Peanut allergy is majorly related to severe food induced allergic *** food including cow's milk,hen's eggs,soy,wheat,peanuts,tree nuts(walnuts,hazelnuts,almonds,cashews,pecans and pistachios),fish and shellfish are responsible for more than 90%of food ***,we provide promising insights using a large-scale data-driven analysis,comparing the mechanistic feature and biological relevance of different ingredients presents in peanuts,tree nuts(walnuts,almonds,cashews,pecans and pistachios)and ***,we have analysed the chemical compositions of peanuts in different processed form raw,boiled and *** the data-driven approach we are able to generate new hypotheses to explain why nuclear receptors like the peroxisome proliferator-activated receptors(PPARs)and its isoform and their interaction with dietary lipids may have significant effect on allergic *** results obtained from this study will direct future experimeantal and clinical studies to understand the role of dietary lipids and PPARisoforms to exert pro-inflammatory or anti-inflammatory functions on cells of the innate immunity and influence antigen presentation to the cells of the adaptive immunity.

Near infrared light-induced dynamic modulation of enzymatic activity through polyphenol-functionalized liquid metal nanodroplets

Dynamic manipulation of enzymatic activity is a challenging task for applications in chemical and pharmaceutical industries due to the difficult modification and variable conformation of various ***, we report a new strategy for reversible dynamic modulation of enzymatic activity by near-infrared light-induced photothermal conversion based on polyphenol-functionalized liquid metal nanodroplets(LM). The metal-phenolic nanocoating not only provides colloidal stability of LM nanodroplets but also generates nanointerfaces for the assembly of various enzymes on the LM nanodroplets. Upon near infrared(NIR) irradiation, the localized microenvironmental heating through photothermal effect of the LM nanodroplets allows tailoring the enzymatic activity without affecting the bulk temperature. A library of functional enzymes, including proteinase K, glucoamylase, glucose oxidase, and Bst DNA polymerase, is integrated to perform a reversible control and enhanced activities even after five times of cycles, demonstrating great potential in bacterial fermentation, bacteriostasis, and target gene amplification.

3D extrusion of multi-biomaterial lattices using an environmentally informed workflow

The conventional building material palette has been proven limited in terms of adaptability to our current environmental challenges. Innovations in computational design and digital manufacturing have supported the broadening of biomaterial applications as an alternative. While biomaterials are characteristically responsive to stimuli such as temperature and humidity, their unpredictable behaviour is a hurdle to standardization and architectural utilisation. To examine the nexus between material formulation, computation and manufacturing, multi-biomaterial lattice structures were produced through an environmentally informed workflow. Customized biomaterial development resulted in three candidate biopolymer blends with varying levels of hydro-responsiveness and transparency. The computational strategy included a machine learning clustering algorithm to customise results and dictate material distribution outputs. To test the workflow, environmental data of solar radiation exposure and solar heat gain from a specific location was used to inform the material deposition via pneumatic extrusion for the design and digital fabrication of a deformation-controlled prototype of 350 mm × 350 mm. This led to a series of multi-biomaterial wall panel components that can be applied at architectural scale. In future, these techniques can support the incorporation of living elements to be embedded within the built environment for truly animate architecture.

Heterogeneous populations of neural stem cells contribute to myelin repair

As ingenious as nature＇s invention of myelin sheaths within the mammalian nervous system is, as fatal can be damage to this specialized lipid structure. Long-term loss of electrical insulation and of further supportive functions myelin provides to axons, as seen in demyelinating diseases such as multiple sclerosis （MS）, leads to neurodegeneration and results in progressive disabilities. Multiple lines of evidence have demon-strated the increasing inability of oligodendrocyte precursor cells （OPCs） to replace lost oligodendrocytes （OLs） in order to restore lost myelin. Much research has been dedicated to reveal potential reasons for this regeneration deficit but despite promising approaches no remyelination-promoting drugs have successfully been developed yet. In addition to OPCs neural stem cells of the adult central nervous system also hold a high potential to generate myelinating OLs. There are at least two neural stem cell niches in the brain, the subventricular zone lining the lateral ventricles and the subgranular zone of the dentate gyrus, and an additional source of neural stem cells has been located in the central canal of the spinal cord. While a substantial body of literature has described their neurogenic capacity, still little is known about the oligodendrogenic potential of these cells, even if some animal studies have provided proof of their contribution to remyelination. In this review, we summarize and discuss these studies, taking into account the different niches, the heterogeneity within and between stem cell niches and present current strategies of how to promote stem cell-mediated myelin repair.

