algorithm with VIIRS 375 m active fire data.
