Optimization of micropatterned poly(lactic-coglycolic acid) films for enhancing dorsal root ganglion cell orientation and extension

Nerve conduits have been a viable alternative to the ‘gold standard’ autograft for treating small peripheral nerve gap injuries. However, they often produce inadequate functional recovery outcomes and are ineffective in large gap injuries. Ridge/groove surface micropatterning has been shown to promote neural cell orientation and guide growth. However, optimization of the ratio of ridge/groove parameters to promote orientation and extension for dorsal root ganglion （DRG） cells on poly（lactic-co-glycolic acid）（PLGA） films has not been previously conducted. Photolithography and micro-molding were used to define various combinations of ridge/groove dimensions on PLGA films. The DRG cells obtained from chicken embryos were cultured on micropatterned PLGA films for cell orientation and migration *** of the films occurred during the test period, however, this did not cause deformation or distortion of the micropatterns. Results from the DRG cell orientation test suggest that when the ridge/groove ratio equals 1 （ridge/groove width parameters are equal, i.e., 10 μm/10 μm （even））, the degree of alignment depends on the size of the ridges and grooves, when the ratio is smaller than 1 （groove controlled） the alignment increases as the ridge size decreases, and when the ratio is larger than 1 （ridge controlled）, the alignment is reduced as the width of the grooves decreases. The migration rate and neurite extension of DRG neurons were greatest on 10 μm/10 μm and 30 μm/30 μm micropatterned PLGA films. Based on the data, the 10 μm/10 μm and 30 μm/30 μm micropatterned PLGA films are the optimized ridge/groove surface patterns for the construction of nerve repair devices.

Controlled release of FK506 from micropatterned PLGA films:potential for application in peripheral nerve repair

After decades of research,peripheral nerve injury and repair still frequently results in paralysis,chronic pain and neuropathies leading to severe disability in *** clinically available nerve conduits only provide crude guidance of regenerating axons across nerve gap without additional ***506（Tacrolimus）,an FDA approved immunosuppressant,has been shown to enhance peripheral nerve regeneration but carries harsh side-effects when delivered *** objective of this study was to develop and evaluate a bioresorbable drug delivery system capable of local extended delivery of FK506 that also provides topological guidance cues to guide axon growth via *** was used to create micropatterned poly（lactide-co-glycolic acid）（PLGA） films embedded with ***-patterned,10/10 μm（ridge/groove width）,and 30/30 μm patterned films loaded with 0,1,and 3 μg/cm2 FK506 were manufactured and *** vitro FK506 rate of release testing indicated that the films are capable of an extended（at least 56 days）,controlled,and scalable release of *** extension bioactivity assay indicated that FK506 released from the films（concentration of samples tested ranged between 8.46–19.7 ng/m L） maintained its neural bioactivity and promoted neurite extension similar to control FK506 dosages（10 ng/m L FK506）.The multi-functional FK506 embedded,micropatterned poly（lactide-co-glycolic acid） films developed in this study have potential to be used in the construction of peripheral nerve repair devices.

The scaffold pore size influences many critical physical aspects of tissue engineering,including tissue infiltration,biodegradation rate,and mechanical *** measurements of pore sizes from scanning electron micrographs using ImageJ/FIJI are commonly used to characterize scaffolds,but these methods are both time-consuming and subject to user *** semi-automated analysis tools are limited by a lack of accessibility or limited sample size in their verification *** work here describes the development of a new mATLAB algorithm,PoreScript,to address these *** algorithm was verified using three common scaffold fabrication methods(e.g.,salt leaching,gas foaming,emulsion templating)with varying pore sizes and shapes to demonstrate the versatility of this new *** results demonstrate that the pore size characterization using PoreScript is comparable to manual pore size *** PoreScript algorithm was further evaluated to determine the effect of user-input and image parameters(relative image magnification,pixel intensity threshold,and pore structure).Overall,this work validates the accuracy of the PoreScript algorithm across several fabrication methods and provides user-guidance for semi-automated image analysis and increased throughput of scaffold characterization.