Novel approaches for the development of peripheral nerve regenerative therapies

Schwann cells are the myelinating glial cells of the peripheral nervous system（PNS）.By establishing lipid-rich myelin sheaths around large-caliber axons,they ensure that electrical signal transmission is accelerated-a process referred to as saltatory signal *** from this prominent physiological function,these cells also exert important pathophysiological roles in PNS injuries or dis- eases. In contrast to the central nervous system （CNS）, the adult PNS retains a remarkably high degree of intrinsic re- generation. As a consequence, transected axons and dam- aged myelin sheaths can be repaired and nerve functional- ity can be restored. This spontaneous regenerative capacity depends on （inter） actions of macrophages, neurons, and Schwann cells.

Laser powder bed fusion(LPBF)for the fabrication of dense components used for tooling applications,is highly *** stresses,which evolve in the additively manufactured part,are inherent to LPBF *** additional stress contribution in high-carbon steels arises from the austenite-to-martensite phase transformation,which may eventually lead to cracking or even *** an alternative to pre-heating the base plate,which is not striven by industry,lowering the martensite content which forms in the part,is essential for the fabrication of dense parts by LPBF of high-carbon tool steels which are then adapted to *** this study,a successful strategy demonstrates the processing of the Fe85Cr4Mo1V1W8C1(wt%)high-carbon steel by LPBF into dense parts(99.8%).The hierarchical microstructure consists of austenitic and martensitic grains separated by elemental segregations in which nanoscopic carbide particles form a network.A high density of microsegregation was observed at the molten pool boundary ultimately forming a *** LPBF-fabricated steel shows a yield strength,ultimate compressive stress,and total strain of 1210 MPa,3556 MPa,and 27.4%,*** mechanical and wear performance is rated against the industrially employed and highly wear-resistant 1.2379 tool steel taken as the *** its lower macro-hardness,the LPBF steel(58.6 HRC,0.0061 mm^(3) Nm^(-1))shows a higher wear resistance than the reference steel(62.6 HRC,0.0078 mm^(3) Nm^(-1)).This behavior results from the wear-induced formation of martensite in a microscale thick layer directly at the worn surface,as it was proven via high-energy X-ray diffraction mapping.

BACKGROUND Biliary microlithiasis/sludge is detected in approximately 30%of patients with idiopathic acute pancreatitis(IAP).As recurrent biliary pancreatitis can be prevented,the underlying aetiology of IAP should be *** To develop a machine learning(ML)based decision tool for the use of endosonography(EUS)in pancreatitis patients to detect sludge and *** We retrospectively used routinely recorded clinical and laboratory parameters of 218 consecutive patients with confirmed AP admitted to our tertiary care hospital between 2015 and *** who did not receive EUS as part of the diagnostic work-up and whose pancreatitis episode could be adequately explained by other causes than biliary sludge and microlithiasis were *** trained supervised ML classifiers using H_(2)*** automatically selecting the best suitable predictor model to predict microlithiasis/*** predictor model was further validated in two independent retrospective cohorts from two tertiary care centers(117 patients).RESULTS Twenty-eight categorized patients’variables recorded at admission were identified to compute the predictor model with an accuracy of 0.84[95%confidence interval(CI):0.791-0.9185],positive predictive value of 0.84,and negative predictive value of 0.80 in the identification cohort(218 patients).In the validation cohort,the robustness of the prediction model was confirmed with an accuracy of 0.76(95%CI:0.673-0.8347),positive predictive value of 0.76,and negative predictive value of 0.78(117 patients).CONCLUSION We present a robust and validated ML-based predictor model consisting of routinely recorded parameters at admission that can predict biliary sludge and microlithiasis as the cause of AP.
