社会建构论作为一种知识的元理论,同经验主义一样,是有关知识的一切主张之理论,同时也是一种实践着的理论。作为一种新的研究范式,社会建构论打开了通往多种理解、多元价值和多样化生活的大门。传统的实证主义研究方法和实践可以被社会建构论范式整合。许多新的研究方式,如叙事研究、行动研究、话语研究等,得到社会建构论强有力的方法论支持。社会建构论赋予人一种批判意识,要求澄清某种特定的社会建构的实践意义,并关注对美好未来的创造。对于社会建构论是"反实在论"或"相对主义"的批评大多出于某些误解。

本文依据怀尔实验室研究出的预示方法,初步估计了航天飞机的脉动压力环境。特别着重叙述平行燃烧型航天飞机发射过程中的跨音速阶段。由轻量级轨道飞机和单独的固体火箭助推器组成的基线外形,和一条典型的飞行轨道一起,被当作预示估算的模型。在跨音速马赫数区域,对各临界脉动压力环境作了预示,其最大脉动压力级如下:1)肩部引起的气流分离(157分贝),2)结尾激波振荡(164分贝),3)突超物引起的气流分离(168分贝),4)压缩角引起的气流分离(148～157分贝),5)激波引起的干扰流(161分贝),6)无激波干扰流(150分贝),7)附体紊流边界层流(137分贝)。总的说,预示的压力环境与风洞试验结果的比较表明一致性很好。预示的上述环境的1/3倍频带谱,一般为如下三种类型之一:1)附体紊流边界层谱(典型高频),2)均匀分离流和无激波干扰振荡谱(典型中频)和3)激波振荡和激波干扰流谱(典型低频)。对羽状排气引起的分离流环境做了预示。仅有固体火箭助推器羽状排气的影响是重要的,其脉动压力级与压缩角引起的分离流和激波振荡的压力级相当。

Dear Editor,Asian rice(Oryza sativa)is the staple food for half the world and is a model crop that has been extensively *** contributes20%of calories to the human diet(Stein et al.,2018).With the increase in global population and rapid changes in climate,rice breeders need to develop new and sustainable cultivars with higher yields,healthier grains,and reduced environmental footprints(Wing et al.,2018).Since the first gold-standard reference genome of rice variety Nipponbare was published(International Rice Genome Sequencing Project,2005),an increasing number of rice accessions have been sequenced,assembled,and annotated with global ***,a single reference genome is obviously insufficient to perform the genetic difference analysis for rice ***,the pan-genome has been proposed as a solution,which allows the discovery of more presence-absence variants compared with single-reference genome-based studies(Zhao et al.,2018).Over the past years,several databases,such as RAP-db(https://***),RGAP(http://***),and Gramene(https://***),have long-term served rice genomic research by providing information based on one or a series of individual reference *** integrate and utilize the genomic information of multiple accessions,we performed comparative analyses and established the user-friendly Rice Gene Index(RGI;https://***)*** is the first gene-based pan-genome database for rice.

Western corn rootworm （WCR, Diabrotica virgifera virgifera LeConte） is highly sensitive to orally delivered double-stranded RNA （dsRNA）. RNAi in WCR is systemic and spreads throughout the insect body. This raises the question whether transitive RNAi is a mechanism that functions in WCR to amplify the RNAi response via production of secondary siRNA. Secondary siRNA production is achieved through RNA-dependent RNA polymerase （RdRP） activity in other eukaryotic organisms, but RdRP has not been identified in WCR and any other insects. This study visualized the spread of the RNAi- mediated knockdown of Dr v-ATPase C mRNA throughout the WCR gut and other tissues using high-sensitivity branched DNA in situ hybridization. Furthermore, we did not detect either secondary siRNA production or transitive RNAi in WCR through siRNA sequence profile analysis. Nucleotide mismatched sequences introduced into either the sense or antisense strand of v-ATPase C dsRNAs were maintained in siRNAs derived from WCR fed with the mismatched dsRNAs in a strand specific manner. The distribution of all siRNAs was restricted to within the original target sequence regions, which may indicate the lack of new dsRNA synthesis leading to production of secondary siRNA. Thus, the systemic spread of RNAi in WCR may be derived from the original dsRNA molecules taken up from the gut lumen. These results indicate that the initial dsRNA dose is important for a lethal systemic RNAi response in WCR and have implications in developing effective dsRNA traits to control WCR and in resistance management to prolong the durability of RNAi trait technology.

Complement receptor 2 （CR2/CD21） is predominantly expressed on the surface of mature B cells where it forms part of a coreceptor complex that functions, in part, to modulate B-cell receptor signal strength. CR2/CD21 expression is tightly regulated throughout B-cell development such that CR2/CD21 cannot be detected on pre-B or terminally differentiated plasma cells. CR2/CD21 expression is upregulated at B-cell maturation and can be induced by IL-4 and CD40 signaling pathways. We have previously characterized elements in the proximal promoter and first intron of CR2/CD21 that are involved in regulating basal and tissue-specific expression. We now extend these analyses to the CR2/CD21 core promoter. We show that in mature B cells, CR2/~D21 transcription proceeds from a focused TSS regulated by a non-consensus TATA box, an initiator element and a downstream promoter element. Furthermore, occupancy of the general transcriptional machinery in pre-B versus mature B-cell lines correlate with CR2/CD21 expression level and indicate that promoter accessibility must switch from inactive to active during the transitional B-cell window.