Effects of Supplemental Glutamine and Lysine on Growth Performance of Broiler Chickens

The optimum levels of Lysine and Glutamine needed for growth performance and maintenance of the chicken broilers were evaluated in a randomized 3 × 4 factorial arrangement of dietary treatments. The battery cages measured 99 × 66 × 25 cm that can be sufficient for 5 birds. Day old Chicken broilers totaling 180 were assigned to dietary treatments comprising of 3 concentrations of Lysine (0.85, 1.14, and 1.42) each in combination with 4 concentrations of Glutamine (0, 1, 2, and 3). Each dietary treatment was replicated 3 times and each replication had 5 birds. The birds were given feed and water ad libitum with a 23-hour light regimen for a period of 4 weeks. Then, the experimental birds were evaluated for body weight gain, feed consumption, and feed conversion in order to determine their optimum requirement for dietary Lysine and Glutamine. Based on the findings of this study, the highest performance was observed in birds fed the diet supplemented with 1.42 lysine and 1% glutamine, but the highest improvement in feed conversion was observed in diet contain 1.14 and 1.42 with 1% and 3% glutamine, respectively. Birds fed 1.42 lysine and 1% glutamine had the highest total body weight gain and feed consumption. The lysine requirements in the diet for Chicken are between 1.14 and 1.42 with glutamine level of 1%.

The Effect of Irradiance Related Temperature on microalgae Growth in a Tubular Photo Bioreactor for Cleaner Energy

In microalgae based biofuel technology, the light is one of the important factors for the proper growth of microalgae cells as microalgae is a photosynthetic microorganism. For a large scale outdoor culture the irradiance of sunlight and associated temperature is also need to consider. In this study aims to present computational model of microalgae growth taking effect of solar irradiance and corresponding temperature in a tubular photo bioreactor for an outdoor culture system. We consider the transient behavior of temperature inside the photo bioreactor for a microalgae culture. The optimum range of temperature for outdoor cultivation of microalgae is about 22℃ - 27℃ and out of this range the microalgae cell growth inhibits. many correlations have already been established to investigate the algal productivity based on the dynamic conditions of temperature in case of full scale outdoor cultivation. However, none of them are validated yet numerically considering the model as a function of weather conditions, operational behavior and design criteria. A tubular photobioreactor (PBR) with length 20.5 m and radius 0.05 m has taken account as a simulation model. The PBR is horizontally placed as temperature variations can be observed with greater accuracy. As the solar irradiance varies at any geographic latitude for a year and so thus temperature, equations and parameters are established relating the irradiance with the temperature to simulate the effect. We observed some significant effects of temperature on the growth of microalgae. moreover, for the maximum growth of the cells we should control the surrounding temperature.

A Note on Self-Accelerating Olver and Olver-Gauss Beams

The recent introduction by Belafhal et al. [Opt. and Photon. J. 5, 234-246 (2015)] of mth-order Olver beams as a novel class of self-accelerating nondiffracting solutions to the paraxial equation is a direct contradiction to the seminal work of Berry and Balazs who determined that the infinite-energy Airy wave packet is the only accelerating nondiffracting solution to the (1 + 1)D Schrödinger equation. It is shown in this note that the work of Belafhal et al. is valid only for m=0, which coincides with the Airy solution.

One Step Forward, Two Steps Back: Biconvergence of Washed Harmonic Series

We examine variations of the harmonic series by grouping terms into “washings” that alternate sign with the number of terms in a washing growing exponentially with respect to a fixed base. The bases x = 1 and x = ∞ correspond to the alternating harmonic series and the usual harmonic series;we first consider other positive integral bases and further we consider positive real number bases with a unique way to make sense of adding a non-integral number of terms together. In both cases, we prove a remarkable result regarding the difference between the upper and lower convergent values of the series, and give some analysis of this behavior.

Introduction: This study evaluated the difference in operative and clinical outcomes for patients with advanced ovarian cancer after primary debulking surgery (PDS) versus neoadjuvant chemotherapy (NACT) followed by interval debulking surgery (IDS) in Bangladesh. Methods: Sixty patients with advanced epithelial ovarian cancer presenting to the department of Gynaecological Oncology at the National Institute of Cancer Research and Hospital were prospectively enrolled. Thirty patients underwent primary debulking surgery and thirty patients received NACT followed by IDS. Genetic differentiation between and within Northern Native American language groups:an argument for the expansion of the Native American CODIS database
